Desensitisation of human platelets against stimulation by thromboxane mimics--reduction in receptor capacity and loss of functional response.

Number and affinity of thromboxane A2/PGH2 receptors on gel-filtered human platelets was determined using the receptor antagonist 3H-SQ 29548 as a tracer compound. Preincubation of platelet rich plasma (PRP) with U 46619 reduced receptor number to 31% of control platelets without affecting the affinity (2 x 10(-9) mol/l). Pretreated platelets lost their reactivity to stimulation by U 46619 but not to thrombin within 3 h. It is concluded that preincubation of platelets leads to a homologeous platelet desensitisation and down regulation of thromboxane receptors.

[Pretherapeutic testing of sensitivity to cytostatic drugs in primary cultures of selected urologic tumors].

In 30 patients with malignant urologic tumours (scrotal tumours, renal tumours, tumours of the urinary bladder) an in vitro cultivation of the primary tumor was tried. According to the technique of Tannenberger and Bacigalupo 21 primary cell cultures could be applied. This corresponds to an accession rate of 70%. After formation of a cell lawn and addition of cytostatics of the arbitrarily selected medicaments vinblastin, bleomycin, cis-DDP, actinomycin D the reaction of the cells on the drugs was judged light-microscopically and electrophysiologically by measuring the transmembrane potential 24 hours after the application of medicaments. Thus apart from a usual oncobiogram which has as its fundament the morphology of the cells an electrooncobiogram could be established. The authors dealt with the methodology of the TMP-measurement after Redmann as a relatively new method for the pretherapeutic sensitisation test of tumours. The reactions of the cells to the cytostatics mentioned were so multifarious in the 3 groups of tumours that no conclusions of general validity could be drawn. These different modes of reaction are to be regarded as an expression of the biological individuality of human tumours. Apart from answering theoretical questionings the TMP-measurements might be used in the pretherapeutic sensitisation test of cytostatics in malignant renal tumours.

A method for radioimmunological assay of glisoxepide (author's transl).

A radioimmunoassay for the antidiabetic drug 3-(4-[beta-(5-methyl-isoxazol-3-carboxyamido)-ethyl]-benzenesulfonyl)-ureido-piperidine-4-carboxic acid (glisoxepide, I, Pro-Diaban) is described. Antisera were obtained by immunizing rabbits with a piperidine carboxylic acid analogue of glisoxepide coupled to bovine serum albumine. The carboxylic acid analogue coupled to [3H]-1-tyrosine methyl ester was used as the tracer. Antisera with high sensitivity were obtained. With some glisoxepide analogues the specificity of the antisera was investigated. For good binding the isoxazole and the benzene ring as well as the appropriate distance between both rings are essential. The assay can be carried out without extraction of the compound from the serum. Dextran coated charcoal is used for separation of free and bound glisoxepide.

Polyclonal hyperviscosity syndrome.

The clinical course of two patients who presented with polyclonal hyperviscosity syndrome is described. Polymerizing IgG rheumatoid factors were isolated from the serum of both patients. The presence of these polymers with their extraordinary rheologic properties was the principal determinant of the abnormal rheological properties of the plasma. Neither patient had a discrete rheumatic disease. In both, a lymphoproliferative syndrome was present with features suggestive of the "pseudolymphoma" of Sjögren's syndrome.
