[Effects of Shengmai Chenggu capsule on proliferation and vascular endothelial growth factor secretion of vascular endothelial cells].

OBJECTIVE: To study the effect of the serum of rabbits fed with Shengmai Chenggu capsule (SMCGC), a preparation of traditional Chinese medicine, on the proliferation and vascular endothelial growth factor (VEGF) secretion of cultured ECV304 cells. METHODS: The drug-containing serum from rabbits with intragastric administration of SMCGC at different doses was obtained for treatment of ECV304 cells cultured in vitro, with the cells treated with blank serum and ligustrazine serving as the control. The proliferation of ECV304 cells was detected by MTT assay and VEGF concentration in the culture medium of the cells measured by means of ELISA. The ultrastructure of ECV304 cells treated with the sera for 12 h was observed by transmission electron microscopy. RESULTS: After treatment with SMCGC-containing rabbit serum for 12 and 36 h, respectively, the proliferation of ECV304 cells in all dosage groups showed no significant increases (P>0.05), and the sera of rabbits fed with medium and high doses of SMCGC significantly inhibited the proliferation of ECV304 cells after a 36-hour treatment (P<0.01). The high-dose rabbit serum might even cause apoptosis of ECV304 cells, and low-dose serum significantly stimulated VEGF secretion of the ECV304 cells (P<0.01). CONCLUSIONS: SMCGC does not promote the proliferation of ECV304 cells, and the serum of low-dose SMCGC-treated rabbits may stimulate VEGF secretion of the ECV304 cells, which might be the main mechanism of SMCGC in stimulating angiogenesis.

[Syntheses and spectral studies of functionalized ZnS nanoparticles as fluorescence probes].

In this work, nano-ZnS has been successfully prepared. The nano-ZnS has also been modified with sodium mercaptoacetic acid. The functionalized nanoparticles are water-soluble and biocompatible. All the nanoparticles have been characterized by IR spectra, UV spectra, fluorescence spectra and TEM images. In comparison with single organic fluorophore, the nanoparticles probes are brighter and more photostable, and do not suffer from blinking. The nanoparticles have a narrow, tunable, symmetric emission spectrum and a broad, continuous excitation spectrum. They are also photochemically stable. Effects of proteins and nucleic acids on the UV spectra and fluorescence spectra of the functionalized ZnS have also been studied. The intensities of UV spectra and fluorescence spectra of the functionalized ZnS are enhanced by proteins, and, however, are quenched by nucleic acids. The functionalized colloidal solutions prepared are hopeful of use as fluorescence probes in biological staining and diagnostics.

[Expression of survivin gene and its relation with the expression of bcl-2 and bax protein in epithelial ovarian cancer].

OBJECTIVE: To study the expression of apoptosis related gene survivin and its relation with expression of bcl-2, bax protein in epithelial ovarian cancer. METHODS: Expression of survivin gene was evaluated by reverse transcription polymerase chain reaction (RT-PCR) in 35 cases of epithelial ovarian cancer, and compared with that in 10 cases of borderline cancer, 10 cases of benign tumors and 10 cases of normal tissue. Expression of bcl-2 and bax was detected by immunohistochemistry streptomycin-avidin-biotin-peroxidase complex (SABC) method, and correlation between them was analyzed. RESULTS: Expression of survivin gene was detected in a significantly greater proportion in epithelial ovarian cancer and borderline tumors than benign tumors and normal tissue. There was no relationship between survivin gene expression and FIGO stage, histologic grade, pathological type and lymphatic metastasis. Expression of bcl-2 and bax protein was positively and negatively correlated with expression of survivin gene respectively. CONCLUSION: survivin gene may play an important role in pathogenesis of ovarian cancer, apoptosis related gene bcl-2 may have a synergic role and bax have an antagonistic role with survivin gene in formation and progression of ovarian cancer.
