Behavioural and body temperature effects of meningococcal lipopolysaccharide after intraventricular injection in adult fowls Gallus domesticus.

The effects of the meningococcal lipopolysaccharide (LPS), given into the II cerebral ventricle of adult fowls, were studied on behaviour and body temperature. Immediately after the administration a marked and dose- dependent behavioural sedation or sleep lasting over 2 hours or more according to the dose was observed. The body temperature effects of LPS were biphasic, i.e. and immediate initial phase of hypothermia lasting about 100 min, followed by a longer-lasting fever response. A pretreatment with phentolamine, an antagonist at alpha-adrenoceptors prevented the hypothermic phase following the endotoxin administration. In conclusion, present experiments show that in comparison to 0-somatic antigen of Shigella dysenteriae, meningococcal endotoxin possesses more marked behavioural effects and a different profile in body temperature effects.

Legionellosis in an infant: first case in Italy.

Among several children admitted in Hospital with febrile acute pneumonia, it has been found - by indirect immunofluorescence assay a significant seroconversion for Legionella pneumophila serogroup 1, compatible with a recent infection, in an 18-month-old boy. The clinical course of the disease was favourable, and a prompt recovery as observed after administration of an antibiotic (cefuroxime) stable to the bacterial beta-lactamase.

Variability of antibody-specific induction and regulation of plaque-forming cells in cultures of normal human peripheral blood lymphocytes.

PFC have been detected in normal human PBL cultures, immunized with sheep red blood cells (SRBC) and supplemented with Concanavalin-A (Con-A) on day 0 (suppressive effect), and on day 2 (helper effect). Five PBL cultures responded punctually on established days, while four cultures failed to reproduce an enhancing effect when Con-A was added on day 2. In one case only no suppression was observed following addition of Con-A on day 0. Furthermore, it should be taken into consideration the dose of Con-A employed that can give suppressive, enhancing and no effects irrespective of the day of its addition in culture. On the basis of these considerations, it has been concluded that such a method is not suitable for clinical purposes, for instance in patients with immunological disorders, since the individual response to Con-A is extremely variable.

Bacterial endotoxin-immunocompetent cells interaction: newer aspects.

Bacterial endotoxin or LPS are constituents of the outer membrane of the cell wall of Gram-negative bacteria. When they invade human or mammalian organisms, they induce many biological effects. Lipid A represents the toxic part of the molecule. In the present communication, the AA. review some of the actions of LPS on immunocompetent cells, with particular reference to mitogenicity, adjuvanticity and tumor regression. Newer aspects of LPS immunobiology are emerging from the collected data. Of particular interest seems to be the relationship among T cells, Macrophages and B cells in LPS-induced adjuvanticity. These cells play a primary role in the potentiation of antibody production and their effects are mostly mediated via LK or MK. About possible mechanisms of tumor regression LPS-dependent, TNF and TNS features are illustrated. Because of the great extent of the present subject, the AA. are not able to include in this context some novel effects of endotoxin on immunocompetent cells as PGs release, SAA protein induction and colony forming activity.

[Intensive treatment of septic shock].

In order to identify a septic shock syndrome, the Limulus test (positive results in 10 cases out of 12) is preferable to hemoculture (positive in 9 cases out of 22), as a valid tool for a precocious diagnosis and to support both the clinical diagnosis and the precocious case finding of the septic conditions. According to the Authors, as far as intensive treatment is concerned, the simultaneous finding of the central venous pressure (CVP) and the lung capillary (LC) allows an accurate evaluation of the rate of cardiac impairment and the alterations to the distribution of blood volume in connection with treatment. The Authors have also reported the data related to the clinical course of illness and mortality rate (68%) in 21 cases of septic shock, as well as the hemodynamic parameters with the pertinent correlations (CVP-PAP-Pcp and Da-v) in 12 patients affected by septic shock from the very beginning and throughout the whole period of treatment.
