The Adaptive Roles of Active Transposable Elements in Insect Hosts.

Active transposable elements (TEs) are capable of generating new insertions in genomes and have historically been viewed as genomic parasites due to their largely detrimental or neutral effects. However, emerging evidence suggests that these elements also play a crucial role in driving adaptive evolution in insects. This mini-review synthesizes recent findings on how active TEs contribute to insect adaptation through various mechanisms, including regulation of gene expression, structural variation, and epigenetic effects. Notable examples of adaptation driven by active TEs include their roles in insecticide resistance, morphological adaptations, tolerance to harsh climates, and antiviral immune responses. We argue that while host silencing mechanisms, such as the piRNA pathway, tightly regulate TE activity to minimize harmful effects, the context-dependent activation of active TEs can generate beneficial genetic variation that enhances insect adaptations to anthropogenic and climatic pressures. Future research that integrates long-read sequencing, single-cell omics, and gene editing techniques will provide a robust mechanistic foundation for understanding the adaptive significance of active TEs in insects, with important implications for pest management, pollinator protection, and evolutionary biology.

LDHC4 promotes ovarian cancer progression through H4K12 lactylation to regulate PGK1 expression and modulate glycolysis.

OBJECTIVE: Ovarian cancer (OC) pathogenesis involves metabolic and epigenetic alterations, yet the underlying mechanisms remain unclear. Here, we sought to investigate the role and regulatory mechanism of lactate dehydrogenase C4 (LDHC4) in OC progression. METHODS: Multi-omics approaches were employed, including analyses of The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx) cohorts, tissue microarrays, molecular biology experiments, and in vivo mouse models. LDHC4 expression was modulated in OC cell lines (A2780 and ES-2) to assess its effects on proliferation, migration, invasion, and metastasis. Lactylproteomic profiling, cleavage under targets and tagmentation (CUT&Tag), and chromatin analyses were conducted to explore epigenetic mechanisms. The functional role of the downstream glycolytic enzyme phosphoglycerate kinase 1 (PGK1) was examined using pharmacological inhibition. Molecular docking and xenograft models were used to evaluate the therapeutic potential of targeting H4K12lac (lactylation of histone H4 at lysine 12). RESULTS: LDHC4 was significantly overexpressed in OC tissues and associated with poor overall survival (OS) (hazard ratio [HR]&#x2009;=&#x2009;4.017, 95% confidence interval [CI]: 2.308&#x2013;6.989, P&#x2009;<&#x2009;0.0001). It promoted proliferation, migration, invasion, and metastasis in vitro and in vivo. LDHC4 overexpression increased global lactylation, notably upregulating H4K12lac, which was enriched at the PGK1 promoter. Inhibition of PGK1 with CBR-470-1 (half-maximal inhibitory concentration [IC&#x2085;&#x2080;]&#x2009;=&#x2009;14.56 &#xb5;M) suppressed OC growth and metastasis. Importantly, Elbasvir, identified as a high-affinity H4K12lac inhibitor, significantly reduced tumor burden in mouse xenografts across multiple doses (10&#x2013;20&#xa0;mg&#xb7;kg&#x207b;&#xb9;) and downregulated H4K12lac and Ki-67 expression. CONCLUSION: We conclude that LDHC4 promotes OC progression via lactylation-mediated epigenetic upregulation of PGK1. Targeting this pathway through H4K12lac inhibitors such as Elbasvir thus emerges as a viable therapeutic approach for OC.

Determination of phthalates in water samples using polyaniline-based solid-phase microextraction coupled with gas chromatography.

A simple solid-phase microextraction (SPME) device, coupled with gas chromatography-flame ionization detection (GC-FID), was developed to detect trace levels of phthalates in environmental water samples. Polyaniline (PANI) was chosen as the sorbent for the SPME device and was electrochemically deposited on a stainless steel wire to achieve high thermal and mechanical stability. The porous structure of the PANI film, characterized by scanning electron microscopy (SEM), suggested large extraction capability. Key parameters were optimized and five phthalates were selected to evaluate the SPME-GC procedures. The method was also applied to the analysis of lake and river water samples. Control experiments were carried out using commercial polyacrylate (PA) fiber. The new PANI-SPME-GC method offers high accuracy, precision and sensitivity and low detection limits. Thus, the method developed could be used as a new way to monitor the trace levels of phthalates in water medium. A possible extraction mechanism was investigated using electrochemical impedance spectroscopy (EIS).

Low-temperature preparation and microwave photocatalytic activity study of TiO2-mounted activated carbon.

TiO2 thin films were deposited on granular activated carbon by a dip-coating method at low temperature (373 K), using microwave radiation to enhance the crystallization of titania nanoparticles. Uniform and continuous anatase titania films were deposited on the surface of activated carbon. BET surface area of TiO2-mounted activated carbon (TiO2/AC) decreased a little in comparison with activated carbon. TiO2/AC possessed strong optical absorption capacity with a band gap absorption edge around 360 nm. The photocatalytic activity did not increase when the as-synthesized TiO2/AC was thermally treated, but was much higher than commercial P-25 in degradation of phenol by irradiation of electrodeless discharge lamps (EDLs).

Photoelectrocatalytic treatment of pentachlorophenol in aqueous solution using a rutile nanotube-like TiO2/Ti electrode.

Taking pentachlorophenol (PCP) as a reference, we investigated the photoelectrocatalytic degradation of organic pollutants using a rutile nanotube-like TiO(2)/Ti film electrode. The nanotube-like TiO2 electrode was prepared by first oxidizing the surface of a titanium sheet to form rutile TiO2 and then treating it to form the tubular structure in NaOH aqueous solution. The occurrence of PCP degradation was indicated by the decrease in pH, concentration of PCP and TOC, and by the formation of chloride ions. The photoelectrochemical (PEC) efficiency of the nanotube-like TiO2/Ti electrode has been determined in terms of degradation of PCP and the incident photo-to-current conversion efficiency (IPCE). The experimental results showed that PCP could be degraded more efficiently by a photoelectrocatalytic process than by a photocatalytic or electrochemical oxidation alone. A significant photoelectrochemical synergetic effect was observed. The kinetic constant of PEC degradation of PCP using a nanotube-like TiO2 electrode was over 60% higher than that using a TiO2 film electrode. It is noted that chloride ion and hydrogen ion concentration increased with irradiation time in the PEC degradation of PCP; PCP was gradually mineralized and the complete minimization of PCP took more time than its degradation.

Degradation of atrazine by microwave-assisted electrodeless discharge mercury lamp in aqueous solution.

The present study investigates the degradation of atrazine (2-chloro-4-(ethyl amino)-6-isopropyl amino-s-triazine) in aqueous solution by a developed new method, namely by means of a microwave-assisted electrodeless discharge mercury lamp (MW-EDML). An experimental design was conducted to assess the influence of various parameters: pH value, initial concentration, amount of EDML, initial volume and coexisted solvent. Atrazine was degraded completely by EDML in a relatively short time (i.e. t(1/2)=1.2 min for 10 mg/l). Additionally, the identification of main degradation products during atrazine degradation process was conducted by gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-mass spectrometry (LC-MS). This study proposes the degradation mechanism including four possible pathways for atrazine degradation according to the degradation products.

Ultrasonic metamaterials with negative modulus.

The emergence of artificially designed subwavelength electromagnetic materials, denoted metamaterials, has significantly broadened the range of material responses found in nature. However, the acoustic analogue to electromagnetic metamaterials has, so far, not been investigated. We report a new class of ultrasonic metamaterials consisting of an array of subwavelength Helmholtz resonators with designed acoustic inductance and capacitance. These materials have an effective dynamic modulus with negative values near the resonance frequency. As a result, these ultrasonic metamaterials can convey acoustic waves with a group velocity antiparallel to phase velocity, as observed experimentally. On the basis of homogenized-media theory, we calculated the dispersion and transmission, which agrees well with experiments near 30 kHz. As the negative dynamic modulus leads to a richness of surface states with very large wavevectors, this new class of acoustic metamaterials may offer interesting applications, such as acoustic negative refraction and superlensing below the diffraction limit.

Raman enhancement factor of a single tunable nanoplasmonic resonator.

We have developed a novel technique to precisely determine the Raman enhancement factor in single nanoplasmonic resonators (TNPRs). TNPRs are lithographically defined metallodielectric nanoparticles composed of two silver disks stacked vertically, separated by a silica layer. At resonance, the local electromagnetic fields are enhanced at the TNPR surface, making it an ideal surface-enhanced Raman scattering (SERS) active substrate. The ability to control the dimensions of the metallic and dielectric layers offers the unique advantage of fine-tuning the plasmon resonance frequency to maximize the enhancement of the Raman signal. Furthermore, by selective shielding of the outer surface of the metallic structure, the efficiency can be further enhanced by guiding the molecular assembly to the locations that exhibit strong electromagnetic fields. We experimentally demonstrate SERS enhancement factors of (6.1+/-0.3)x10(10), with the highest enhancement factor being achieved by using an individual nanoparticle. By using nanofabrication techniques, we eliminate the issues such as large size variations, cluster aggregation, and interparticle effects common in preparing SERS substrates using conventional chemical synthesis or batch fabrication methods. TNPRs produce very controllable and repeatable SERS signals at the desired locations and, thus, make an ideal candidate for device integration.

[Indirect ELISA for the pyrethroids-permethrin residue detection].

In order to detect the permethrin (Py) residue in environment samples, Ovalbumin(OVA)was used as protein carrier to couple with semi-antigen permethrin by active ester method. Indirect competitive ELISA (icELISA) was established. The most appropriate titration of coating antigen was 0.45 microg/mL and optimal dilution was 1/2 500 correspondingly. The standard curve of icELISA was also established and the curve indicated that the lowest detection limit was 0.l microg/mL, the curve had a favorable linear relation with the concentration range of 10 to approximately 800 microg/mL, recoveries of permethrin (>97%) were satisfactory.

[Microwave assisted photocatalytic degradation of phenol in aqueous solution].

Experimental study on microwave assisted photocatalytic degradation of phenol in aqueous solution was carried out with modified domestic microwave oven, electrodeless discharge lamps (EDLs) and TiO2 catalyst. Results showed that the removal rate of phenol in the MAPC process was 92% after 30 min reaction. Loss of total organic carbon (TOC) was 84% . The optimal conditions for the MAPC method were 10 mg/L phenol, 900 W MW output energy, 50 mL solution volume, EDLs-3, 1-4 g/l. catalyst dosage and 15 mL/min of flowing velocity in the circulating mode. The MAPC process was promising in treating phenolic waste water.

Photocatalytic degradation of methylene blue in TiO2 aqueous suspensions using microwave powered electrodeless discharge lamps.

Photocatalytic degradation of methylene blue (MB) in TiO(2) aqueous suspensions using microwave (MW) powered electrodeless discharge lamps (EDLs) was studied. MB of initial concentration 100 mg/l was mainly decomposed in the process of photocatalytic degradation using EDLs (PCD/EDLs) after 15 min of irradiation. The corresponding mineralization efficiency was 45%. The influence of factors as EDLs, solution volume and TiO(2) catalyst dosage on the decomposition of MB in the PCD/EDLs process was also investigated. The optimal decomposition efficiency was observed when EDLs-4 (four 10 mm x 50 mm EDLs), solution volume of 50 ml and TiO(2) catalyst dosage of 1-4 g/l were used in the study. The PCD/EDLs process was promising in treating MB polluted water.

Sub-diffraction-limited optical imaging with a silver superlens.

Recent theory has predicted a superlens that is capable of producing sub-diffraction-limited images. This superlens would allow the recovery of evanescent waves in an image via the excitation of surface plasmons. Using silver as a natural optical superlens, we demonstrated sub-diffraction-limited imaging with 60-nanometer half-pitch resolution, or one-sixth of the illumination wavelength. By proper design of the working wavelength and the thickness of silver that allows access to a broad spectrum of subwavelength features, we also showed that arbitrary nanostructures can be imaged with good fidelity. The optical superlens promises exciting avenues to nanoscale optical imaging and ultrasmall optoelectronic devices.

Solid-phase extraction of methadone enantiomers and benzodiazepines in biological fluids by two polymeric cartridges for liquid chromatographic analysis.

The aim of this work was to present the advantages of two polymeric cartridges (Oasis HLB from Waters and Abselut Nexus from Varian) for the solid-phase extraction of methadone enantiomers and its major metabolite, 2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine (EDDP) and of some benzodiazepines (diazepam, flunitrazepam, nitrazepam, oxazepam) in serum and urine in comparison with classical C18-bonded-silica cartridges or liquid extraction. After addition of serum or urine samples, these two cartridges were washed with a water-methanol mixture (95:5, v/v) and eluted with diethylether. After rapid evaporation, the residue was regenerated with mobile phase and injected either in a chiral column (Cyclobond I-2000 RSP) for methadone enantiomers and its metabolite or in a reversed-phase column (Symmetry Shield RP8) for benzodiazepines. The results showed that the chromatograms of blank serum and urine were cleaner than those obtained from classical solid-phase extraction or liquid extraction. The recoveries from these two polymeric cartridges were higher (95-102%) than those obtained by the two previous classical methods and the total time for extraction and solvent evaporation was also shorter (about 6-7 min). For methadone and benzodiazepine extraction, the use of acidic or alkaline buffer was not necessary.

Focusing surface plasmons with a plasmonic lens.

We report the focusing of surface plasmon polaritons by circular and elliptical structures milled into optically thick metallic films or plasmonic lenses. Both theoretical and experimental data for the electromagnetic nearfield is presented. The nearfield is mapped experimentally using nearfield scanning optical microscopy and plasmonic lithography. We find that the intensity at the focal points of the plasmonic lenses increases with size.

Characterization of graphislactone A as the antioxidant and free radical-scavenging substance from the culture of Cephalosporium sp. IFB-E001, an endophytic fungus in Trachelospermum jasminoides.

The extracts derived from cultures of 1626 endophytic strains harbored in Trachelospermum jasminoides were assayed for more potent antioxidant and/or free radical-scavenging agents. The free radical-scavenging assessment was carried out using l,l-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radical assays, and the antioxidant actions on linoleic acid and human low-density lipoprotein (LDL) models. After extensive spectroscopic analyses, graphislactone A was characterized as the most bioactive secondary metabolite of endophytic Cephalosporium sp. IFB-E001 with its free radical-scavenging (in a dose-dependent manner) and antioxidant activities ascertained in vitro to be stronger than those of butylated hydroxytoluene (BHT) and ascorbic acid, the two positive references coassayed in the study. From the demonstrated efficacy of graphislactone A in preventing and protecting against oxidative injury, it can be predicted that this metabolite could be a potential agent in the management of oxidative damage-initiated diseases.

Free radical scavenging and antioxidant activities of EPS2, an exopolysaccharide produced by a marine filamentous fungus Keissleriella sp. YS 4108.

The reactive oxygen species (ROS) and free radical-initiated reactions are ascertained to play multiple roles in degenerative or pathological events such as aging, cancer, heart dysfunction and Alzheimer's disease. EPS2 with a mean molecular weight of 1.3 x 10(5) was characterized as an antioxidant exopolysaccharide from the broth of a marine filamentous fungus Keissleriella sp. YS 4108. Compositionally, it is composed of galactose, glucose, rhamnose, mannose and glucuronic acid in an approximate proportion of 50:8:1:1:0.4. The radical eliminating and antioxidant actions of the glycan was assessed in different in vitro systems showing that EPS2 exhibited profound scavenging activities in superoxide radical. As a reinforcement of the action, similar radical scavenging effects of EPS2 were also discerned with both site-specific and non site-specific hydroxyl radical using the deoxyribose assay method. Moreover, EPS2 effectively blocked as well the non site-specific strand-breaking of DNA induced by the Fenton reaction at concentrations of 0.1 and 1 mg/mL. Further investigation of the effect of EPS2 on human low density lipoprotein (LDL) system demonstrated that it significantly inhibited copper-mediated oxidation of LDL in a dose-dependent manner. These results suggest that EPS2, possessing pronounced free radical scavenging and antioxidant activities, could be of considerable preventive and therapeutic significance to some life-threatening health problems such as cancer, atherogenesis and Alzheimer's disease which pathologically initiated by the presence of free radicals leading to the inevitable peroxidation of important biomolecules.

Effect of temperature on enantiomer separation of oxzepam and lorazepam by high-performance liquid chromatography on a beta-cyclodextrin derivatized bonded chiral stationary phase.

A reversed-phase high-performance liquid chromatography (HPLC) method with beta-cyclodextrin (beta-CD) derivatized as chiral stationary phase is used to directly separate oxazepam (Oxa) and lorazepam (Lor) enantiomers. The effect of temperature on the direct HPLC separation of Oxa and Lor enantiomers is studied for the commercially available beta-CD derivatized bonded chiral stationary phase. Chromatographic peak coalescence, appearing as a plateau between the resolved peaks, is observed at column temperatures of above 13 degrees C. Peak coalescence on the beta-CD derivatized bonded column is attributable to racemization of the Oxa enantiomer. By reducing the column temperature to 13 degrees C, the enantiomeric composition of Oxa and Lor could be determined on the chiral column. This method is expected to be useful for the resolution of 3-hydroxybenzodiazepines. At the same time, the separation mechanism is studied by calculating the thermodynamic parameters. The results reveal that the separation of Oxa and Lor enantiomer is a case of enthalpy-controlled separation, inclusion mechanism does not control the separation. The interaction between Oxa and beta-CD is an additionally strong pi-pi interaction or hydrogen bonding, but that between Lor or beta-CD derivatized is a weak pi-pi interaction or hydrogen bonding.

[The application of the gene-recombinated cell in the detection of PCBs compounds in environmental samples].

To develop novel and rapid bioassay systems for detection of PCBs in environmental and biological samples, two lines of gene-recombinated cells containing reporter gene, green fluorescent protein (GFP) and luciferase (Luc), were be chosen to detect the PCBs in the water, sediment and biological samples from environment. The result showed that the correlation between RFU of GFP and Luc activity and PCBs standard sample concentration was fine, r was 0.99188 and 0.98239 respectively. Compared with the instrument analysis with GC-ECD, the correlation between RFU of GFP and Luc activity and PCBs concentration in environmental and biological extracts was also fine. The method offered a new way of screening and semi-quantitative bioassay for PCBs compounds in environmental and biological samples.
