Unsupervised multiscale clustering of single-cell transcriptomes to identify hierarchical structures of cell subtypes.

BACKGROUND: Cell clustering is an essential step in uncovering cellular architectures in single-cell RNA sequencing (scRNA-seq) data. However, the existing cell clustering approaches are not well designed to dissect complex structures of cellular landscapes at a finer resolution. RESULTS: Here, we develop a multiscale clustering (MSC) approach to construct a sparse cell-cell correlation network for unsupervised identification of de novo cell types and subtypes across multiple resolutions. Based upon simulated silver- and gold-standard data as well as real scRNA-seq data in diseases, MSC demonstrates significantly improved performance compared to established benchmark methods and reveals a biologically meaningful cell hierarchy to facilitate the discovery of novel disease-associated cell subtypes and mechanisms. CONCLUSIONS: We present MSC as a new single-cell multiscale clustering framework as a powerful tool for advancing discoveries in disease-associated cell populations using single-cell sequencing data.

Obstetrics and gynecology residents as teachers of medical students: predictors of excellence.

OBJECTIVE: The purpose of this study was to assess variables that might predict which intern candidates will become excellent teachers of medical students. STUDY DESIGN: This retrospective cohort study compared demographic characteristics, previous work experience, United States Medical Licensing Examinations scores, honors on core clerkships, membership in Alpha Omega Alpha, and match list ranking of 43 residents to identify predictors of excellent teaching evaluations during residency. RESULTS: Fifteen residents (35%) were identified as excellent teachers. They were more likely to have had previous work experience, to be older, or to be male. They were not more likely to have higher United States Medical Licensing Examinations test scores, more honors grades, Alpha Omega Alpha membership, or a higher rank list position. CONCLUSION: Work experience, age, and male gender are associated with increased likelihood of being identified as an excellent teacher by medical students. Programs in which residents have a significant role as teachers of students may consider these factors in the residency selection process.

Comparative study on rapid dot-immunogold staining and two immunogold silver staining assays for diagnosing schistosomiasis japonica.

A fast, specific, sensitive, convenient, and economical rapid-dot-immunogold staining (R-Dot-IGS) assay was used to detect serum antibodies in patients infected with Schistosoma japonicum. The soluble egg antigen of Schistosoma japonicum was added onto microspore membrane. After pre-reacting and blocking, the serum to be detected and sheep anti-human IgG labeled with chloroauric acid were added sequentially. The assay took 15 minutes. For comparison, the dot-immunogold silver staining (Dot-IGSS) and rapid micro-volume Dot-IGSS (RM-Dot-IGSS) assay were also performed. The positive rate to detect the serum of schistosomiasis japonica by the R-Dot-IGS, Dot-IGSS and RM-Dot-IGSS assay was 98%, 98% and 100%, respectively. Samples from 50 healthy controls, 10 cases of clonorchiasis, and 10 cases of paragonimiasis showed negative reactions except for one case of clonorchiasis with RM-Dot-IGSS assay. Compared with Dot-IGSS and RM-Dot-IGSS, R-Dot-IGS assay has similar sensitivity and specificity, but the latter is quicker, simpler, and cheaper. Therefore, R-Dot-IGS is strongly recommended for rapid diagnosis of schistosomiasis japonica both in epidemiological study and in the clinic.

New technique for in-situ measurement of ammonia distribution in industrial combustion systems.

A new technique of a laser gas monitoring system with a linear Charge-Coupled Device (CCD) has been developed to measure in-situ distribution of ammonia concentrations at various temperatures. The concentration is determined by measuring absorption with a tunable 13CO2 waveguide laser that is frequency modulated between two adjacent emission lines. One line coincides with a strong ammonia absorption line, while the other is used as a reference. By its referencing the system is auto-calibrated continuously. The high spectral resolution of the laser suppresses interference by other species. Multi-light path is used to achieve distribution of ammonia concentration in the flue gas channel, which is inhomogeneous. The linear CCD is used to replace detectors, which can avoid errors caused by parameter inconsistency of multi-detector and enable lower cost, fast and exact data acquisition and processing with computer by video interface. The measuring technique presented in this paper can be used to protect industrial environment against pollution in the chemical industry, several power plants and waste incinerators.
