In vitro assays for adhesion and migration of osteoblastic cells (Saos-2) on titanium surfaces.

The first event occurring at the boundary between a metal implant and living tissue is the attachment of cells onto the metal surface of the implant. The attachment characteristics of the metal in this situation are critical in determining its biocompatibility and usefulness as artificial bone and tooth implants. Using the human osteosarcoma cell line Saos-2, we attempted to establish simple and reliable methods for evaluating the attachment of cultured osteoblastic cells onto titanium samples that had been subjected to various surface treatments. Fluorescence actin imaging showed that cells cultured on titanium with hydrofluoric acid etching (HF-Ti) exhibited delayed spreading of their cytoplasm, as compared to cells cultured for the same length of time on nitrided titanium or physically polished titanium. The HF-Ti-cultured cells also exhibited poor assembly of focal contacts, as visualized by vinculin immunofluorescence. Furthermore, in motility assays based on an in vitro wound model, cells cultured on HF-Ti migrated more slowly than cells cultured on other titanium surfaces. These data suggest that Saos-2 cells attach less effectively to the HF-Ti surface. The methods described in this study should be useful for assessing the initial interactions of cultured cells with various materials, including metals.

[Controlled study on treatment of cervical spondylopathy of the nerve root type with acupuncture, moxibustion and massage as main].

OBJECTIVE: To probe into clinical value of comprehensive program of acupuncture, moxibustion and massage as main for treatment of cervical spondylopathy of the nerve root type. METHODS: Five centers, single blind, randomized controlled method were used, 660 cases were divided into a treatment group of 317 cases and a control group of 311 cases. They were treated respectively with comprehensive program of acupuncture, moxibustion and massage as main, and comprehensive program of physical therapy as main. Establish syndrome detection scale and multiply dimensional effect assessment indexes, and evaluate the therapeutic effects and safety. RESULTS: The cured rate, the cured-markedly effective rate were 42.9%, 64.4% in the treatment group, respectively, better than 16.7%, 36.3% in the control group (P<0.01); after treatment of 2 weeks, clinical symptoms improved in the both groups, but the treatment group was better than the control group in the improvement degrees of neck-shoulder-limb pain, neck rigidity, abnormality of cervical anteflexion, etc. (P<0.01 or P<0.05); the treatment group was shorter than the control group in the time of producing the effect and therapeutic course (P<0.01). CONCLUSION: Comprehensive program of acupuncture, moxibustion and massage as main is safe and effective for treatment of cervical spondylopathy, with a better therapeutic effect compared with the comprehensive program of physical therapy.

[Surveys on the implementation of "rules on stop smoking in the public places in Beijing" from 1997 to 2004].

OBJECTIVE: To find out the situation of the implementation on "rules on stop smoking in the public places in Beijing", a study was carried out to compare the results from 1997 and 2001. METHODS: Thousands of residents aged above 15 in a family were sampled by stratified sampling, and 4 districts were selected as study locales. RESULTS: (1) About 40% of the department or school leaders considered much about the work on banning of smoking with 70 % of the departments or schools having posters of signs on 'ban on smoking' in the meeting rooms. (2) Some of the departments or schools had inspectors looking after the issue. (3) 60% of the people had never or 10% had seen that the inspectors ever criticizing or punishing the smokers who violating the rules. (4) People still often saw some one would smoke in public (the rates were 19.53% , 37.89% , 6.35% respectively in 1997,2001,2004 ). (5) People received the information of these rules through TV news or newspaper. CONCLUSION: Although success had been achieved since the implementation of the Rules, it did not seem to have had substantial progress.

[Multicenter randomized controlled study on acupuncture-massage comprehensive program for treatment of cervical spondylosis of arterial type].

OBJECTIVE: To probe into clinical value of the acupuncture-massage comprehensive program for treatment of cervical spondylosis of arterial type (CSA). METHODS: By using three-center, single-blind, randomized, controlled method, the eligible subjects were randomly divided into an acupuncture-massage group, an acupuncture group, a massage group, 60 cases in each group. They were treated with acupuncture-massage comprehensive program, simple acupuncture and simple massage, respectively. And multiple-dimensional indexes were established to evaluate the therapeutic effect and safety. RESULTS: The baselines of the general data were no different among the 3 groups. The cured-markedly effective and the effective rate were 68.3% and 88.3% in the acupuncture-massage group, 51.7% and 75.0% in the acupuncture group, and 50.0% and 76.7% in the massage group, respectively, the acupuncture-massage group being better than the other 2 groups (P<0.05); after treatment, the total cumulative scores for symptoms in the 3 groups decreased significantly (all P<0.01), the acupuncture-massage group being superior to the other 2 groups (P<0.05); and the physical signs in the 3 groups improved significantly (P<0.01), with no significant differences among the 3 groups (P>0.05); the time inducing the effect in the acupuncture-massage group was shorter than that in the other 2 groups (P<0.05 or P<0.01); and no adverse effects were found in the 3 groups. CONCLUSION: This acupuncture-massage comprehensive therapy has a definite therapeutic effect on CSA with safety, and it is of superiority as compared with the simple acupuncture or the simple massage.

Process formation of the renal glomerular podocyte: is there common molecular machinery for processes of podocytes and neurons?

The renal glomerular podocyte exhibits a highly arborized morphology. In comparison with the neuron, which is the best studied process-bearing cell, the podocyte major processes share many cell biological characteristics with neuronal dendrites. Both podocytes and neurons develop microtubule-based thick processes with branching morphology and both have thin actin-based projections (i.e. podocyte foot processes and dendritic spines). Formation of podocyte processes and neuronal dendrites depends on the assembly of microtubules. Because the assembly of microtubules is regulated by phosphorylation of microtubule-associated proteins, inhibition of protein phosphatases abolishes and inhibition of protein kinases promotes process formation. Podocytes and dendrites also share the machinery of intracellular traffic of membranous vesicles, as well as cytoskeletal elements, which is indispensable for the elongation of these processes. Furthermore, these two cell types share expression of various molecules working for signal transduction, transmembranous transport and intercellular contacts. Such common gene expression implies a similar transcriptional regulation in these cells. Concerning the formation of podocyte foot processes and dendritic branches, actin filaments are thought to play a central role in orchestrating the function of various molecules and the regulation of actin assembly is necessary to establish and maintain such sophisticated cellular architecture. The molecular mechanism of foot process formation seems to include Rho family small GTP-binding proteins, which are known to be responsible for the establishment of dendritic branching morphology.
