Evaluation of dogs with digit masses: 117 cases (1981-1991).

Medical records of 117 dogs with digit masses were reviewed. Of 124 digit masses, 76 (61%) were malignant neoplasms, 25 (20%) were benign neoplasms, and 23 (19%) were pyogranulomatous inflammation. Of 29 digits with radiographic evidence of bone lysis, 24 (83%) were affected by malignant masses, whereas only 5 of 29 (17%) digits with radiographic evidence of bone lysis were affected by benign or pyogranulomatous masses. Only 1 of 19 (5%) dogs with melanoma had radiographic evidence of lysis, but 20 of 25 (80%) dogs with squamous cell carcinoma had radiographic evidence of bone lysis. Thoracic radiographs of 95 dogs were available. Six of 19 (32%) dogs with melanoma had radiographic evidence of pulmonary metastasis at the time of diagnosis, whereas 3 of 24 (13%) dogs with squamous cell carcinoma had radiographic evidence of pulmonary metastasis at the time of diagnosis. Twenty-nine digit neoplasms were squamous cell carcinoma, and 19 of 29 (66%) arose from the subungual epithelium. Of 19 dogs with squamous cell carcinoma originating from the subungual epithelium, 18 (95%) survived for at least 1 year, whereas only 6 of 10 (60%) dogs with squamous cell carcinoma originating in other parts of the digit survived for at least 1 year. Furthermore, of 19 dogs with squamous cell carcinoma originating from the subungual epithelium, 14 (74%) survived at least 2 years, whereas only 4 of 9 (44%) dogs with squamous cell carcinoma originating in other parts of the digit survived for 2 years. Dogs with melanoma of the digits had a median survival time of 12 months, with 10 of 24 (42%) alive at 1 year and 3 of 23 (13%) alive at 2 years.(ABSTRACT TRUNCATED AT 250 WORDS)

In vitro and in vivo assays of isopropanol for mutagenicity.

To assess the mutagenic potential of isopropanol, an in vitro Chinese hamster ovary (CHO) cell/HGPRT gene mutation assay and a bone marrow micronucleus study in mice were conducted. In the CHO/HGPRT assay, concentration levels ranged from 0.5 to 5.0 mg/ml. No elevated mutant frequencies attributable to treatment were observed in the test under either activated or non-activated conditions. In the micronucleus assay, mice were injected intraperitoneally (IP) with either 350, 1,173, or 2,500 mg/kg of isopropanol at constant volumes of 10 ml/kg. No increased incidence of micronuclei was observed in bone marrow polychromatic erythrocytes (PCEs) harvested at 24, 48, or 72 hr post-dosing. In both assays, negative and positive control mutant frequencies were within historical control ranges. These results, in conjunction with previously published data, clearly demonstrate that isopropanol is not a mutagen.

Chronic morpholine exposure of rats.

The chronic toxicity and carcinogenic potential of morpholine were evaluated in 60 Sprague-Dawley rats/sex/group receiving morpholine at mean inhalation exposure concentrations of 0, 10, 50 and 150 ppm for 6 hr/day, 5 days/week, for 104 weeks. Survival, body weight gains, organ weights, hematology, and clinical chemistries were normal in exposed groups and comparable to those of the control animals. The incidences of palpable tissue masses and of histologically confirmed neoplasia were comparable among all groups, including the control groups, and were typical of the strain and age of the rats tested. In-life clinical examinations revealed increased incidences of irritation around the eyes and nares, chromadacryorrhea, and urine stains on the fur, predominantly in high-dose animals. Morpholine exposure was associated with corneal irritation seen by ophthalmoscopic examination and confirmed microscopically as keratitis limited to the highest exposure group. Irritation of the maxillary and nasoturbinates as indicated by infiltration of neutrophils, focal squamous metaplasia of the turbinate epithelium, and necrosis of the turbinate bone was observed in high-dose animals. Therefore, chronic exposure of rats to morpholine for 2 years at concentrations of 150 ppm or less revealed no carcinogenic potential or chronic systemic toxicity. Consistent with its known irritating properties, morpholine produced only local irritation, which was limited almost exclusively to high-dose animals.

Evaluation of Pluronic Polyol F127 as a vehicle for petroleum hydrocarbons in the Salmonella/microsomal assay.

Complex hydrocarbon mixtures have proven difficult to evaluate in in vitro mutagenicity assays owing to their insolubility in aqueous environments. Pluronic Polyol F127 (BASF Wyandotte, Parsippany, NJ), prepared as a 50% (w/w) solution in absolute ethanol, proved effective in emulsifying various petroleum hydrocarbon fractions. Its effectiveness in the Salmonella/microsomal assay was evaluated using model solutions each comprising a polycyclic aromatic hydrocarbon (PAH) dissolved in mineral oil. The PAHs used were benzo(a)pyrene, 3-methylcholanthrene, and 7,12-dimethylbenz(a)anthracene. Model solutions were evaluated neat and as emulsions with the Pluronic F127 solution or Tween 80. Similar levels of each PAH were prepared in dimethyl sulfoxide (DMSO) for comparison. Cytotoxicity and mutagenesis were evaluated in the preincubation technique using strain TA97. Little or no cytotoxicity or mutagenesis was evident for model solutions tested neat. However, emulsification of these PAH-laden mixtures with the Pluronic F127 solution yielded cytotoxic and mutagenic responses similar to, or greater than, those observed for PAHs delivered in DMSO. Model mixtures emulsified with Tween 80 were less active. Study results demonstrate that Pluronic F127, prepared as a 50% (w/w) solution in absolute ethanol, is an effective vehicle for evaluating the mutagenic potential of complex hydrocarbon mixtures containing PAHs in the Salmonella/microsomal assay. Since PAHs are a class of insoluble hydrocarbons, the results also suggest the potential usefulness of the Pluronic F127 solution to detect the mutagenicity of other insoluble hydrocarbons and hydrocarbon mixtures.

Ventricular arrhythmias in dogs undergoing splenectomy: a prospective study.

Fifty dogs undergoing splenectomy for splenic masses (n = 40), torsion of the splenic pedicle (n = 5), and immune-mediated disease (n = 5) were evaluated preoperatively and and postoperatively for ventricular arrhythmias and the relationship between ventricular arrhythmia splenic disease. The ability of 1-minute electrocardiograms recorded every 6 hours (ECGs/q6hr) to detect ventricular arrhythmia was compared with continuous 48-hour Holter monitoring. Based on continuous Holter monitoring, splenectomized dogs had a high incidence (22 of 50) of rapid ventricular tachycardia. The incidence of rapid ventricular tachycardia was significantly higher in dogs with ruptured splenic masses (16 of 23) than without rupture (1 of 17) (P < .001). When the results of ECG/q6hr were compared with the results of continuous Holter monitoring ECG/q6hr was normal in 29% (4 of 14) of dogs with rapid ventricular tachycardia at > 3,000 ventricular extrasystoles (VE)/hr; 50% (4 of 8) of dogs with rapid ventricular tachycardia at 1,000 to 3,000 VE/hr and 100% (6 of 6) of dogs with 10 to 300 VE/hr without rapid ventricular tachycardia. Although dogs undergoing splenectomy had a high incidence of ventricular arrhythmias, one-minute ECGs/q6h were unreliable for detection of ventricular arrhythmias even when high-frequency extrasystoles occurred.

Nocardiosis. A literature review with selected case reports in two dogs.

Two dogs with systemic nocardiosis are presented and the pathobiology, diagnosis, and treatment of nocardial infections are discussed. Both dogs had nonspecific respiratory signs and depression. The diagnosis was made by isolation of the organism only after surgical drainage was established and appropriate tissues were cultured. The response to surgical drainage and antimicrobial therapy was dramatic in both dogs, but one dog experienced a drug reaction to trimethoprim-potentiated sulfonamide. Although systemic nocardial infections traditionally have had a grave prognosis, through early diagnosis, surgical intervention, and the use of newer, safer and synergistically acting antimicrobials, the prognosis has improved. This article reviews current human and veterinary literature regarding the microbiology, pathogenesis, and treatment of nocardiosis and reports on the successful treatment of systemic nocardiosis in two dogs.

Complications associated with high-dose prednisolone sodium succinate therapy in dogs with neurological injury.

A retrospective study of 105 dogs with neurological injury was conducted to evaluate the type and prevalence of complications encountered when treatment consisted of a high-dose corticosteroid protocol. All dogs were treated with high-dose prednisolone sodium succinate (HDPSS) (30 mg/kg body weight, administered intravenously [i.v.] q 6 hrs for 36 hrs). Thirty-five (33.3%) cases developed complications including diarrhea (n = 11), melena (n = 11), vomiting (n = 6), hematochezia (n = 3), hematemesis (n = 1), anorexia (n = 1), or a combination (n = 2). Most complications resolved without additional treatments after termination of the HDPSS therapy. None of the complications were considered serious, and none prolonged hospitalization.
