Molecular relationship of an atypical Azospirillum strain 4T to other Azospirillum species.

DNA/DNA hybridization, plasmid content and partial 16S rDNA sequence were determined to confirm the relationship between two Azospirillum strains, 4B and 4T, isolated from the same rice rhizosphere. The partial 16S rDNA sequence was determined for 9 strains belonging to 5 Azospirillum species which included Azospirillum lipoferum strains 4B and 4T, and was compared to a set of ribosomal sequences from other bacteria of the alpha subdivision of the Proteobacteria. Four Azospirillum species sequences were found to form a coherent group, whereas A. halopraeferens was found to be more divergent. The five Azospirillum species were closely related to Magnetospirillum and to a lesser extent to Rhodospirillum, with which they share some morphological features. The partial 16S rDNA sequences of the two strains 4B and 4T were identical and confirmed the closeness of these two strains. Plasmid content and DNA/DNA hybridization data (S1 nuclease and delta Tm) grouped Azospirillum strains 4B and 4T with the other strains of species Azospirillum lipoferum (USA5a, 59b, BR17). Other biochemical and genetic characters confirmed the grouping of Azospirillum strains 4B and 4T together with A. lipoferum.

Isolation of Azospirillum lipoferum 4T Tn5 Mutants Affected in Melanization and Laccase Activity.

Azospirillum lipoferum 4T has original properties such as nonmotility, melanin synthesis, and laccase activity. Following random Tn5 mutagenesis in A. lipoferum 4T, we obtained 10 mutants which were affected in melanization and laccase activity. The class 1 mutants, with intermediate levels of laccase activity, showed some coloration; the class 2 mutants, which were completely negative for laccase activity, were also colorless. The Tn5 localization on the chromosome or on the cryptic 300-MDa plasmid of A. lipoferum 4T was proven by hybridization for all class 1 mutants or for most class 2 mutants, respectively.

[The abdomen in multiple-injured patients. Comparative study on 225 multiple-injured patients with and without abdominal lesions. Incidence of abdominal complications].

Of 225 patients with multiple injuries and a high lesional score, 100 presented traumatic abdominal lesions. Comparison of the two groups confirmed both the usefulness and validity of a traumatic index and the reliability of abdominal puncture-lavage. Abdominal complications were analyzed. Mortality in all multiple injury cases, with or without abdominal lesions, was identical in this series where emergency surgery was performed only for lesions affecting vital organs, the mean age of patients with abdominal lesion being significantly lower. An attitude to adopt when deciding whether surgery is necessary is proposed.

[Intraperitoneal bladder rupture (author's transl)].

On-duty radiologists should keep in mind some peculiar circumstances leading to intraperitoneale bladder rupture without fracture of pelvic bones. It is thus possible to identify the classical pattern of dog's ears topping the bladder, on the abdomen plain film, and the diagnosis may be confirmed by intravenous urography. An illustration is given by the reported case: a young man, after drinking too much, received a kick into the lap, followed by a severe pain and total urinary retention with a flat bladder.

[Cellulitis of the leg as the presentation of pandiaphysitis in the child (author's transl)].

The authors report 6 cases of acute osteomyelitis of the leg, presenting with a very severe subcutaneous cellulitis and progressing to sub-periosteal abscess, then pandiaphysitis with major bone destruction at the 15th day, despite antibiotics. However, all these children recovered without sequelae, apart from one case. Early diagnosis was difficult and treatment started after an excessive delay in the majority of cases. If it can be performed on an emergency basis, Technetium bone scan is useful in making the diagnosis. In the absence of the type of examination, any case of severe subcutaneous cellulitis in a child must be considered as a potential osteomyelitis and treated as such.

[Carbonic anhydrases of bovine erythrocytes: comparative study of the anhydrase and esterase activity of different forms].

Comparative study of esterase activities (p- and o-nitrophenylacetate) allowed to characterize three groups of bovine erythrocyte carbonic anhydrases:--the first one includes CI, CII (isozyme of CI) and CIr ("artificial" product of CI).--the second one includes native CIv1 and "artificial" CIv1, first conformational variants of CI,--finally CIv2, second "artificial" conformational variant of CI. Possible modifications of the enzyme site between the first and the other enzyme groups are discussed. Except CIv2 of lower activity, all the products have identical carbonic anhydrase activity. The catalytic constants Km ap and kcat ap for hydrolysis of p-nitrophenylacetate have been determined for all enzymes; this study confirms the lower activity of CIv2.

Development of a polymerase chain reaction assay for identification and detection of the fish pathogen Flavobacterium psychrophilum.

A PCR-based method was developed to identify and detect Flavobacterium psychrophilum, the causative agent of "cold-water disease" and "rainbow trout fry syndrome" in salmonid fish. Two oligonucleotide primers were designed by comparing the 16S rRNA sequence of all taxa in the genus Flavobacterium and of representative species in most related genera within rRNA superfamily V. Purified chromosomal DNAs from all these bacterial species, from 25 F. psychrophilum isolates and from several other fish-pathogenic bacteria were used to assess the specificity of the reaction. Amplification products were generated only with F. psychrophilum DNA. The detection level, equivalent to approximately 10 to 100 bacterial cells, was increased 10-fold by hybridization with a radioactive probe. Preliminary experiments demonstrated that this procedure can also be applied to samples of infected tissue. This PCR assay is therefore a rapid, specific, and sensitive alternative to conventional plate culture methods for the identification and detection of F. psychrophilum.
