Glycosylated haemoglobin and glycosylated albumin: evaluation of different methods in diabetic control.

The levels of glycosylated haemoglobins and albumin were determined in 56 diabetic patients aged between 5 and 19 years. The glycohaemoglobin levels were measured by two different methods, cation-exchange chromatography and a colorimetric chemical method. The glycosylated albumin levels were measured by a chemical method in plasma albumin purified with Affi-gel Blue. The results were compared with other indicators such as blood glucose and urinary glucose. The postprandial blood glucose levels were well correlated with the levels of glycoproteins: glycohaemoglobin by the column method (r = 0.886), glycohaemoglobin by the chemical method (r = 0.72) and glycoalbumin (r = 0.662). The urinary levels of glucose showed a weak correlation with blood glucose levels (r = 0.214) and with glycoalbumin (r = 0.298), but no correlation with glycohaemoglobin. Significant correlations between the changes of glycosylated albumin levels and glycohaemoglobin were found (r = 0.78). The result of the comparison of the chemical method with the column method for glycohaemoglobin determination showed a high correlation between the values (r = 0.95). There were significant differences in glycosylated protein levels between normal and diabetics. There were also significant differences in glycohaemoglobin and glycoalbumin values between fairly managed and poorly managed groups of diabetics. Of the three parameters, glycoalbumin, glycohaemoglobin and plasma glucose, the greatest absolute changes were found in the levels of glycoalbumin.

Endotoxaemia in cystic fibrosis: response to antibiotics.

In a study of 6 children with cystic fibrosis receiving intravenous antibiotics for pseudomonas lung infection, serum endotoxin values were monitored by a modification of the limulus lysate technique. The values fell with treatment, reflecting a response that was not always apparent on clinical assessment. Endotoxin concentrations may offer a more precise way of monitoring the effects of antibiotic treatment in CF patients.

Ultrastructural studies on spermatogenesis in a sex-ratio-mutant strain of Drosophila simulans.

The aberrant sex-ratio mutation in D. simulans used for this study is a temperature-sensitive autosomal recessive. Homozygous males raised at 16 degrees C produce about 2% of F1, but those raised at 26 degrees C, have a normal sex ratio. Ultrastructural studies of spermatogenesis have revealed many anomalies in the germ cells of flies raised at 16 degrees C, but the same flies raised at 26 degrees C had few anomalies. The earliest spermatogenic stage with noticeable abnormalities was the primary spermatocyte. In later stages there were pronounced abnormalities in nucleoli, chromatin condensation, nuclear shape, Golgi complex, acrosome, and microtubules. There is asynchronous differentiation of spermatids within a bundle. Some of the abnormalities encountered are disorganization or loss of microtubules of the axoneme, degenerating nebenkern derivatives, and increased numbers of lysosomes, multilamellate bodies, and multivesicular bodies. At the lower temperature, more than half of the sperm within the same bundle were found in different stages of degeneration. Genetic analysis suggests that the sex-ratio gene causes abnormalities and degeneration of most of the Y-bearing sperm. However, counts of abnormal sperm at the ultrastructural level indicate that some X-bearing sperm must also undergo degeneration. These observations show that the sex-ratio gene is of variable penetrance at different temperatures in the primary spermatocyte and of differential penetrance in X- and Y-bearing sperm.

Immunoglobulins M and D as antigen-binding receptors on the same cell, with shared specificity.

Antigen-binding cells specific for Type 3 pneumococcal polysaccharide were detected among normal human peripheral blood lymphocytes, tonsillar lymphocytes, and umbilical cord blood lymphocytes. By using mu and delta-specific antiserum it was shown that some specific antigen-binding cells simultaneously expressed both IgM and IgD on their surface. Both classes on individual cells were able to bind the same antigen, suggesting that the variable regions of each immunoglobulin class, when present on the same cell membrane, are coded by the same V gene.

On cell lethals in Drosophila.

Somatic crossing over in flies heterozygous for a dominant Minute mutant may result in two cells, homozygous for wild type and Minute, respectively. If both cells took part in development, a twin spot of two genotypes, different from the rest of the body, would result. Two-hundred wild type spots were found, none of which was accompanied by an adjacent twin spot homozygous for Minute. The absence of these spots shows that their cell lethality is not overcome by transport, including diffusion, across a few cells of gene-dependent material from the neighboring cells which carry one or two wild type alleles of Minute.
