Characterisation of a sperm coating auto-antigen reacting with antisperm antibodies of infertile males using monoclonal antibodies.

OBJECTIVE: In a previous study a number of sperm-specific antigens were identified which reacted with antisperm antibodies from both infertile and vasovasostomised males. To investigate the localisation and distribution of these antigens and their role in male fertility, monoclonal antibodies were raised against them; immunoblotting techniques were used to select only those antibodies which competed with human antisperm antibodies for these human auto-antigens. DESIGN: One antibody, NW21, reacted with an 18 kDa auto-antigen present on epididymal sperm but absent from testicular sperm. Immunohistochemical studies showed that the antigen is produced in small basal cells between the columnar epithelium of the corpus epididymis, passes up into the tubule and then coats sperm passing along the epididymis. Sperm stored in the cauda epididymis and ductus deferens stain strongly for this sperm coating glycoprotein. CONCLUSIONS: The localisation of this antigen supports the suggestion that auto-immune infertility may represent a response to epididymal rather than testicular sperm. Monoclonal antibodies raised to unique and immunologically accessible sperm coating proteins, produced in the epididymis rather than in the testis, would seem to present an excellent theoretical solution to male contraception.

Effect of natural estrogen on the serum level of follicle-stimulating hormone (FSH), estradiol and estrone in post-menopausal women and its effect on endometrium.

Serum FSH, estradiol and estrone levels were assayed in 35 post-menopausal women who received the three commonly available estrogen preparations Premarin, Progynova and Harmogen. The results were compared with the levels during treatment with Dixarit. Hormone levels did not change when Dixarit was used. The levels of estrone and estradiol increased significantly (p less than 0.01) when estrogen preparations were used. There was no abnormality in the endometrium at the end of the treatment period.

Determination with Hi-Gonavis of luteinizing hormone levels in urine compared with those in plasma.

Hi-Gonavis, an immunological luteinizing hormone/human chorionic gonadotrophin (LH/hCG) test kit, was used to measure the periovulatory LH excretion in the urine of 25 patients over two cycles. The LH plasma levels was simultaneously determined by radioimmunoassay. The lower detection limit of Hi-Gonavis is 12.5 i.u. of LH/l of urine. Urinary LH levels of 50 i.u./l were associated with plasma levels of at least 12 i.u./l. Such values were found usually on the day before the mid-cycle peak of LH. The pre-ovulatory increasing plasma LH levels correlated closely with the urinary values. The urinary LH level reflects that in plasma; the Hi-Gonavis method can be used to predict the occurrence of ovulation.
