Experimental studies on PNP suicide gene therapy of hepatoma.

To investigate the killing effect of PNP/MeP-dR suicide gene system on hepatoma cells, pcDNA3.0/PNP, an eukaryotic expression vector harboring E. coli PNP gene, was transfected into human hepatoma HepG2 cells by liposome-mediated method. A HepG2 cell line with stable PNP gene expression, HepG2/PNP, was established with presence of G418 selection. The cell growth curves were determined with trypan blue staining. The sensitivity of HepG2/PNP to MeP-dR and bystander effects were assayed by MTT and FCM methods. The enzymatic activity of the product of PNP gene was determined by HPLC method. The cytotoxic effects of MeP-dR on HepG2/PNP cells were obvious (IC50 = 4.5 micromol/L) and all HepG2/PNP cells were killed 4 days after the treatment with 100 micromol/L MeP-dR. In mixed cultures containing increasing percentages of HepG2/PNP cells, total population killing was demonstrated when HepG2/PNP cells accounted for as few as 5% of all HepG2 cells 8 days after the treatment with 100 micromol MeP-dR. High-pressure liquid chromatography (HPLC) demonstrated that the PNP enzyme could convert MeP-dR into 6-MP. PNP/MeP-dR suicide gene system had an advantage over traditional suicide gene systems for hepatoma gene therapy. Our e results suggest that high-level bystander effects of this system result in significant anti-tumor responses to hepatoma gene therapy, especially in vivo.

Determination of the small band gap of carbon nanotubes using the ambipolar random telegraph signal.

The ambipolar random telegraph signal (RTS) (i.e., RTS in both hole conduction at negative gate biases and electron conduction at positive gate biases) is observed in an ambipolar carbon nanotube field-effect transistor (CNT-FET). Then, the ambipolar RTS is used to extract the small band gap of the SWNT. The determination of the small band gap CNT using RTS demonstrates a potentially high accuracy and stability. Other methods are provided to confirm the small band gap of the SWNT.

Single-branch endograft for treating stanford type B aortic dissections with entry tears in proximity to the left subclavian artery.

PURPOSE: To present a preliminary experience with a single-branch endograft for excluding Stanford type B aortic dissections with entry tears adjacent to the left subclavian artery (LSA). METHODS: From February to August 2004, 16 symptomatic patients (15 men; mean age 57.8 years, range 41-73) having a Stanford B aortic dissection with a proximal tear <15 mm from the LSA orifice were treated with a single-branch thoracic endograft constructed of Z-shaped nitinol stents to which a Dacron graft had been sutured. The tubular main stent-graft had a branched segment connected to it for implantation in the LSA. The deployment method is the same as for a straight endograft except that the branch is deployed first in the LSA to stabilize the entire device. The main body of endograft is then deployed into the descending aorta to seal the tear, block flow from entering the false lumen, and enlarge the true lumen. RESULTS: Sixteen branched endografts were deployed to seal the entry tears, with a technical success rate of 94% (15/16). One case was converted to surgery when the branch became trapped in the LSA at the left thoracic outlet. Two proximal endoleaks were treated with additional tubular endografts. All the proximal tears were sealed by the stent-grafts, and the compressed true lumens were widened. No paraplegia or distal organ or limb ischemia was noted, nor was there any mortality or complications. By 3 months post treatment, symptoms had abated, thrombosis had formed in the false lumen, and the true lumen had resumed its normal diameter in 15 of the 16 stent-graft patients. One patient has a distal re-entry at the level of the visceral arteries that is being observed. CONCLUSIONS: The single-branch thoracic aortic endograft provides a simpler, safer, and more effective means of treating aortic dissections with entry tears in proximity to the LSA.

Hands-free human computer interaction via an electromyogram-based classification algorithm.

A four-electrode system for hands-free computer cursor control, based on the digital processing of Electromyogram (EMG) signals is proposed. The electrodes are located over the right frontalis, the procerus, the left temporalis and the right temporalis muscles in the head. This system is meant to enable individuals paralyzed from the neck down (e.g., due to Spinal Cord Injury) to interact with computers using point-and-click graphic interfaces. The intention is to translate electromyograms derived from muscle contractions associated with specific facial movements into five cursor actions, namely: Left, Right, Up, Down and Left-click. This translation is accomplished by a digital signal processing classification algorithm that takes advantage of the divergent spectral nature of the EMG signals produced by the frontalis, temporalis, and procerus muscles, respectively. The effectiveness of the algorithm is evaluated by comparing its performance to that of a previously developed three-electrode EMG-based algorithm, using Matlab simulations. The results indicate that the algorithm classifies with great accuracy and provides a marked improvement over the previous three-electrode system.

User stress detection in human-computer interactions.

The emerging research area of Affective Computing seeks to advance the field of Human-Computer Interaction (HCI) by enabling computers to interact with users in ways appropriate to their affective states. Affect recognition, including the use of psychophysiologcal measures (e.g. heart rate), facial expressions, speech recognition etc. to derive an assessment of user affective state based on factors from the current task context, is an important foundation required for the development of Affective Computing. Our research focuses on the use of three physiological signals: Blood Volume Pulse (BVP), Galvanic Skin Response (GSR) and Pupil Diameter (PD), to automatically monitor the level of stress in computer users. This paper reports on the hardware and software instrumentation development and signal processing approach used to detect the stress level of a subject interacting with a computer, within the framework of a specific experimental task, which is called the 'Stroop Test'. For this experiment, a computer game was implemented and adapted to make the subject experience the Stroop Effect, evoked by the mismatch between the font color and the meaning of a certain word (name of a color) displayed, while his/her BVP, GSR and PD signals were continuously recorded. Several data processing techniques were applied to extract effective attributes of the stress level of the subjects throughout the experiment. Current results indicate that there exists interesting similarity among changes in those three signals and the shift in the emotional states when stress stimuli are applied to the interaction environment.

Changes in epidemiology and clinical characteristics of cervical cancer over the past 50 years.

OBJECTIVE: To evaluate the changes in the epidemiology and clinical characteristics of cervical cancer over the past 50 years, and explore appropriate treatment corresponding to these changes. METHODS: The clinical and pathological data of 1557 patients with invasive cervical cancer treated between January, 1955 and December, 2004 were retrospectively analyzed. RESULTS AND CONCLUSIONS: The average age of cervical cancer onset gradually decreased over the past 50 years, from 56.27+/-8.45 in 1955-1964 to 43.81+/-8.9 years in 1995-2004, whereas the ratio of young (< or =35 years old) patients rose from 3.42% to 24.91%. The ratio of early clinical stage (stages I-II) and non-squamous cancer also steadily increased (P<0.05, respectively). The tumor stage, pathological type and rate of lymph node metastasis were all significantly different among different age groups (P<0.05). In particular, the young (< or =35 years old) group had evidently higher ratios of non-squamous and advanced stage (III-IV) cancers with a higher rate of lymphatic metastasis in comparison with other age groups (P<0.01, respectively). Because of the changes in epidemiology and clinical characteristics of cervical cancer, it is necessary to modify the conventional treatment regimens and explore reasonable therapy corresponding to these changes. Preservation of reproductive endocrine function ought to be fully considered in cervical cancer treatment in women at childbearing age. Neoadjuvant intraarterial chemotherapy is an useful method for cervical cancer treatment at present.

[Generation of chemical-inducible activation tagging T-DNA insertion lines of Arabidopsis thaliana].

Using an estrogen-inducible expression XVE (LexA-VP16-Estragon Receptor) system, we have generated approximately 40 000 independent T-DNA insertion lines of Arabidopsis thaliana. Segregation analyses of about 18000 lines indicated that 51.6% of them contain single T-DNA insertions and that the average insertion number is 1.38 copies per line. Mutants displaying a variety of morphological alterations were identified, including those that affect development of roots, hypocotyls, leaves, floral organs and seeds as well as the flowering time.

[Expression and significance of angiopoietin-1 and angiopoietin-2 in oral squamous cell cacinoma].

BACKGROUND & OBJECTIVE: Recently, Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) have been identified as potential angiogenic factors. The local balance between them and other angiogenic factors determines the growth, stableness, or regression of blood vessels. This study was to investigate the correlations of Ang-1 and Ang-2 expression to clinicopathologic features, microvessel density (MVD), and vessel maturation index (VMI) of oral squamous cell cacinoma (OSCC), and evaluate the significance of Ang-1 and Ang-2 coexpression in tumor angiogenesis and vessel maturation. METHODS: The expression of Ang-1 and Ang-2 in 41 specimens of OSCC, 30 specimens of adjacent noncancerous oral tissues, and 10 specimens of normal oral mucosa was detected by immunohistochemistry. MVD and VMI were assessed with double-labeling immunohistochemical staining against CD34 and alpha-smooth muscle actin (alpha-SMA). RESULTS: The positive rate of Ang-1 was significantly lower in OSCC than in adjacent noncancerous oral tssues and normal oral mucosa (41.46% vs. 90.00% and 90.0%, P < 0.05); the positive rate of Ang-2 was significantly higher in OSCC than in adjacent noncancerous oral tssues and normal oral mucosa (51.22% vs. 26.67% and 0, P < 0.05). The differences in Ang-1 and Ang-2 expression between adjacent noncancerous oral tssues and normal oral mucosa were not significant (P > 0.05). The positive rate of Ang-1 was significantly higher in highly differentiated tumor than in moderately differentiated tumor (56.00% vs. 18.75%, P < 0.05); the positive rate of Ang-2 was significantly higher in tumor with lymph node metastasis than in tumor without lymph node metastasis (84.62% vs. 35.71%, P < 0.01). Ang-1 and Ang-2 expression were closely correlated with angiogenesis and vessel maturation of tumor (P < 0.05). Ang-1 and Ang-2 had antagonistic effect on the regulation of tumor vessel maturation. CONCLUSION: The down-regulation of Ang-1 and up-regulation of Ang-2 are closely related to tumor angiogenesis and vessel maturation.

No association between the genetic polymorphisms within RTN4 and schizophrenia in the Chinese population.

The RTN4 gene on chromosome 2p13-14 has been reported to be over-expressed in schizophrenia by a cDNA subtractive hybridization experiment between postmortem human frontal cerebral cortices from Canadian schizophrenia individuals and neurological controls. The same study also reported a high prevalence of homozygous CAA insertion in the schizophrenia. In a replication attempt to investigate the role of RTN4 in the etiology of schizophrenia, we genotyped the CAA insertion polymorphism and other three genetic polymorphisms (a TATC deletion in the 3'-untranslated region and two single nucleotide polymorphisms in the 5' region) within RTN4 and conducted a case-control study in the Chinese population. There were no significant discrepancies in allele and genotype frequencies of the four polymorphisms individually and haplotype distribution between the cases and the controls. Our current data suggest that the genetic polymorphisms within RTN4 are unlikely to confer an increased susceptibility to schizophrenia in the Chinese population.

charge storage behavior of nanowire transistors functionalized with bis(terpyridine)-Fe(II) molecules: dependence on molecular structure.

We studied the influence of three bis(terpyridine)-Fe(II) molecules-(X-tpy)2FeCl2 (X = H (1), SAc (2), and 4-phenyl-SAc (3)-on charge storage of a nanowire transistor. The molecules were assembled on the surface of an indium oxide nanowire that forms the conduction channel of the transistor. We found that the charge storage characteristics of such a device strongly depends on the structure of the terpyridine ligand: both retention time (tau) and threshold voltage shift (DeltaVth) increased in the order of 1 < 2 < 3, with tau of 200 s, 12 h, and 287 h and DeltaVth at 4.8, 12, and 28 V, respectively. Furthermore, when we placed the devices with molecules 1 and 3 in a vacuum and recorded the I-Vg curves in a two-day period, we observed higher hysteresis stability for device with molecule 3. For example, DeltaVth was reduced from 4.8 to 1.7 V for the device with molecule 1, while there was no reduction in DeltaVth for the device with molecule 2. These results suggest that thiolate headgroup and/or longer ligand length raises the charge tunneling barrier and results in longer charge retention and wider, more stable memory window. This work demonstrates the potential of chemical synthesis toward tailored device characteristics.

Waveguide-coupled octagonal microdisk channel add-drop filters.

We report planar waveguide-coupled octagonal microdisk channel add-drop filters on silicon nitride-on-silica substrates. Using ray optics, we show that discrete octagonal cavities have wave-front-matched four- and eight-bounce ray orbits. From a planar waveguide-coupled 50-microm-sized octagonal microdisk filter, our experiment demonstrates TM-polarized eight-bounce modes with a coupling efficiency of approximately 56%, Q approximately equal to 1150, and a finesse of approximately 5. Using the two-dimensional finite-difference time-domain method, we obtain qualitative agreement between measurement and simulation of a smaller-sized waveguide-coupled octagonal microdisk filter.

Self-assembled spiral nanoarchitecture and supramolecular chirality in Langmuir-Blodgett films of an achiral amphiphilic barbituric acid.

An amphiphilic barbituric acid derivative was found to form stable monolayers showing a clear phase transition at the air/water interface. It is interesting to find that the deposited Langmuir-Blodgett (LB) films of the compound showed circular dichroism (CD) although the molecule itself was achiral. AFM measurements on the transferred one-layer LB film revealed that spiral nanoarchitectures were formed. It was further found that the supramolecular chirality of the LB films was related to symmetry breaking at the interface. Hydrogen bonding and the pi-pi stacking between the neighboring molecules resulted in chiral fibers which formed the spiral structures. To the best of our knowledge, this is the first report on the chirality of the molecular assemblies and spiral nanostructures formed through the air/water interface by achiral molecules.

Discovery of a potent, selective and orally active canine COX-2 inhibitor, 2-(3-difluoromethyl-5-phenyl-pyrazol-1-yl)-5-methanesulfonyl-pyridine.

Structure-activity relationship (SAR) studies of 2-[3-di(and tri)fluoromethyl-5-arylpyrazol-1-yl]-5-methanesulfonylpyridine derivatives for canine COX enzymes are described. This led to the identification of 12a as a lead candidate for further progression. The in vitro and in vivo activity of 12a for the canine COX-2 enzyme as well as its in vivo efficacy and pharmacokinetic properties in dog are highlighted.

[Expression of TMP Fc in Pichia pastoris and identification of its biological activity].

The DNA coding for the fusion protein of thromobopoietin mimetic peptide (TMP) and human IgG1 Fc fragment was amplified from recombinant plasmid pET28a/TMPFc, inserted into pPICZalphaA and transformed into Pichia pastoris using electroporation. The recombinants of correct phenotype were identified after screening on MDH and MMH culture medium. The fusion gene was verified with PCR and western blot. MTT method was used to test the activity of TMPFc in promoting the growth of Ba/ F3-mpl cell. The TMPFc with a 64 000 molecular weight was a secretary protein in the system and its expression amounted to 65% of the total protein in the medium supernatant. The TMPFc showed a promotive effect on the growth of Ba/F3-mpl in vitro. A significant portion of the secretary protein existed as dimer, which provided material for studying the dimer in future.

An automated pontine-wave detection system.

STUDY OBJECTIVES: Pontine-waves (P-waves), the pontine component of ponto-geniculo-occipital waves, represent a close marker of brainstem phasic events and are associated with cardiorespiratory changes in sleep. Because visual scoring is subjective and cumbersome, we developed an automated P-wave analysis system, which could detect and classify P-waves as clusters or as isolated events in bipolar recordings of the pontine electroencephalogram (EEG) of conscious rats. DESIGN: A computer algorithm was developed to extract and normalize each half-wave of the pontine EEG according to the background noise level. Candidate events for different P-wave patterns (uniphasic, biphasic, and triphasic) were compared to a corresponding set of amplitude and duration thresholds to identify P-waves and to reject artifacts. PARTICIPANTS: Ten adult male Sprague-Dawley rats were instrumented for chronic polysomnography. MEASUREMENTS AND RESULTS: Two human experts manually scored each recording, and their consensus score was used as the "gold standard" for algorithm optimization and validation. The algorithm's scoring thresholds were optimized on a training set of 5 six-hour polysomnographic records, yielding 96.8% accuracy and 97.7% sensitivity versus human consensus scoring. Validation of the algorithm, using the optimized threshold values, was conducted using a set of 5 independent recordings, resulting in 94.8% accuracy and 94.7% sensitivity versus human consensus scoring. CONCLUSIONS: We have developed and validated an automated system for detection and classification of P-waves in conscious rats with advantages over human scoring, including increased speed and perfect reliability.

Digital signal processing methods for the evaluation of Blood Volume Pulse (BVP) waveform changes due to exercise.

Previous research by our group has revealed that the Blood Volume Pulse (BVP) waveform recorded using an infrared finger photoplethysmograph (PPG) undergoes changes as the subject performs physical exercise. In particular, a reduction in the depth of the Dicrotic Notch has been observed. There is an interest in characterizing those changes through a single parameter to measure the level of exercise the subject has reached, at any time during an exercise session. This paper reports on the comparison of three Digital Signal Processing approaches designed to reflect the BVP waveform changes through a single parameter, which could be obtained automatically from the digitized BVP signal. The first approach derives a single parameter from the distribution found in the average histogram of several time-aligned and averaged BVP beats. Our second approach analyzes the ratio observed between the first harmonic and higher harmonics in the BVP signal. The third approach evaluates the Dicrotic Notch depth directly from the BVP waveform, tracking sample values about the local minimum defined by the Dicrotic Notch. Our study, involving observations from 10 subjects, ranks these three approaches according to their ability to reflect the changes in BVP due to exercise.

[Synthesis and function analysis of a new thrombopoietin (TPO) mimic peptide].

In order to find a new TPO-mimic peptide with similar activity to TPO while reducing the side effects, a TPO-mimic peptide (P1) screened from a random peptide library was restructured. The new structure of the TPO mimic peptide (P2) was synthesized. After coupling P2 with Dextran 10 and performing intermolecular oxidation, dextran-coupled and dimerized form of P2 were obtained, naming D-P2 and (P2)(2) respectively. The activities of the peptides in vitro were measured with MTT method. The results showed that the EC50 of P2 was 20 nmol/L, 700 times higher than P1. The EC50 of D-P2 and (P2)(2) were 0.35 nmol/L and 0.14 nmol/L, respectively. After administrating to the mouse, the peptides increased the number of platelets in the blood circulation obviously without influence on other blood cells. In conclusion, the TPO-mimic peptides have prospects in treating diseases related with thrombocytopenia.
