Programmed cell death in intervertebral disc degeneration.

Intervertebral disc (IVD) degeneration is largely a process of destruction and failure of the extracellular matrix (ECM), and symptomatic IVD degeneration is thought to be one of the leading causes of morbidity or life quality deterioration in the elderly. To date, however, the mechanism of IVD degeneration is still not fully understood. Cellular loss from cell death in the process of IVD degeneration has long been confirmed and considered to contribute to ECM degradation, but the causes and the manners of IVD cell death remain unclear. Programmed cell death (PCD) is executed by an active cellular process and is extensively involved in many physiological and pathological processes, including embryonic development and human degenerative diseases. Thus, the relationship between PCD and IVD degeneration has become a new research focus of interest in recent years. By reviewing the available literature concentrated on PCD in IVD and discussing the methodology of detecting PCD in IVD cells, its inducing factors, the relationship of cell death to ECM degradation, and the potential therapy for IVD degeneration by modulation of PCD, we conclude that IVD cells undergo PCD via different signal transduction pathways in response to different stimuli, that PCD may play a role in the process of IVD degeneration, and that modulation of PCD might be a potential therapeutic strategy for IVD degeneration.

[Curative effects of Fuzheng Huayu capsules on hepatic fibrosis and the functional mechanisms: a review].

Deficiency of Qi (healthy energy) and blood stasis are the basic pathological changes of hepatic fibrosis according to the theories of traditional Chinese medicine. Fuzheng Huayu Capsule, a compound Chinese herbal medicine for hepatic fibrosis, is produced in the light of this pathological mechanism. More than a decade of clinical studies and experimental researches show that this medicine has effects of protecting hepatic cells, relieving liver injury, and controlling the development of hepatic fibrosis. It has definite functional mechanisms on anti-hepatic fibrosis. It is a safe and effective medicine for hepatic fibrosis, and deserves to be well introduced to clinic.

[Alteration of cytokines and mucin in lower respiratory tract in allergic rhinitis model in rats].

OBJECTIVE: To investigate the relationship between allergic rhinitis (AR) and asthma as well as the mechanisms related with it. METHODS: Sixty healthy rats were randomly divided into AR group and control group. AR model was established by intraperitoneal injection of ovalbumin (OVA) and nasal challenge with OVA. Nasal mucosa and lung tissue from both groups were stained with hematoxylin-eosin (HE), alcian-blue and periodic acid-schiff (AB-PAS), respectively. At the same time, the lung tissue was studied by transmission electron microscopy (TEM). Enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry were used to examine the level of interleukin-4 (IL-4) in bronchial alveolus lavage fluid (BALF) and the expression of intercellular adhesion molecule-1 (ICAM-1) and MUC5AC in nasal and lung tissue, respectively. RESULTS: Infiltration of inflammatory cells in nasal mucosa and lung tissue of AR model in rat was evident. Cilia destruction of bronchial epithelial cells of AR model was found. The level of IL4 in BALF of AR group (58.10 +/- 7.92) pg/ml was significant higher compared with that in control group (24.66 +/- 2.07) pg/ml. The expression of ICAM-1 (0.66 +/- 0.24) and MUC5AC (0.71 +/- 0.10) in lung tissue were both significantly higher than that in control group (0.23 +/- 0.02, 0.29 +/- 0.03). CONCLUSIONS: Allergic inflammation in nasal mucosa not only leads to changes in both histopathology and immunology, but also initiates the inflammation in lower respiratory tract mainly causing the change of cytokines and mucin.

[Alteration of survivin gene expression in ovarian cancer cell line CAOV3 after chemotherapy in vitro].

OBJECTIVE: To study the alteration of survivin gene expression in ovarian cancer cell CAOV3 after chemotherapy in vitro. METHODS: After the secondary culture of ovarian cancer cell line CAOV3, inhibition of ovarian cancer cell growth induced by paclitaxel or carboplatin at different concentration was detected by methyl thiazolyl tetrazolium (MTT). Alteration of survivin mRNA expression was evaluated by reverse transcription polymerase chain reaction (RT-PCR) in ovarian cancer cells which had been cultured with paclitaxel or carboplatin of high concentration (200, 500 mg/L) for 1 d or of low concentration (20, 50 mg/L) for 1, 3 and 5 d, respectively. Meanwhile, CAOV3 cell apoptosis was detected by flow cytometry. RESULTS: The inhibition rates on growth of ovarian cancer cell CAOV3 by different concentrations of paclitaxel or carboplatin after 48 h were 94%, 88%, 34%, 22%, 13% and 97%, 46%, 14%, 9%, 7%. The apoptosis rates of CAOV3 in low concentration paclitaxel for 1, 3, and 5 d were 15%, 23% and 29%. And the apoptosis rate in high concentration paclitaxel for 1 d was 43%. The apoptosis rate of CAOV3 in low concentration carboplatin for 1, 3, and 5 d were 14%, 22% and 26%. And in high concentration carboplatin for 1 d the rate was 19%. Expression of survivin mRNA increased distinctly after culturing in paclitaxel or carboplatin of low concentration for 3 d and of high concentration for 1 d compared to the control group (P < 0.01), the increase was especially evident in paclitaxel treated group (P < 0.01). CONCLUSION: Survivin gene may play an inhibitory role in the chemotherapy of ovarian cancer cell CAOV3 in vitro.
