Application of support vector machine (SVM) for prediction toxic activity of different data sets.

As a new method, support vector machine (SVM) were applied for prediction of toxicity of different data sets compared with other two common methods, multiple linear regression (MLR) and RBFNN. Quantitative structure-activity relationships (QSAR) models based on calculated molecular descriptors have been clearly established. Among them, SVM model gave the highest q(2) and correlation coefficient R. It indicates that the SVM performed better generalization ability than the MLR and RBFNN methods, especially in the test set and the whole data set. This eventually leads to better generalization than neural networks, which implement the empirical risk minimization principle and may not converge to global solutions. We would expect SVM method as a powerful tool for the prediction of molecular properties.

QSAR study of natural, synthetic and environmental endocrine disrupting compounds for binding to the androgen receptor.

A large data set of 146 natural, synthetic and environmental chemicals belonging to a broad range of structural classes have been tested for their relative binding affinity (expressed as log (RBA)) to the androgen receptor (AR). These chemicals commonly termed endocrine disrupting compounds (EDCs) present a variety of adverse effects in humans and animals. As assays for binding affinity remains a time-consuming task, it is important to develop predictive methods. In this work, quantitative structure-activity relationships (QSARs) were determined using three methods, multiple linear regression (MLR), radical basis function neural network (RBFNN) and support vector machine (SVM). Five descriptors, accounting for hydrogen-bonding interaction, distribution of atomic charges and molecular branching degree, were selected from a heuristic method to build predictive QSAR models. Comparison of the results obtained from three models showed that the SVM method exhibited the best overall performances, with a RMS error of 0.54 log (RBA) units for the training set, 0.59 for the test set, and 0.55 for the whole set. Moreover, six linear QSAR models were constructed for some specific families based on their chemical structures. These predictive toxicology models, should be useful to rapidly identify potential androgenic endocrine disrupting compounds.

Scavenging effects of natural phenols on oxidizing intermediates of peroxynitrite.

Most of peroxynitrite (ONOO-/ONOOH) is formed via the diffusion-limited reaction between nitric oxide and superoxide. In biological systems, the decomposition of ONOO- yields 30-35% of carbonate radical (CO3*-) and nitrogen dioxide (NO2*), which are strongly oxidizing intermediates and are suggested to take a part of the responsibility for the toxicity of nitric oxide (NO*) or ONOO-. Therefore, the current study focuses on the scavenging activities of phenols toward CO3*- and NO2* to protect biomolecules from damage caused by NO* or ONOO- using the technique of pulse radiolysis. From the build-up kinetic of the phenoxyl radicals and the decay kinetic of CO3*- radical, the rate constants of scavenging reactions were determined to be 1.9-3.4 x 10(8) M(-1) x s(-1) and 0.11-1.9 x 10(8) M(-1) x s(-1) for CO3*- and NO2* respectively. The results indicated that the tested phenols are the efficient scavengers of CO3*- and NO2*.

Glyceroglycolipids from Serratula strangulata.

The rhizomes of Serratula strangulata yielded three glyceroglycolipids, i.e. 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-(6-amine-6-deoxy-alpha-D-glucosyl)-glycerol, 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-(6-p-hydroxy-phenyl-propionamido-6-deoxy-alpha-D-glucosyl)-glycerol and 1,2-di-O-(9Z,12Z,15Z-octadecatrienoyl)-3-O-[alpha-D-glucose(1-6)-beta-D-allose]-glycerol, as well as a known sesquiterpene lactone and three known phytoecdysones. Their structures were elucidated on the basis of spectral data, especially by 2D NMR spectroscopic methods and chemical conversion. These compounds exhibited significant antibacterial and antitumor activities.

[Studies on transgenic tobacco plants expressing two kinds of insect resistant genes].

A synthetic Bt cry1Ac gene fussed with a secretary signal coding sequences at 5' end and a modified gna gene were used to construct a plant expression vector pBSGS1M+ and this vector was transferred into tobacco (Nicotiana tabacum L.) by Agrobacterium-mediated transformation method. Results of PCR, Southern blot and Slot blot analysis indicated that both the chimeric Bt cry1Ac and gna genes were integrated into the genomes of transformed plants. Western blot analysis indicated that at least the cry1Ac protein was produced in transgenic plants. Upon insect bioassay using cotton bollworm (Heliothis armigera Hubner), the mortality of insect larvae on 60% regenerated plants reached 100% in 5 days post infestation and the growth of the survived larvae was seriously inhibited; The results from insect bioassay with peach aphid (Myzus persicae) showed that the transgenic plants were aphid-resistant, evidenced by a 50%-60% reduction in aphid population density, even over 80% for some individual transgenic plants. These results reflect that the modification of the two insect resistant genes and construction of the expression vector are correct and could be valuable for later application in crop breeding for insect resistance.

[Study on effect of jinshui liujun jian oral liquid on serum superoxide dismutase activity and malonyldialdehyde content in mice with chronic bronchitis].

OBJECTIVE: To study the effect of Jinshui Liujun Jian Oral Liquid (JLJOL) on serum superoxide dismutase (SOD) activity and malonyldialdehyde (MDA) content in mice with chronic bronchitis. METHODS: JLJOL was given to the chronic bronchitis mice model (induced by smoking) through gastrogavage, and then SOD activity and MDA content were tested. RESULTS: SOD activity in model mice after JLJOL treatment was 0.67 +/- 0.15 NU/L, which was significantly higher than that in the untreated model (0.39 +/- 0.13 NU/L, P < 0.01). But the MDA content in treated mice was significantly lower than that in untreated one (9.26 +/- 2.90 nmol/L vs 16.07 +/- 5.62 nmol/L, P < 0.01). CONCLUSION: JLJOL could scavenge the injury of free radical on organism.

A clerodane diterpene with antibacterial activity from Ajuga lupulina.

The structure of a new diterpene, C30H46O11, with antibacterial activity against Pseudomonas aeruginose and Escherichia coli, isolated from the fresh whole plants of Ajuga lupulina (Labiatae) was established to be 2 beta-hydroxy-2-methylbutanoyl-3 alpha-lupulin (3-deoxy-14,15-dihydro-2-hydroxy-15-methoxycaryoptinol 2-methylbutanoate), by means of X-ray crystallographic analysis. The present study confirms that the two six-membered rings are in ideal chair conformations.

[Clinical and experimental study on jinshui liujun decoction in treating chronic bronchitis].

OBJECTIVE: To assess the effect of Jinshui Liujun decoction (JSL) in treating chronic bronchitis. METHODS: The effect of JSL on expelling the phlegm and strengthening the body resistance were observed. RESULTS: JSL was proved to be efficacious in improving immune function of the body and pulmonary ventilatory function (P < 0.01). JSL could not only increase the secretion of rats' trachea mucosa and dilute sputum, but also increase the motion speed of the pigeons' trachea cilia, thus facilitate the removal of sputum. Compared with the normal saline control group, both animal experiments displayed significant statistical difference (P < 0.01), but without significant difference when compared the JSL group with the positive drug Juhong Tanke Ye control group. CONCLUSION: JSL has the effect of resolving phlegm as well as improving human immunological and pulmonary ventilatory functions.

Calcium channel blockade and anti-free-radical actions of panaxatriol saponins in cultured myocardiocytes.

AIM: To identify the calcium channel blockade and anti-free-radical actions of panaxatriol saponins Re, Rf, Rg1, Rg2, Rh1, and oleanolic acid saponin Ro. METHODS: On ventricular myocytes of Wistar rats, single channel activities of B, L, and T type calcium channels were recorded with the cell-attached configuration of patch-clamp technic, and free radical contents were measured with electron spin resenance method. RESULTS: Re, Rg1, Rg2, and Rh1 shortened the open times, prolonged the close times, and reduced the open-state probabilities of B, L, and T type calcium channels; Rf shortened the open time, prolonged the close time and reduced the open-state probability of L type calcium channel; Ro did not influence the activity of calcium channels (60 mumol.L-1). Re, Rg1, Rg2, and Rh1 antagonized the increase of free radical content induced by xanthine 0.42 mmol.L-1-xanthine oxidase 5.3 nmol.L-1; Ro and Rf had no effect (30 mumol.L-1). CONCLUSION: Re, Rg1, Rg2, and Rh1 had both the calcium channel blockade and antifree-radical actions. Rf had blockade action on L type calcium channel.

Morphogenesis and ductal development of the prostatic complex of the guinea pig.

The morphogenesis of glandular architecture of the three lobes of prostate gland of the guinea pig, lateral, dorsal, and coagulating gland was studied from 35 days gestation to 90 postnatal days. Epithelial ductal tubules of various lobes of the gland were microdissected after treatment by collagenase and displayed two dimensionally. The number of ductal tips was counted, and the volume of the ductal network was quantified using a graphic tablet. The results show that the growth and ductal morphogenesis fall into two phases: prenatal and postnatal. The first outgrowth of prostatic buds begins at 35 days gestation (gestational length is 65 days). Ductal growth and branching continues over the next 15-20 days and by 55 days gestation, approximately 60%, 79%, and 71% of the adult number of ductal tips of the lateral and dorsal lobes and coagulating gland respectively, are formed. The figures increase to 89%, 84%, and 106%, respectively, by birth. There is little increase in number of ductal tips thereafter. Postnatal growth is accomplished mainly by elongation of existing ductal network with a little additional branching but with an increase in size (volume) of the tubules. Canalization of ductal tubules occurs prenatally in all lobes but postnatal functional cytodifferentiation takes a slightly different pace among them. Ductal morphogenesis of the guinea pig prostate gland differs significantly in time-course from that of the mouse in which ductal development occurs mainly postnatally.

Origin and evolutionary characteristics of antigenic reassortant influenza A (H1N2) viruses isolated from man in China.

During the 1988/1989 influenza season, five antigenic reassortant influenza A (H1N2) viruses not previously isolated from man were isolated in Hebei province, People's Republic of China. All isolates contained haemagglutinins (HAs) and neuraminidases (NAs) which were antigenically similar to those of the recent Russian (H1N1) and Hong Kong influenza A (H3N2) viruses, respectively. The results of antigenic and nucleotide sequence analyses revealed that the genes encoding the polymerase, nucleoprotein, NA, matrix and non-structural proteins of the reassortant A/Hebei/24/89 (H1N2) virus were derived from the H3N2 parent virus, whereas its HA gene was from the H1N1 parent virus. The nucleotide sequences of the HA (encoding the HA1 subunit) and NA genes of the reassortant viruses were also determined. Phylogenetic trees constructed from these data by the neighbour-joining method revealed that the HA gene of the reassortant virus was closely related to those of recent human H1N1 viruses, whereas the NA gene was related to a recent human Hong Kong (H3N2) virus lineage.
