Canalesolide A, a Structurally Unique Polyhydroxy Macrolide from the Marine Cyanobacterium Okeania sp. with Potent Antitrypanosomal Activity.

The discovery of structurally novel natural products remains central to expanding biologically relevant chemical space, particularly within underexplored marine metabolite classes. Herein, we report the discovery and complete structural elucidation of canalesolide A, a new polyhydroxylated macrolide isolated from the marine cyanobacterium Okeania sp. The compound was identified through an integrated workflow combining phenotypic screening against Trypanosoma brucei and LC-MS/MS-based molecular networking, enabling rapid prioritization of bioactive fractions and dereplication of known metabolite families. Spectroscopic analysis revealed that canalesolide A belongs to the bastimolide-related class of macrolides but exhibits a distinct structural architecture. Its structure was established by integrating ultrahigh-resolution NMR spectroscopy, empirical configurational analysis of polyol systems, targeted model compound synthesis, and controlled chemical degradation and derivatization. This combined strategy resolved stereochemical motifs that were inaccessible by direct analysis of the intact macrolide alone, providing a transferable approach for assigning densely oxygenated marine macrolides. Genome mining identified the putative biosynthetic gene cluster and proposed biosynthetic pathway for a bastimolide-related macrolide. Canalesolide A displays potent, low nanomolar antitrypanosomal activity against human-infective subspecies of T. brucei with rapid elimination of parasites within 1 h at 1 μM. Although moderate mammalian cytotoxicity was observed, preliminary in vivo efficacy/toxicity studies in infected mice suggest a narrow therapeutic window highlighting the need for improved selectivity. This study expands the structural and biosynthetic diversity of polyhydroxylated macrolides and establishes a generalizable framework for resolving stereochemically complex natural products.

Five-coordinate Pd(II) orthometallated triarylphosphite complexes.

The reaction of the orthopalladated triarylphosphite complexes [{Pd(mu-Cl){kappa(2)-P,C-P(OC(6)H(2)-2,4-R(2))(OC(6)H(3)-2,4-R(2))}(2)] (R = H, (t)Bu) with bis(2-diphenylphosphinoethyl)phenylphosphine leads to a five-coordinate palladium(II) (R = H) and a mixture containing four-and five-coordinate species (R = (t)Bu). The crystal structure of the five-coordinate species [Pd{kappa(2)-P,C-(P(OC(6)H(4))(OC(6)H(5))(2)}{bis(2-diphenylphosphinoethyl)phenylphosphine}][SbF(6)] is presented. This complex reacts with hydrogen peroxide or [AuCl(tht)] to give four-coordinate complexes in which the displaced phosphine residue is either oxidised or coordinated to gold chloride; this demonstrates that the five-coordinate complexes are labile in solution. By contrast, the reactions of the dimeric precursors with 1,1,1-tris(diphenylphosphinomethyl)ethane give four-coordinate complexes in the solid state, although evidence is presented that the smaller phosphite-containing system is five-coordinate at room temperature or higher in solution.

Structural studies of the phase, aggregation and surface behaviour of 1-alkyl-3-methylimidazolium halide + water mixtures.

The surface, phase and aggregation behaviour of mixtures of 1-alkyl-3-methylimidazolium halide, [C(n)mim]X, where n is the alkyl chain length, with water has been explored using a variety of methods. Critical micelle concentrations (cmc) and micelle structures have been determined for aqueous [C(n)mim]Br solutions for n=2, 4, 6, 8, and 10. Small-angle neutron scattering (SANS) measurements reveal that for the n=8 and 10 systems, at concentrations just above the cmc, small near-spherical aggregates exist, which, after initial growth, possess core radii (aggregation numbers) at intermediate concentrations of 10.5+/-0.5 Angstrom (22+/-2) and 13.2+/-0.5 Angstrom (40+/-3), respectively, for n=8 and n=10. Towards higher concentrations, the aggregates appear to grow, with the aggregates in the [C(10)mim]Br system becoming increasingly elongated (prolate) with increasing concentration. No evident aggregates are formed in the systems with n=2 and 4. In the n=6 system, it appears that oblate aggregates with radius approximately 9 Angstrom form at the cmc and that the radius increases with increasing concentration. For longer alkyl chain lengths, at high concentrations lyotropic mesophases form in some systems. The mesophase region for the [C(8)mim]Cl system has been explored across the composition range using X-ray diffraction and (2)H NMR spectroscopy. Both techniques suggest that a major hexagonal phase with lattice parameter of 29.5+/-0.5 Angstrom coexists with a minor lamellar phase (23.5+/-0.3 Angstrom) or possibly a second hexagonal phase (27.1+/-0.4 Angstrom). The area per adsorbed molecule at the surface of [C(8)mim]Br solutions has been measured as a function of concentration using neutron reflectometry. A minimum in the area per molecule behaviour is coincident with a minimum identified in the surface tension isotherm occurring close to the cmc. The data suggest depletion of [C(8)mim]Br from the surface region occurs at concentrations immediately above the cmc.

BF3.OEt2 and TMSOTf: A synergistic combination of Lewis acids.

The combination of BF3.OEt2 and TMSOTf gives BF2OTf.OEt2, which is a more powerful Lewis acid than its components and especially effective in CH3CN solvent; the complex formed has been characterised by 1H, 19F, 11B and 31P (using Et3PO as an additive) NMR spectroscopy.

Piperazine additions to C60--a facile approach to fullerene substitution.

A range of fullerene monoadducts can be generated via the photochemical reaction of piperazine derivatives with C60. Addend functionality can also be efficiently incorporated by transformation of the hydroxyl-substituted adduct prepared in this fashion. Reaction yields and process simplicity compete with current standard procedures for fullerene mono-functionalisation.

Interfacial pH at an isolated silica-water surface.

Tethering a pH-sensitive dye to a silica surface enables the interfacial pH to be measured optically and is found to be 2 pH units lower than that of the bulk. The positive H+ ions are attracted by the surface potential and have an enhanced concentration consistent with the previously determined surface potential of order -120 mV, with respect to zero in the bulk. A stable near-surface charged layer once formed is not disrupted by the bulk pH.

Aggregation behavior of aqueous solutions of ionic liquids.

The aggregation behavior in aqueous solutions of three ionic liquids based on the 1-alkyl-3-methylimidazolium cation has been investigated by means of surface tension, conductivity, and small-angle neutron scattering (SANS) measurements. From analysis of the SANS data, models for the shapes and sizes of aggregates have been proposed: the short-chain 1-butyl-3-methylimidazolium tetrafluoroborate [C4mim] [BF4] system can be best modeled by treating it as a dispersion of polydisperse spherical aggregates that form above a critical aggregation concentration, whereas the 1-octyl-3-methylimidazolium iodide, [C8mim] [I], solutions can be modeled as a system of regularly sized near-spherical charged micelles that form above a critical micelle concentration. Solutions of 1-octyl-3-methylimidazolium chloride, [C8mim]-[Cl], display weak long-range ordering of possibly disklike particles culminating in the formation of structures with distinct long-range order at higher concentrations.
