Kinetics of cadmium accumulation and elimination in carp Cyprinus carpio tissues.

Carp (Cyprinus carpio) were tested for cadmium accumulation and elimination during and after a simulated pollution exposure. Fish were distributed in two 1000-l indoor concrete aquaria supplied with a continuous flow (8 l min(-1)) of well water. The cadmium concentration was maintained at 53 microg l(-1) in one aquarium and 443 microg l(-1) in the other aquarium for 127 days. The exposure phase was followed by a 43-day depuration period. The cadmium accumulation in liver, kidney and muscle was measured by means of ICP-MS. The data showed that cadmium exposure produces significant cadmium uptake in tissues. Cadmium concentrations increased sharply in kidney and liver, whereas the pollutant level in muscle was only significant after 106 days. After 127 days of Cd exposure (53 microg l(-1)), the cadmium concentration in kidney was 4-fold higher than in liver and 50-fold higher than in muscle for a toxic level of 53 microg l(-1). At a Cd of 443 microg l(-1), kidney cadmium content was 2-fold higher than in liver and 100-fold higher than in muscle. In kidney and liver, the toxic concentration increased as the concentration of pollutant in water increased. During the 43 depuration days, the loss of accumulated cadmium was rapid and immediate in muscle. Conversely, no loss of cadmium was observed in kidney and liver.

Value of enterobacterial repetitive intergenic consensus PCR for study of Pasteurella multocida strains isolated from mouths of dogs.

Fifty-six Pasteurella multocida strains (40 P. multocida subsp. septica and 16 P. multocida subsp. multocida strains) isolated from the mouths of 56 dogs among the 134 living in a French canine military training center (132e Groupe Cynophile de l'Armée de Terre, Suippes, France) were studied by use of enterobacterial repetitive intergenic consensus-PCR (ERIC-PCR) and restriction fragment length polymorphism (RFLP) techniques. Both techniques showed genomic heterogeneity of the strains studied. However, RFLP was more discriminatory than ERIC-PCR for differentiating P. multocida strains. All but three pairs of strains were discriminated by RFLP, suggesting a limited circulation of strains between these dogs living in proximity. Although ERIC-PCR is easier and faster to perform, it cannot be recommended for epidemiological studies of P. multocida strains.

Lactate metabolism and glucose turnover in the subterranean crustacean niphargus virei during post-hypoxic recovery

Glucose and lactate metabolism were studied in a hypoxia-resistant subterranean crustacean, Niphargus virei, using an injection of l-[U-14C]lactate and tracer d-[6-3H]glucose either in normoxic conditions or after a 24 h exposure to severe hypoxic. Post-hypoxic animals (H animals) were compared with two treatment groups of normoxic animals. In the first normoxic group (NLL animals), animals were simultaneously injected with labelled and unlabelled lactate to obtain a lactate load similar to that of H animals. In the second normoxic group (N, control animals), animals were only injected with labelled lactate. During a 24 h recovery period, the incorporation of 14C and 3H into glycogen, lactate, glucose, amino acids, lipids and CO2 was measured. During recovery, glucose turnover rate was enhanced in H and depressed in NLL compared with N animals. However, when energy expenditure was taken into account, the changes were due only to a reduction of glucose turnover rate by lactate load. It was concluded that gluconeogenesis was not the main source of glyconeogenesis. Equivalent lactate loading in NLL and H animals resulted in an equivalent enhancement (fivefold) of lactate utilization in both groups when energy expenditure was taken into account. Lactate label incorporation appeared later in glycogen than in glucose, but remained high 24 h after the injection. Since glucose is mainly an extracellular metabolite, this observation may be consistent with the hypothesis of two distinct sites for glycogen restoration in hypogean crustaceans: a gluconeogenic organ (a liver equivalent) and a glyconeogenic organ (a muscle equivalent). The oxidative pathways of glucose and lactate were depressed in post-hypoxic N. virei and to a lesser extent in the NLL group. Since there is no evidence of marked protein utilization, it is postulated that, during recovery, repayment of the O2 debt relies on an increase in lipid utilization. During recovery from severe hypoxia or after a lactate load, the subterranean N. virei appeared to implement a strategy of lactate removal quite different from that observed in epigean crustaceans, favouring lactate-supported gluco- and glyconeogenesis and rapid glycogen replenishment instead of rapid lactate removal via oxidative pathways.

Cadmium bioaccumulation in carp (Cyprinus carpio) tissues during long-term high exposure: analysis by inductively coupled plasma-mass spectrometry.

This work was an attempt to investigate cadmium bioaccumulation in the carp (Cyprinus carpio) during simulated pollution. The fish, weighing 100 g, were kept for 140 days in a 1000-liter indoor concrete tank supplied with a continuous flow (8 liters min-1) of unchlorinated, aerated, and filtered well water, the cadmium concentration of which was maintained at 450 microg liter-1. Such a high cadmium concentration was chosen to achieve metal saturation of the fish organs. Carp were fed during exposure. Cadmium accumulation in liver, kidney, and muscle was measured by inductively coupled plasma-mass spectrometry (ICP-MS), which is one of the most sensitive analytical techniques. The reported data indicate that cadmium exposure results in significant cadmium uptake, but the pattern of this uptake varies with the organ. For kidney and liver, cadmium concentration increased rapidly to the saturation level, probably because of the limited ability to store the cadmium as exposure persisted. A positive correlation between the increase in metallothioneins in tissues and the increase in metal tolerance in fish has been suggested. The cadmium concentration increase in muscle was significant only after 3 months, and, then, increased as an exponential function of the exposure time. When the storage capacity limits of the liver and kidney are reached, cadmium accumulation in muscle is stimulated. In this experiment, the high contamination levels reached suggest that such muscle would be unsuitable for human consumption. After 140 days of exposure, the cadmium concentrations in muscle, liver, and kidney were respectively 9 +/- 1, 91 +/- 7, and 250 +/- 16 mg kg-1 dry wt.

Good immunogenicity of GBM strain inactivated hepatitis A vaccine in healthy male adults.

A formalin-inactivated aluminium hydroxide adsorbed hepatitis A vaccine was evaluated in a dose-response study on 195 healthy male adults (age range: 18-31 years) in two French hospitals (Lyon, Rouen). Four doses (20, 40, 80, 160 RIA antigen units) were administered intramuscularly (i.m.) in two injections over a 6-month period. At the time of the first vaccine injection, 32 subjects (16.4%) were found positive (> 20 mIU ml-1) for HAV antibody (total Ig RIA HAVAB assay, Abbott Laboratories) and were excluded from the analysis of immunogenicity criteria. Fourteen days after the first vaccine injection, 78.1% (95% confidence interval (CI): 62-90) of seronegative subjects who received the 160 RIA antigen unit dose seroconverted with a geometric mean titre (GMT) of 43 mIU ml-1 (95% CI: 33-56). Seroconversion was 100% (95% CI: 91-100) at 1 month with a GMT of 95 mIU ml-1 (95% CI: 79-112). Statistical analysis revealed a significant dose-related effect (p < 0.0001) on GMT by multivariate regression analysis of the results after the first injection. Biological safety was evaluated and alanine aminotransferase and aspartate aminotransferase levels were similar prior to and 14 days after the first injection in the four groups. Reactions after injection were similar in the four dosage groups: 6.2% of subjects reported immediate reactions after first vaccination (feeling sick, spontaneous pain, headache), 8.9% reported local reactions at the site of injection (spontaneous pain, haematoma, local adenopathy) and 13.5% reported general reactions ('flu-like' syndrome, gastrointestinal tract disorders, fatigue, headache).(ABSTRACT TRUNCATED AT 250 WORDS)

Exposure to enteroviruses and hepatitis A virus among divers in environmental waters in France, first biological and serological survey of a controlled cohort.

An epidemiological study of hepatitis A and enteroviruses was conducted in a military diving training school, by evaluating the viral contamination of water using an ultrafiltration concentration technique, and assessing seroconversion and the presence of virus in stool specimens obtained from 109 divers and 48 controls. Three of 29 water specimens were positive for enterovirus by cell culture and 9 by molecular hybridization. There was little or no risk of virus infection during the training course (49 h exposure) because there was no significant difference between divers and controls for both viral isolation and seroconversion. However, a higher percentage of coxsackievirus B4 and B5 seropositive divers suggests that these were more exposed during previous water training. No hepatitis A virus (HAV) detection and no seroconversion to HAV was observed. The rate of HAV seropositive subjects was 17% in this 24.5-year-old population.

Transient adult T-cell leukemia/lymphoma picture during varicella infection in an HTLV-1 carrier.

HTLV-1 (human T-lymphotropic virus type 1) is associated with tropical spastic paraparesis, adult T-cell lymphoma (ATL), and also with opportunistic infections. The risk for developing ATL in HTLV-1 healthy carriers is low, between 1 and 4%. Nothing is known about the events promoting the evolution from the healthy carrier state to symptomatic ATL. We describe the case of a 44-year-old French Caribbean man with a chronic and recurrent strongyloidiasis in which the occurrence of a hemorrhagic and necrotic varicella led to the discovery of an infection by HTLV-1 and an acute form of ATL. All hematological data were normal before the onset of varicella. ATL completely disappeared at the same time as the varicella healed. This leads us to hypothesize that acute infections such as the reactivation of varicella-zoster may act as a promoting factor for the development of ATL in healthy HTLV-1 carriers.

Use of cRNA digoxigenin-labelled probes for detection of enteroviruses in humans and in the environment.

A dot blot hybridization test for enteroviruses is described using non-radioactive digoxigenin-labelled probes and a chemiluminescent detection. The use of a 5' non-coding riboprobe which detects all enteroviruses and a VP1 probe that detects the three serotypes of polioviruses allows the rapid detection of polioviruses and non-polio enteroviruses in human specimens or environmental water samples. The assay is strictly enterovirus specific and sensitive (800 fg RNA) and offers several advantages over conventional diagnosis or radioactive probes.

[Incidence of nosocomial infections in a military hospital].

Over a 2-year period, 479 cases of nosocomial infections were identified in our hospital by a surveillance method based on the bacteriological laboratory results. The monthly incidence rate ranged from 1.8 to 4 percent of all in-patients. With this method, a 2 percent background noise and a 4 percent alarm threshold could be defined. The most frequent nosocomial infections were urinary tract infections (77 percent), purulent skin infections (12 percent) and septicaemias (10 percent). Compared during 15 days with the most sensitive surveillance method of the National Nosocomial Infections Surveillance System, our method proved insufficient to detect nosocomial lung infections and superficial surgical wound infections. On the other hand, it was highly satisfactory to watch for urinary tract infections and septicaemias. Improvements that would not put a heavy burden on the work of clinical departments are suggested.

Inhibitory effect of anaesthesia with 2-phenoxyethanol as compared to MS222 on glucose release in isolated hepatocytes from rainbow trout (Salmo gairdneri).

1. Glucose production by freshly isolated hepatocytes from rainbow trout was studied after anaesthesia of the animals with 2-phenoxy ethanol (2PE) or tricaine methanesulphonate (MS222). 2. At the end of the procedure, hepatic contents of glycogen, glucose, lactate, ATP, ADP, AMP, were not significantly different between the two treatments. 3. Glucose production was considerably lower for 2PE than for MS222 anaesthetized trouts. This discrepancy results probably from an inhibition of glycogenolysis, suggesting that 2PE anaesthetized animals were less stressed than MS222 anaesthetized ones.

Determination of glucose turnover in sea bass Dicentrarchus labrax. Comparative aspects of glucose utilization.

1. Parameters of in vivo glucose utilization by sea bass (132 +/- 6 g, mean +/- SEM) acclimated at 15 degrees C in sea-water were measured after single injection of labelled glucose. 2. Glucose turnover rate (RG; mumol . min-1 . kg-1) was found to be 0.55-065 (2-3H glucose) and 0.34 +/- 0.42 (U14C glucose). 3. Glucose transit time was 443-449 min, glucose mass 233-261 mumol . kg-1, and glucose recycling 37%. 4. Oxygen consumption (MO2) amounted to 94 +/- 6.2 mumol . min-1 . kg-1. 5. The comparison with other fish species, mammals and birds, taking into account body size, temperature, diet, exercise, in poikilotherms and homeotherms leads to the calculation of a glucose turnover index (RGI = RG x 6 x 100 x MO2(-1)). 6. Value of this, generally lower in ectotherm teleosts (2-9), than in endotherms: mammals, birds and thunidae (22-60), confirms the minor quantitative importance of glucose in the metabolism of most fish.

Effect of acclimation temperature on Km values towards GDP-mannose of the microsomal dolichol phosphate mannosyltransferase activity of trout liver (Salmo gairdnerii).

Apparent Km value for GDP-mannose remained stable when trout were raised and when enzyme activity was assayed at temps corresponding to the natural biological temp (between 5 and 15 degrees C). The Km value increased when the temperature of the assay was higher than 21 degrees C. Marked increase of the Km value was observed when trout were acclimated at 21 degrees C, whatever the temperature of the assay between 5 and 30 degrees C.
