Histidine supplementation for treatment of anaemia of uraemia.

The effects of supplemental histidine on the anaemia of 16 chronically uraemic patients and 26 patients undergoing maintenance dialysis were assessed. Patients were given either L-histidine 4 g/day or placebo for a mean of 17-5 weeks in a randomized double-blind controlled study. Haemoglobin and packed cell volume increased slightly and to a similar degree in the patients on dialysis receiving placebo and histidine, but this change was statistically significant only in those on placebo. Plasma histidine rose significantly in the histidine-treated patients undergoing dialysis and decreased in the uraemic patients receiving placebo. These results suggest that supplementary L-histidine in patients with uraemia or patients undergoing maintenance dialysis does not improve anaemia.

Immunochemical isolation and characterization of ovalbumin messenger ribonucleic acid.

Hen oviduct ovalbumin messenger RNA has been purified to apparent homogeneity and its physical and molecular properties have been examined. Purification was achieved through the use of indirect immunoprecipitation to isolate ovalbumin synthesizing polysomes and the use of poly(U)-Sepharose chromatography to separate quantitatively ovalbumin messenger RNA from ribosomal RNA. Ovalbumin mRNA was purified 90 to 100-fold over oviduct polysomal RNA as judged by both the rate of hybridization to a complementary DNA and by translation in a rabbit reticulocyte lysate protein-synthesizing system. Isolated ovalbumin mRNA migrates as a single sharp symmetrical peak on sucrose gradient sedimentation and polyacrylamide gel electrophoresis. The molecular weight of ovalbumin mRNA determined by sedimentation in denaturing dimethylsulfoxide gradients is 700,000 (equivalent to 2,180 nucleotides). The complexity of purified ovalbumin mRNA determined from the relative rate of hybridization to a complementary DNA is 2,280 nucleotides. Since ovalbumin synthesis requires only 1,161 nucleotides, ovalbumin mRNA appears to contain approximately 1,150 untranslated nucleotides. The average length of the polyadenylate sequence in ovalbumin mRNA is only 44 nucleotides and it does not account for significant fraction of the untranslated nucleotides.

A high-efficiency HeLa cell nuclear transcription extract.

A HeLa cell nuclear transcription extract that is approximately 20 times more efficient than standard HeLa cell transcription extracts was developed. Transcription of the strong adenovirus II major late promoter by this extract results in the synthesis of 1.5-4 molecules of product RNA per molecule of template, indicating that the extract is capable of multiple rounds of initiation. Standard HeLa cell nuclear extracts transcribe closed circular and linear adenovirus major late promoter templates with equal efficiency. In contrast, the new extract exhibits an increase of approximately twofold on transcription of a closed circular, as opposed to a linear, major late promoter template.
