Baking stability of acesulfame K.

The stability of acesulfame K during baking was investigated at different baking temperatures and baking times. The contents of acesulfame K in baked and unbaked doughs were determined by HPLC. The recovery rate of acesulfame K was independent of the baking conditions chosen and correlated with the recovery rate of acesulfame K in the unbaked doughs. As the stability of acesulfame K cannot only be affected by baking temperature and baking time but also by pH value and moisture content of the baked goods, additional stability investigations were performed with acidic fillings and apple pie. Even under these extreme baking conditions no decomposition of acesulfame K could be detected.

Evaluation of the genotoxic potential of sorbic acid and potassium sorbate.

The genotoxic potential of sorbic acid and potassium sorbate was investigated in vivo and in vitro. Oral administration of sorbic acid (up to 5000 mg/kg body weight) did not induce sister chromatid exchanges or the formation of micronuclei in bone marrow cells of mice. Intraperitoneal treatment of rats with 400-1200 mg potassium sorbate/kg body weight did not alter the elution profile of DNA from isolated liver cells in the in vivo alkaline elution assay. Sorbic acid did not induce DNA repair in cultured human A549 cells in the unscheduled DNA synthesis (UDS) assay. In vitro incubation of the cells with 1-1000 micrograms potassium sorbate/ml, in the absence or presence of rat liver homogenate, did not result in the formation of DNA single-strand breaks in the alkaline elution assay. These results demonstrate that sorbic acid and its potassium salt are not genotoxic in vivo or in vitro. In contrast to sorbic acid and potassium sorbate, sodium sorbate is very sensitive to oxidative degradation; the main oxidation product was identified to be 4,5-oxohexenoate, which was mutagenic in the Ames test.

Structure-activity relationship of anthracyclines in vitro.

The cytotoxic activities of several natural and semisynthetic anthracyclines against L1210 leukemia and two human colon tumor cells (Colon 4, HT 29) in vitro were examined after short (1 h) and long (7 days) incubation times and correlated with the water/octanol partition coefficients and the DNA-binding affinity of the compounds. Analysis of equation in which cytotoxicity against L1210 (1-h incubation) was parabolically related to the partition coefficient revealed an almost exclusive correlation (r = 0.80) between the cytotoxicity and the parameters, and this correlation was only slightly improved by addition of DNA-binding affinity (r = 0.85). On the other hand, cytotoxic activities displayed after continuous incubation were partially related to both partition coefficients (parabolic dependence) and DNA-binding affinities (linear dependence). In this case the correlation between the activity and partition coefficient (r = 0.67) was significantly improved by addition of DNA-binding affinity (r = 0.90). Similar results were also obtained for human colon tumor cells although the corresponding correlation coefficients were generally of lower value, indicating that cytotoxic activity of anthracyclines against these primary resistant cells may be influenced by additional factors not yet determined.

Indication and duration of the prednisolone therapy of chronic pulmonary sarcoidosis--a prognostic-therapeutic screening as a result of a controlled trial.

Report on a controlled therapeutic trial with prednisolone of chronic pulmonary sarcoidosis. The patients comprised three groups differing from the dose of prednisolone and the duration of the treatment. 194 clinically and histologically proven cases were completely treated. Two years after ending the treatment the relapse rates were about 20 to 30% in each group. This corresponds well with the rate of spontaneous regression. These results are an indicator that in most cases the underlying pathogenic process of immune deficiency is calming down during treatment and this leads to a possible change in therapeutic indication:--treating all patients for only one year, --taking into account that a relapse will occur in about one third of all cases,--avoiding supertreatment of the other two thirds with a not harmless and costly drug.

[The prognosis of intrathoracic sarcoidosis on the basis of the clinical picture and the histological feature in the mediastinal lymph nodes (author's transl)].

The clinical course of (pulmonary) sarcoidosis is difficultly foreseeable with regard to the fate of a given patient. There are some clinical features to get a probably good or probably poor prognosis, but there are only a few presumptions about the correlation between the histologic patterns with the clinical course. Lymph nodes were removed by mediastinoscopy. The authors have the intention with the aid of the histologic picture in these lymph nodes to examine, if 1. micro-cellular reactions and pure epithelioid-cell granulomas may have a better prognosis than 2. central necrosis, hyalinization, and fibrosis, whereas 3. the number of giant cells seems to be without prognostic significance.

Prognostic significance of lymphopenia in pulmonary sarcoidosis.

Since BOTTGER (1966, 1969) AND HOFFBRAND (1968) had described the absolute lymphopenia in pulmonary sarcoidosis as a sign of immune deficiency there were several authors who tried to elucidate different problems of sarcoidosis (RACOVEANU et al. 1970, STOJAN et al. 1971, OSODA et al. 1972). One of these problems is the effort to correlate absolute lymphopenia to prognostic parameters. Three controlled therapeutic trials were performed and compared: 59 cases with clinically and histologically proven sarcoidosis with spontaneous regression and 180 cases with different regimens of prednisolone: --a steady increase of the absolute number of lymphocytes in cases without and with treatment is a prognostically good sign, --no increase or even decrease is a poor sign, -- a very low and unchanged number is worst. This seems to confirm--among other things--that course and prognosis of sarcoidosis really may not be affected by time-limited corticotherapy.

[Prognostic evaluation of the absolute lymphopenia during prednisolone therapy of chronic pulmonary sarcoidosis (author's transl)].

From a controlled prednisolone therapy trial 130 patients with chronic pulmonary sarcoidosis who had already finished their drug-therapy were evaluated. The relapse rate was 28.5% within the following two years. It is obvious, that the long-time prognosis of cases with very low absolute numbers of lymphocytes in the peripheral blood (1000 and less/mul) is significantly poorer than in all other cases.
