Toxicity and bioaccumulation of cadmium in the colonial green alga Scenedesmus obliquus.

A laboratory investigation was conducted to study the extent and efficiency of cadmium bioaccumulation in Scenedesmus obliquus by subjecting this alga to varied sublethal Cd concentrations. The influence of cell population age on Cd bioaccumulation was also studied. Under the experimental conditions employed, growth was not significantly affected by Cd concentrations ranging from 0.01 ppm to 1.00 ppm. At concentrations above 1.00 ppm, however, growth was inhibited markedly. Increases in external Cd concentration caused an increase in total bioaccumulation over the entire range of concentrations, which did not significantly affect growth. Efficiency of Cd bioaccumulation was also concentration dependent, but maximum accumulation efficiency occurred in a medium with a Cd concentration lower than that medium in which maximum total bioaccumulation occurred. Age of the cell population influenced the extent of Cd bioaccumulation. Rapidly growing, young cultures accumulated less Cd than older cultures approaching stationary growth phase.

Determination of paraquat in sunflower seeds by reversed-phase high-performance liquid chromatography.

The herbicide paraquat was determined with extracts from 1-g samples of sunflower seeds. The liquid chromatography procedure utilized a microparticle (10 micron) C18 reversed-phase column and isocratic elution with 27% acetonitrile in water, 10 mM in the sodium salt of octanesulfonic acid. Eluted paraquat was detected at 254 and 280 nm and quantitated by paraquat internal standard peak height ratios. This procedure provided linear working curves over the concentration range of 0-20 microgram/g of paraquat. Recovery of paraquat varied from 89-101%, with an average recovery of 93%. Good agreement was obtained in the comparison of results of the described procedure with those from a well established UV procedure.

Determination of thallium in urine with Zeeman effect graphite furnace atomic absorption.

Thallium is determined in urine with a simple dilution as the only sample pretreatment. Zeeman effect background correction provides adequate accommodation for non-atomic absorption such that sufficient diluted urine can be introduced into the furnace to achieve a detection limit of 0.5 ng/mL. Precision and accuracy were evaluated with Environmental Protection Agency water samples and in vitro spiked urine pools; precision of 15% at the 6 ng/mL level was observed. Diluted urine was used in calibration, with only 4 to 5% difference in slope observed within run among randomly selected urine specimens.

Reference range data for assessing exposure to selected environmental toxicants.

We analyzed blood and urine specimens from 32 charter boat captains, anglers, and spouses from both groups, who reportedly ate fish from Lakes Michigan, Huron, or Erie, for selected environmental toxicants. The toxicants measured in serum were polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), coplanar polychlorinated biphenyls, other polychlorinated biphenyls (PCBs), and persistent pesticides. Nonpersistent pesticides and elements were measured in urine; and elements were measured in blood. Internal dose levels of these toxicants will be compared to reference range data that we have compiled. These reference range data will be used to ascertain the exposure status of individuals or groups within this study.
