Pseudolarix acid B, a new tubulin-binding agent, inhibits angiogenesis by interacting with a novel binding site on tubulin.

Tubulin-binding agents have received considerable interest as potential tumor-selective angiogenesis-targeting drugs. Herein, we report that pseudolarix acid B (PAB), isolated from the traditional Chinese medicinal plant Pseudolarix kaempferi Gordon, is a tubulin-binding agent. We further demonstrate that PAB significantly and dose-dependently inhibits proliferation, migration, and tube formation by human microvessel enthothelial cells. It is noteworthy that PAB eliminated newly formed endothelial tubes and microvessels both in vitro and in vivo. In addition, PAB dramatically arrested the cell cycle at G2/M phase. PAB also induced endothelial cell retraction, intercellular gap formation, and promoted actin stress fiber formation in conjunction with disruption of the tubulin and actin cytoskeletons. All of these effects occurred at noncytotoxic concentrations of PAB. We found that these effects of PAB are attributable to depolymerization of tubulin by direct interaction with a distinct binding site on tubulin compared with those of colchicine and vinblastine. Taken together, these findings show that PAB is a candidate antiangiogenic agent for use in cancer therapy, and they provide proof of principle for targeting this novel binding site on tubulin as a new strategy for treating cancer.

Synthesis and evolution of PbS nanocrystals through a surfactant-assisted solvothermal route.

We present a surfactant-assisted solvothermal approach for the controllable synthesis of a PbS nanocrystal at low temperature (85 degrees C). Nanotubes (400 nm in length with an outer diameter of 30 nm), bundle-like long nanorods (about 5-15 mum long and an average diameter of 100 nm), nanowires (5-20 mum in length and with a diameter of 20-50 nm), short nanorods (100-300 nm in length and an axial ratio of 5-10), nanoparticles (25 nm in width with an aspect ratio of 2), and nanocubes (a short axis length of 10 nm and a long axis length of 15 nm) were successfully prepared and characterized by transmission electron microscopy, scanning electron microscopy, and powder X-ray diffraction pattern. A series of experimental results indicated that several experimental factors, such as AOT concentration, ratio of [water]/[surfactant], reaction time, and ratio of the reagents, play key roles in the final morphologies of PbS. Possible formation mechanisms of PbS nanorods and nanotubes were proposed.

Direct chitosan-mediated gene delivery to the rabbit knee joints in vitro and in vivo.

Chitosan vector system is expected to be useful for direct gene therapy for joint disease. This study first sought to confirm that foreign genes can be transferred to articular chondrocytes in primary culture. Next, chitosan-DNA nanoparticles containing IL-1Ra or IL-10 gene were injected directly into the knee joint cavities of osteoarthritis rabbits to clarify the in vivo transfer availability of the chitosan vectors. Clear expression of IL-1Ra was detected in the knee joint synovial fluid of the chitosan IL-1Ra-injected group. While no expression was detected in the chitosan IL-10-injected group, this demonstrates that the transfection efficiency of chitosan-DNA nanoparticles was closely related to the type of the gene product. A significant reduction was also noted in the severity of histologic cartilage lesions in the group that received the chitosan IL-1Ra injection. This avenue may therefore represent a promising future treatment for osteoarthritis.

Dynamic assessment of the focal hepatic lesion in rats using ultrasonic contrast agent.

The focal hepatic lesion caused by local injection of absolute alcohol in rats was evaluated with ultrasonic contrast agent and pathologic examination. Twenty adult Wistar rats weighing about 200 g were injected with absolute alcohol (0.05-0.1 mL each one) on the exterior left lobe of the liver under the monitoring of ultrasound. Pulse inversion harmonic imaging was used to evaluate the focal lesion after bolus injection of ultrasonic contrast agent (0.05 mL/200 g) through caudal vein. Seven days later, the focal lesion was studied again as before. The exterior left lobe of liver with focal lesion was incised and underwent pathologic examination. The results showed that all of the focal lesions could be defined clearly after bolus injection of the ultrasonic contrast agent under the mode of pulse inversion harmonic imaging. There was good correlation between the size of the focal lesion measured by ultrasound on the 7th day after the "ablation" under the mode of pulse inversion harmonic imaging and that gotten by pathologic examination (P = 0.39). The focus size measured by ultrasound right after the ablation was larger than that gotten by pathologic examination (P = 0.002). It was concluded that ultrasonic contrast agent plus pulse inversion harmonic imaging could be used to assess the size of the focal hepatic lesion caused by local injection of absolute alcohol in rats.

[The effect on the differentiation and maturation of dendritic cells by lupane acetate of cortex periplocae].

AIM: To investigate the effect of lupane acetate of cortex periplocae (CPLA) on the differentiation, maturation and immune activity of human peripheral blood mononuclear cell (PBMC)-derived dendritic cells (DCs) in vitro. METHODS: PBMC isolated from human peripheral blood was cultured with GM-CSF, IL-4 for 5 d and stimulated with TNF-alpha (as positive control) or CPLA to induce DCs. The morphological characteristics of DC were observed under inverted microscope and transmisson electron microscope. The expressions of CD1a, CD83, CD80 and CD86 were analyzed by flow cytometry. IL-12, IFN-gamma production in the culture supernatant of DCs was detected by ELISA. MTT method was used to determine the proliferation of T cells stimulated by DCs. RESULTS: After 10-days culture with cytokines and CPLA, PBMC developed into mature DCs with typical morphological characteristics and high expressions of CD1a, CD83, CD80 and CD86 on the cellular surface (P<0.05). CPLA enhanced IL-12 and IFN-gamma production by DCs (P<0.05). CPLA-treated DCs markedly stimulated proliferation of T cells (P<0.05). CONCLUSION: CPLA may induce the differentiation and maturation of DC, up-regulate cytokines production and increase the immune activity of DC.

[Construction and identification of recombinant vectors carrying herpes simplex virus thymidine kinase genes expressed in vascular endothelial cells].

BACKGROUND & OBJECTIVE: Blocking tumor angiogenesis is a promising tumoricidal approach. Herpes simplex virus-thymidine kinase (HSV-tk) suicide gene could effectively damage neoplastic vascular endothelial cells. However, the killing effect of currently used HSV-tk system is non-specific due to employing cytomegalovirus (CMV) as its promoter. Kinase domain insert containing receptor (KDR), a kind of vascular endothelial growth factor receptor, is highly expressed on tumor vascular endothelial cells, but lowly expressed on normal tissues. This study was to construct a recombinant adenovirus containing KDR promoter-mediated HSV-tk suicide gene, and analyze its specific expression on vascular endothelial cells. METHODS: KDR-tk fragment was subcloned into corresponding restriction sites of vector with directional cloning method using pAdeasy system to construct recombinant adenoviral plasmid containing KDR promoter-controlled HSV-tk gene (AdKDR-tk). After packaged and amplified in 293 cells, the virus was used to infect KDR-expressed human umbilical venous endothelial cells (HUVECs) and KDR-unexpressed liver cancer cell line HepG2. After administration of ganciclovir (GCV), proliferation of gene-transfected HUVECs and HepG2 cells was evaluated by MTT assay. RESULTS: According to the results of restriction digestion, reverse transcription-polymerase chain reaction (RT-PCR) and PCR, the length, position and orientation of inserted genes were correct. The titer of the recombinant adenovirus was 1 x 10 (10) pfu/ml. Under infection index of 100, with the concentration of GCV increased from 0 to 50 microg/ml, the survival rates of AdKDR-tk-transfected HUVECs and HepG2 cells decreased from 100% to (28.94+/-5.67)% and (75.45+/-2.91)%, respectively (P<0.01). CONCLUSION: Adenovirus-mediated transfection of KDR promoter-HSV-tk gene could direct endothelial specific gene expression.

[A comparative study on biocompatibility of acellular corneal stroma materials prepared by serial digestion methods].

OBJECTIVE: To observe the biocompatibility of the acellular corneal stroma materials prepared by three different methods. METHODS: Three different serial digestion methods were used to produce the acellular corneal stroma materials. The biocompatibility of the materials was investigated by the cell seeding and the materials were implanted into the rabbit corneal stroma layer. RESULTS: The cells in the materials 1 and 2 were not decellularized completely. The rabbit corneal fibroblasts died on the materials 1 and 2 after the cell seeding for 3-4 days. An obvious rejection could be observed after the implantation. The cells in material 3 were decellularized completely and the collagen fibers or elastic fibers were reserved integrally, showing a typical three-dimensional network. The rabbit corneal fibroblasts could expand on the materials in vitro. No obvious rejection could be observed and the materials were gradually absorbed. CONCLUSION: The acellular porcine cornea stroma materials prepared by trypsin-Dnase-Rnase are suitable for reconstruction of the tissue engineered cornea.

[Transcription regulation of 5-Aza-2'-deoxycytidine on maspin gene demethylation in RKO human colorectal cell line].

OBJECTIVE: To detect the methylation status of 5'CpG island in the core promotor of maspin gene in RKO human colorectal cell line, and to explore the transcription regulation of DNA 5'CpG island demethylation on maspin tumor suppressor gene and its effect on the growth of cancer cell. METHODS: The status of 5 'CpG island methylation of maspin gene in RKO human colorectal cell line was analyzed using methylation specific polymerase chain reaction (MSP). After treated with a specific demethylating agent, 5-Aza-2'-deoxycytidine, reverse transcription polymerase chain reaction (RT- PCR) was used to examine maspin gene expression. Cell proliferation was evaluated using MTT assay, distribution of cell cycle and rate of apoptosis were determined using flow cytometry. RESULTS: The 5'CpG island methylation in the core promotor of maspin gene was detected in RKO human colorectal cell line. After treatment with three different concentration of 5-aza-2'-deoxycytidine, the expression of maspin mRNA increased 10.89, 16.91, 23.97 times respectively. MTT array showed the proliferation activity of RKO cell line was obviously reduced after 5-aza-2'-deoxycytidine treatment. The cells were arrested in G(0)/G(1) phase, and the apoptosis rates were 5.17%, 8.71% and 11.23% respectively compared with control group. CONCLUSION: The 5'CpG island methylation is probably responsible for maspin expression silencing in RKO human colorectal cell line, 5-aza-2'-deoxycytidine may effectively cause demethylation and inhibit the growth of tumor cell by reactivating the gene transcription silenced by aberrant hypermethylation.

[Biological characteristics of an established model of ovarian cancer in mice and its homologous cell lines].

BACKGROUND & OBJECTIVE: There are no specific methods for early diagnosis of ovarian cancer, recurrence prevention and drug-resistance. The experimental mouse model of ovarian cancer could help to reveal the biological and genetic features of ovarian cancer, and provide rational basis for further intervention strategy. This study was to establish a model of ovarian cancer in mice and homologous cell line, and analyze its biological characteristics. METHODS: Ovarian cancer was developed in 8-week-old female F1 (C57BL/6N x C3H/He) mice by a single whole-body neutron irradiation of 2.7 Gy from a (252)Cf source. A metastatic cell line was established through serial subcutaneous transplantation of the primary tumor for 11 generations, and then tumor cells were transferred to in vitro cultivation. These cells were cloned for more than 6 months. The biological characteristics of the tumors and the homologous cell line were determined by cellular and molecular biological techniques. RESULTS: The grafted tumors in mice were successively passaged for 11 generations with a successful inoculation rate of 96% during 23 months. A tumor cell line OV99 isolated from the grafted tumors was established after 6 months and grew steadily. Morphologic characters and ultrastructures of OV99 cells were accorded with those of ovarian cancer epithelia. The chromosomal analysis of OV99 cells revealed aneuploid pattern of 76 chromosomes. Flow cytometry (FCM) and reverse transcription-polymerase chain reaction (RT-PCR) showed same features between OV99 cells and positive control ovarian cancer cell line OVHM, including distribution of cell cycle, rapid growth rate and the expression of P21, P185, P53, proliferating nuclear cell antigen (PCNA) and Cyclin D proteins, and MAGE-1 and MAGE-3 mRNA. CONCLUSION: Establishment of the ovarian carcinoma animal model in mice and OV99, a cell line owns biologic characteristics of ovarian cancer cells, provides experimental materials for further investigation of ovarian carcinoma.

[Response to dynamic strain in human periosteal cells grown in vitro].

The objective of this program is to investigate the biological effect of dynamic strain on human periosteal cells in vitro. Using a well-established model, the Flexercell unit, we placed mechanical stress (50,000 microstrain, 1 Hz and sine wave) on human periosteal cells grown in collagen coated flexible membrane. The time points of proliferative and differentiative properties were assessed by means of cell counting, thymidine incorporation, synthesis of alkaline phosphatase and osteocalcin, and long term of mechanical load induced calcium nodules formation was also demonstrated. The results showed that the application of highly controlled strains exerted a significant effect on human periosteal cells by up regulation of osteogenic properties rather than exercised an influence on proliferation. The results suggested that the promoting effects of dynamic strain on human periosteal cells probably contribute to the biological function of mechanical loading bearing.

[Study of cytokines secreted by tumor-draining lymph node cells from lung cancer patients induced with different stimulators].

AIM: To investigate the influence of different stimulators on cytokines secretion of tumor-draining lymph node (TDLN) cells and study antitumor effects and mechanism of TDLN cells. METHODS: IL-12 p70, IFN-gamma, TNF-alpha and NO which were secreted by TDLN cells induced by different stimulators (IL-2 group, IL-2+autologous tumor antigen group, IL-2+GM-CSF+IL-4+autologous tumor antigen group and IL-2+GM-CSF+IL-4+LPS group) were detected by ELISA and Griess after TDLN cells were stimulated for 7, 14 and 21 d. RESULTS: The rate of CD83(+) cells of TDLN cells induced by IL-2+GM-CSF+IL-4+autologous tumor antigen and IL-2+GM-CSF+IL-4+LPS was significantly increased. The TDLN cells in the two groups secreted much more IL-12 p70, IFN-gamma and TNF-alpha than IL-2 group and IL-2+autologous tumor antigen group. The TDLN cells stimulated by IL-2+GM-CSF+IL-4+LPS secreted significantly more NO than the other three groups. TDLN cells in each group secreted IL-12 p70, IFN-gamma and NO at the highest level after TDLN cells were stimulated for 14 d. CONCLUSION: Different stimulators can make TDLN cells produce different number of CD83(+) cells (DC), which gives TDLN cells different antitumor activity.

[Biological and antitumor effects of CD40L on ovarian cancer cell line OVHM].

BACKGROUND & OBJECTIVE: Accumulative evidences have indicated the importance of CD40-CD40L receptor-ligand pair in the initiation and regulation of human immune response. Whether triggering CD40-CD40L signaling pathway could induce antitumor effects in ovarian cancer cells is still unclear. This study was to investigate the biological and antitumor effects of CD40L on ovarian cancer cell line OVHM (CD40-positive). METHODS: CD40L cDNA was transfected into OVHM cells by lipofectamine 2000. Immunofluorescent technique and flow cytometry were used to evaluate the expression of CD40, CD40L, MHC-I, MHC-II, CD80, CD86. Cell proliferation was detected by MTT assay. CD40L/OVHM and OVHM cells were inoculated hypodermically into C57BL/6N x C3H/He F1 (B6C3F1) mice and BALB/c nude mice simultaneously. Tumor volume was measured. The production of IFN-gamma secreted by the spleen cells from CD40L/OVHM and OVHM cell-inoculated mice co-cultured with 20 Gy-irridiated OVHM cells were measured with enzyme-linked immunosorbent assay (ELISA). RESULTS: Expression of CD40L in OVHM/CD40L cells was significantly higher than that in OVHM cells (P<0.05). Moreover, the expression of co-stimulatory molecules CD80 and CD86 of CD40L/OVHM was increased (P<0.05). The proliferation rate of parental and CD40L-expressed OVHM cells were not different. Although, delayed tumor growth was observed when CD40L-expressing OVHM cells and OVHM cells were injected simultaneously and contralaterally, the tumor growth of OVHM and CD40L/OVHM in BALB/c nude mice was not different. The amounts of IFN-gamma secreted by the co-culture of irradiated OVHM cells with spleen cells from mice injected with CD40L/OVHM were much higher than those from OVHM/parental cell inoculated and naive mice. CONCLUSION: The forced expression of CD40L cDNA in OVHM cells can increase the immunogenicity, and thus develop antitumor effects against OVHM-incubated mice. In vivo immunization of immunocompetent mice with CD40L/OVHM improves the generation of cytotoxic lymphocytes against parent ovarian cancer cells.

[The reconstruction of bioengineer active corneal stroma and its biocompatibility study].

This paper aims to explore the biocompatibility of bioengineer active corneal stroma (BACS), as the biological carrier for cornea reconstruction, to provide the basis for future study on clinic application. The cells and immunogenic components of cornea stroma were removed through different extract methods. A complex of functional corneal stroma cells and acellular corneal stroma was used to reconstruct BACS. Their morphological characteristics and ultrastructures were observed with transmission electron microscope. The complex was grafted into interlamellar stromal pockets. Cells were labeled by BrdU to examine the survival and conversion after grafting. The cells could survive and proliferate in acellular corneal stroma. All the nuclei of the corneal stromal cells showed positive labeling with BrdU in the BACS. After 4 weeks, BACS became transparent; after 8 weeks, the bioengineer active cornea stroma was fully reconstructed.

[A novel anti-DNA antibody modified coronary stent for site-specific plasmid DNA delivery].

OBJECTIVE: To explore the feasibility of using an endovascular metal stent as a highly efficient and site-specific gene delivery system. METHODS: Stents were formulated with a collagen coating. Anti-DNA monoclonal antibodies were covalently bound to the collagen surface by a cross linking reagent of N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP). Binding capacity and stability of antibody and plasmid DNA on stents were quantified by radioactive labeling. The gene transduction efficiency was evaluated in cell culture and in rabbits. RESULTS: The amount of antibodies binding on collagen matrix through SPDP reaction was 15 times higher than that of through physical absorption (P < 0.005). The binding stability of plasmid was significantly better than the control groups (P < 0.01). There was no harmful effect on cell growth with the anti-DNA antibody modified stents. The stents retrieved from cell culture after 72 hours of incubation in A10 cells showed numerous transducted cells only infiltrating the surface coating indicating a highly localized and efficient gene delivery pattern. Results of in vivo gene transfer by this modified stent revealed (2.8 +/- 0.7)% of total cells transduction and the higher transduction location was neointimal layer (about 7%). No distal spread of vector was detectable in the anti-DNA antibody modified stent implantation animals. CONCLUSIONS: Anti-DNA antibody modified stents represent a novel highly efficient and site-specific gene delivery system which can deliver various kinds of plasmid vectors. The release of plasmid DNA tethered on the stents could be controlled in some conditions. This novel system provided a novel platform for cardiovascular site-specific gene therapy.

[Application and reliability of balloon occlusion test in treatment of cerebrovascular diseases].

OBJECTIVE: To study the reliability of applying balloon occlusion test (BOT) to evaluate the compensation capacity of cerebral collateral circulation, and investigate the validity of different criterion about BOT and techniques to improve the security of BOT. METHODS: A retrospective analysis was conducted on the results of 86 patients, 39 males and 47 females, aged 49.8 (13 - 72), 67 suffering from intracranial aneurysm and 19 from traumatic carotid cavernous fistula, who received BOT, and on the results of 38 of the 66 patients who accepted artery occlusion after the BOT. RESULTS: The positive rate of diagnosis was 10.47% and the negative rate was 89.53% by conventional BOT; whereas the positive rate of diagnosis was 36.05% and the negative rate was 63.95% with combined cross circulation and conventional test. The brain ischemia rate after cerebral artery occlusion therapy was 8.33%. The BOT-related complication rate was 3.49%. CONCLUSION: (1) The current BOT technique can basically provide reliable results. (2) The cross circulation test and hypotensive provocative test can effectively improve the specificity of BOT. (3) More perfect BOT criterion for the prediction of cerebral tolerance is still to be worked out. Combination of the conventional method and advanced imaging technique and search for an objective and quantificational criterion would be of reward.
