Myeloid leukemia 239Pu-treated mice.

Incidence of myeloid leukemia was studied in 79 mice injected i.v. with 180 kBq 239Pu/kg. Plutonium-treated mice were compared with 70 controls of the same origin and sex (random-bred ICR-SPF female mice). The animals were killed moribund and the disease was diagnosed on the basis of histological and cytologic examination of bone marrow, spleen, liver, peripheral blood, and other tissues. It has been found that 22 plutonium-treated mice (27.8%) and 17 controls (24.3%) were leukemic. The mean survival of diseased animals was 459 +/- 19 days in the tested group and 559 +/- 24 days in controls. It means that the occurrence of myeloid leukemia was not very different in irradiated and control mice, but under plutonium contamination the disease appeared significantly earlier and its incidence was shifted to the younger age.

Electrophoretic mobility of mouse T-cell hybrids.

The hybrid cell line BH2 was derived by fusion between an AKR thymoma BW5147 (HGPRT-) and C57BL thymoma EL-4R (TK-). The hybrid cells showed a near-tetraploid modal number of chromosomes, in contrast to the near-diploid stem-lines of both parental cell populations; most of the BH2 hybrid cells acquired marker chromosomes from both parental cell lines. Inoculation of the parental and hybrid cells into C57BL and AKR mice revealed that the possible admixture of revertant parental cells in the hybrid cell population was less than 10(-4). Anodic electrophoretic mobilities (AEM) of the mouse thymoma lines and their hybrids were compared with each other and with normal mouse lymphoid cells. The AEM of the parental and hybrid T-cell lines was slower than that of mouse T LNC and comparable with AEM of some thymocyte subsets. The mean AEM of parental and hybrid cell lines was 0.69 micrometers/sec/V/cm for BW5147 cells, 0.96 for EL-4R cells and 0.83 for BH2 cells, the mean AEM of the hybrid cell population being identical with the mean of the parental AEM values. The mean AEM was found to be a relatively stable characteristic of each cell line.

T-Cell hydrids: I. Selection of thymidine kinase deficient subline EL-4R from EL-4 cell population.

Thymidine kinase deficient (TK-) cell line EL-4R suitable for production of T-cell hybrids has been selected from the EL-4 cell population by treatment with 5-bromodeoxyuridine. Comparison of the EL-4 and EL-4R cell lines revealed differences in chromosome constitution and electrophoretic mobility of both cell populations. Both, EL-4 and EL-4R cells had a modal number of 39 chromosomes; the percentage of cells with modal number of chromosomes and the percentage of biarmed chromosomes in the EL-4R line was higher than that of the EL-4 line. Anodic electrophoretic mobility of EL-4R cell population was faster than that of EL-4 cells, respective mean values being 0.96 and 0.91 X 10(-4) cm2 s-1 V-1.

Electrophoretic mobility profiles of mouse leukemias: I. Electrophoresis of normal mouse thymus and lymph node cell populations.

Anodic electrophoretic mobility of thymocytes and lymph node cells from random-bred ICR Swiss mice was examined. Three cell populations were identified which differed in their surface charge as reflected in their distinct mean AEM and in their time of appearance during the course of life. The first electrophoretically slow-moving population with a mean AEM of 0.97 microns sec-1 V-1 cm was detected in the early postnatal period. In the first month after birth, the first population was replaced by a second, slower-moving population with a mean AEM of 0.83 microns sec-1 V-1 cm. In the second month after birth, a third, fast-moving population with a mean AEM value of 1.24 microns sec-1 V-1 cm appeared in addition to the second cell population. The third population constituted only a minority of thymus cells and continued to be present, as well as the second cell population, up to two years of age. Lymph node cells of ICR mice showed a typical bimodal electrophoretic distribution; fast LNC displayed a mean AEM of 1.18 microns sec-1 V-1 cm whereas slow LNC that of 0.76 microns sec-1 V-1 cm. The percentage of cells in the fast- and slow-moving LNC subpopulations was similar to the expected percentage of T and B LNC. Fractionation of the ICR LNC on nylon wool columns revealed that the majority of the fast-moving LNC corresponded to the nonadherent Thy 1.2+ lymphocytes and the majority of the slow-moving LNC to the nylon wool-adherent sIg+ lymphocytes.

Cell-surface adhesiveness of mouse leukemias.

Cell-surface adhesiveness of forty-one primary mouse leukemias was quantitatively examined by the Latex (polystyrene) particle adherence (LPA) assay. Mean surface adhesiveness of leukemic cell populations was found to be substantially lower than that of normal mouse thymus, lymph node and spleen cell populations. No significant differences were found among mean LPA-positivity of sixteen thymic lymphomas, sixteen reticulum cell sarcomas and six myelogenous leukemias examined; the LPA-positivity of two nonthymic lymphomas and one undifferentiated leukemia was higher and comparable with that of normal mouse thymus cells. Comparison of the LPA-positivity of normal thymus cells and thymic lymphomas indicated that the malignant conversion of thymus cells was either accompanied or followed by a decreased cell-surface adhesiveness.

Bone sarcomas in 239Pu-treated mice.

Female ICR mice were injected with a monomeric form of 239Pu (180.1 kBq/kg) at the age of 10 weeks and during the long-term contamination the incidence of bone sarcomas was studied. Average survival of 100 control mice was 545 +/- 14 days, the 100 plutonium-given mice survived 420 +/- 10 d and 50 sarcoma-bearing animals 456 +/- 11 days. Most of the 61 induced sarcomas were located in the spine (70%) and in trabecular parts of the other bones. Among these 61 tumors there were 55 osteoblastic osteosarcomas, 4 immature undifferentiated sarcomas and 2 angiosarcomas. In 6 tumor-bearing mice metastases were observed in lungs, liver, intracranial cavity and in mediastinal lymph nodes. Among control mice only one osteogenic mandibular sarcoma was verified. The osteosarcoma incidence was evaluated with respect to the retained activity and cumulative average skeletal dose. The results were discussed and compared with data published by others.

Terminal hematological changes in mice bearing osteosarcomas induced by plutonium-239.

Terminal hematological changes were studied in female ICR mice injected intravenously with 181.1 kBq/kg of monomeric 239Pu. Cellularity and differential cell counts in femoral bone marrow and spleen, as well as peripheral blood counts were determined. The results were evaluated with respect to the quantitative hematological changes and to the diagnosis of proliferative disorders. As far as the nonstochastic response is concerned, evidently decreased cell counts, without abnormalities in myelogram, were observed in the femoral bone marrow of 239Pu-treated mice and only a mild leukopenia was present in peripheral blood. No significant difference was observed between the hematological response of tumor-bearing mice and the response of mice free of osteosarcomas.

3H-TDR labelling of osteoprogenitor cells after 226RA incorporation in mice.

The time course of 3H-TDR labelling index of osteoprogenitor cells and the doses in endosteum of lumbar vertebra and distal femoral epiphysis were autoradiographically determined in young female mice after single injections of 19.7 and 55.9 micronCi/kg body weight of 226Ra. The selected bone areas were examined in animals killed 2 hours to 28 days after the injection of nuclide. It was ascertained that changes in the relative labelling index are depending on the absorbed doses of alpha radiation. The possible relevance of these experimental findings for the explanation of the osteosarcoma induction and localization is discussed.

Activation and phenotyping of LAK generated by exposure to product of cells transformed by interleukin 2 cDNA.

It has been previously found that local administration of X63-m-IL-2 cells transformed by interleukin 2 (IL-2) cDNA and constitutively producing large quantities of IL-2 mediated regressions of murine plasmacytomas and 3-methyl-cholanthrene-induced sarcomas transplanted in syngeneic mice. Here we show that killer cells generated by cultivation of spleen cells in supernatants from X63-m-IL-2 cultures (LAK) or by co-cultivation of murine splenocytes with X63-m-IL-2 cells were cytolytic for natural killer (NK)-sensitive as well as NK-resistant target cells, including the IL-2-producing X63-m-IL-2 cells. Spleen cells cultured in X63-m-IL-2 supernatants or co-cultivated with X63-m-IL-2 cells yielded predominantly Thy 1.2+, CD3+, LFA-1+ lymphocytes. The in vitro results suggest that the LAK cells generated due to the IL-2 production by genetically engineered cells probably help to terminate the treatment by killing the IL-2 producers.

Gene therapy of cancer: use of IL-2 gene transfer and kinetics of local T and NK cell subsets.

Experiments were designed to compare the efficacy of recombinant IL-2 immunotherapy and IL-2 gene therapy of i.p. growing murine plasmacytoma X63-Ag8.653. The kinetics of peritoneal exudate mononuclear cells were monitored during the progression and gene therapy of the plasmacytoma, using cytofluorometric analysis and monoclonal antibodies against T and NK cell subsets. It has been found that the percentage of mice protected against plasmacytoma transplants was higher in mice treated by transfer of genetically manipulated IL-2-producing plasmacytoma cells as compared to the mice repeatedly injected with recombinant IL-2. Intraperitoneal inoculation of the X63-Ag8.653 plasmacytoma led in most of the inoculated mice to an increased percentage of NK+, ASGM1+, Thy 1.2+, CD3+ and TCR alpha beta+ cells in the peritoneal fluid. The presence of macroscopically detectable i.p. tumours was accompanied by a higher increase in the percentage of NK+ and TCR gamma delta+ cells. Local IL-2 gene therapy of the plasmacytoma either prevented or diminished an increase in the percentage of CD3+, Thy 1.2+ and TCR alpha beta+ lymphocytes.
