Sequence homologies between type 1 and type 2A protein phosphatases.

The Mr = 33,000 catalytic fragment of rabbit skeletal muscle type 1 protein phosphatase was digested with trypsin after reduction and alkylation. The resulting peptides were isolated, subjected to automated Edman degradation, and their sequences compared to the deduced peptide sequence of the bovine type 2A protein phosphatase cDNA. Of 10 tryptic peptides from the type 1 phosphatase that were sequenced, nine showed a high degree of homology with the type 2A phosphatase. This provides the first direct sequence comparison suggesting that the type 1 and type 2 protein phosphatases, distinguished functionally by their substrate specificities and sensitivity to inhibitors, make up part of a family of closely related gene products with similar structures.

Phosphorylation of chloroform soluble compounds in plasma membranes of human epidermoid carcinoma A431 cells.

This study investigated a possible role for the epidermal growth factor (EGF) receptor protein tyrosine kinase in phosphoinositide metabolism with plasma membrane vesicles from human epidermoid carcinoma (A431) cells. We found a novel chloroform-soluble product radiolabeled with [gamma-32P]ATP that did not migrate from the origin in the thin layer system designed to separate the phosphoinositides, appeared as a single band of Mr = 3500 on polyacrylamide gels in the presence of dodecyl sulfate, had an ultraviolet absorbance spectrum with a maximum at 275 nm and stained with Coomassie dye. Based on these properties this phosphorylation product is referred to as a proteolipid. The 32P label was not detected in phosphotyrosine [Tyr(P)], phosphoserine [Ser(P)] or phosphothreonine [Thr(P)] and was lost during acid or base hydrolysis. Phosphorylation of proteolipid was increased significantly by EGF, whereas phosphorylation of phosphatidic acid was decreased and labeling of phosphoinositides was unaffected. Thus, it appears that in A431 membranes the EGF receptor/kinase does not utilize phosphatidylinositol as a substrate, but does phosphorylate a membrane proteolipid.

Use of artificial streams for toxicological research.

A review of the literature pertaining to the use of artificial streams as research tools in both toxicant and nontoxicant studies was conducted. This document summarizes that review and provides an assessment of the value of artificial streams in toxicological research. Descriptions are presented of the major types of artificial streams including considerations for the physical design of those streams based on their intended use. Research representative of nontoxicant behavioral, productivity, and trophic relationship studies is summarized to provide a background for the more in-depth discourse on toxicant research. Toxicant research is reviewed in relation to environmental factors such as physical type of artificial stream, light, water, and substrate and experimental conditions including biota tested, method of biota introduction, measured parameters, and toxicant. Comparisons and assessment are presented concerning the use of the appropriate artificial system for addressing various types of toxicant research emphasizing simple vs. complex systems.
