Circular Dammann grating.

A circular Dammann grating that can produce circular equal intensities at various orders in the far field is described. A set of parameters such as order, circular number, uniformity, and diffraction efficiency has been defined to describe the novel diffractive phase elements. Numerical solutions of binary-phase (0, pi) circular Dammann gratings are given. The results of experiments with a four-order circular Dammann grating made by a lithographic technique are presented. This novel diffractive optical element should be highly interesting in a wide variety of practical applications.

Multifunctional double-layered diffractive optical element.

We propose a novel multifunctional double-layered diffractive optical element (DOE) based on the fractional Talbot effect. This DOE consists of two layers: one is the encoding layer, in which multiple sub-DOEs, i.e., multiple optical functions, are encoded; the other is the decoding layer, which is a properly designed Talbot illuminator. This DOE can perform each of the multiple optical functions one by one by shifting the encoding layer. Experimental results demonstrate that this method is efficient. This device should be highly interesting for integrated optics, optical interconnection, secure optical storage, and dynamic optical fiber communications.

Miniaturized three-dimensional endoscopic imaging system based on active stereovision.

A miniaturized three-dimensional endoscopic imaging system is presented. The system consists of two imaging in channels that can be used to obtain an image from an object of interest and to project as tructured light onto the imaged object to measure the surface topology. The structured light was generated with a collimated monochromatic light source and a holographic binary phase grating. The imaging and projection channels were calibrated by use of a modified pinhole camera. The surface profile was extracted by use of triangulation between the projected feature points and the two channel ofthe endoscope. The imaging system was evaluated in three-dimensional measurements of several objects with known geometries. The results show that surface profiles of the objects with different surfaces and dimensions can be obtained at high accuracy. The in vivo measurements at tissue sites of human skin and an oral cavity demonstrated the potential of the technique for clinical applications.

CHB2, a member of the SWI3 gene family, is a global regulator in Arabidopsis.

The SWI/SNF complex is an ATP-dependent chromatin remodeling complex that plays an important role in the regulation of eukaryotic gene expression. Very little is known about the function of SWI/SNF complex in plants compared with animals and yeast. SWI3 is one of the core components of the SWI/SNF chromatin remodeling complexes in yeast. We have identified a putative SWI3-like cDNA clone, CHB2 (AtSWI3B), from Arabidopsis thaliana by screening the expressed sequence tag database. CHB2 encodes a putative protein of 469 amino acids and shares 23% amino acid sequence identity and 64% similarity with the yeast SWI3. The Arabidopsis genome contains four SWI3-like genes, namely CHB1 (AtSWI3A), CHB2 (AtSWI3B), CHB3 (AtSWI3C) and CHB4 (AtSWI3D). The expression of CHB2, CHB3 and CHB4 mRNA was detected in all tissues analyzed by RT-PCR. The expression of CHB1 mRNA, however, could not be detected in the siliques, suggesting that there is differential expression among CHB genes in different Arabidopsis tissues. To investigate the role of CHB2 in plants, Arabidopsis plants were transformed with a gene construct comprising a CHB2 cDNA in the antisense orientation driven by the CaMV 35S promoter. Repression of CHB2 expression resulted in pleiotropic developmental abnormalities including abnormal seedling and leaf phenotypes, dwarfism, delayed flowering and no apical dominance, suggesting a global role for CHB2 in the regulation of gene expression. Our results indicate that CHB2 plays an essential role in plant growth and development.

Repression of gene expression by Arabidopsis HD2 histone deacetylases.

The four HD2 proteins of Arabidopsis thaliana (AtHD2A-D) belong to a unique class of histone deacetylases that is plant specific. Previously, we have demonstrated that one of the members, AtHD2A, can mediate transcriptional repression when targeted to the promoter of a reporter gene. Here, we report that AtHD2B and AtHD2C can also repress gene expression. AtHD2A and AtHD2C differ from AtHD2B and AtHD2D in the composition of their structural domains. Our data show that both structural types play a role in the repression of gene transcription. We demonstrate that AtHD2A can mediate gene repression through interactions with transcription factors in plants. By fusing AtHD2A with the DNA-binding domain of the plant transcriptional factor Pti4, the expression of a GCC box containing reporter gene was repressed. We also demonstrated repression of a GUS gene with GAL4 enhancers using transgenic plants that expressed a GAL4/AtHD2A fusion gene. Furthermore, the expression of the GAL4/AtHD2A protein using the seed-specific napin promoter (NAP2) and the constitutive tCUP promoter demonstrated that repression of transgenes could be achieved in a tissue-specific or unrestricted manner. Targeting of HD2 proteins to specific promoters using transcription factor DNA-binding domains may therefore provide a new technology for silencing target genes and pathways in plants as well as for assessing the function of unknown transcription factors.

Nonvolatile holograms in LiNbO3:Fe:Cu by use of the bleaching effect.

We report our observation of a bleaching effect under an ultraviolet exposure in LiNbO3:Fe:Cu crystals. Two three-step recording-transferring-fixing schemes are proposed to record nonvolatile photorefractive holograms in such crystals. In the schemes two red laser beams and an ultraviolet illumination are used selectively to write the charge grating inthe shallow-level Fe centers, to develop the charge grating inthe deep-level Cu centers by transferring the charge grating in the Fe centers, and to fix only thecharge grating in the Cu centers for unerasable read-out. Experimental results, verifications, and an optimal recording scheme are given. A comparison of the lithium niobate crystals of the same double-doping system of Fe:Mn, Ce:Mn, Ce:Cu, and Fe:Cu is outlined.
