[Reactive oxygen products and antioxidants in cardiovascular diseases].

Cardiovascular diseases account in this country for more than half the mortality. In the submitted review the authors discuss reactive oxygen products as one of the possible risk factors of the mentioned diseases. The second part of the paper deals with antioxidative enzymes and antioxidants in cardiovascular diseases as well as with possibilities of their use in prevention and improvement of adverse clinical conditions.

Sex-related and age-related differences in platelet function in vitro: influence of hematocrit.

Even minor changes in the sodium citrate concentration of citrated plasma from young probands due to diverse hematocrit values were found to affect platelet functional activity following in vitro stimulation. There were no differences between men and women in the platelet aggregation and TXB2 production of young probands with similar hematocrit values. Platelets from elderly persons virtually failed to respond to such changes in plasma sodium citrate. Mechanisms of increased platelet activity in the elderly are discussed. It is assumed that the age-related differences are caused by a more pronounced platelet response even to lower plasma calcium levels in citrated plasma.

[Functional status of thrombocytes in men and women in younger age groups].

Differences in the functional status of platelets in vitro were compared between young adult men and women. Platelets in women were found to have a higher susceptibility to ADP, epinephrine, arachidonic acid and to produce more TXB2. These changes appear to be due to the declined hematocrit and thus to lower concentrations of sodium citrate, a commonly used anticoagulant. Platelet activity was conversely related to plasma sodium citrate. Since calcium chloride was found to offset the inhibitory effect of sodium citrate, it was assumed that the sex-related differences observed in platelet activity were due to varied levels of ionized calcium. The platelet activity was at equal values of hematocrit equal in both sexes. (Tab. 3, Ref. 19.).

[Blood platelets, lipid peroxidation and antioxidants].

The paper reviews published data on the products of platelet lipid peroxidation, their role in blood coagulation as other physiological and pathological processes. The authors discuss the significance of anti-oxidants (selenium, glutathione peroxidase, reduced glutathione and vitamin E) in removing excessive hydroperoxides and thus in the control of platelet functional activity. Changes in platelet arachidonic acid metabolites and in antioxidants were found in a number of pathological conditions such as diabetes mellitus and cardiovascular diseases.

[Functional status of thrombocytes in men of various age groups].

The functional state of thrombocytes was compared in men of three age groups, i.e. below 46 years, within the range of 46 to 59 years, and above 59 years. Thrombocytes of old people were found to be more sensitive to such aggregation inductors as ADP, collagen, arachidonic acid, and adrenaline compared to thrombocytes of subjects from the younger age groups. A further finding was that blood platelets of old men produced significantly greater amount of ATP than recorded in young men and this may account for hyperactivity of thrombocytes in old age.

Age-related changes in human platelet function in vitro.

Platelet aggregation and ATP release were simultaneously measured in platelet-rich plasma samples obtained from humans of various age. In subjects aged over 59 years, an increase was found in platelet sensitivity to ADP and collagen as well as elevated aggregation amplitudes 5 min after induction with low concentrations of the aggregation agents. At 1 mumol/l ADP, old persons had a higher incidence of a secondary aggregation wave. Platelets from old subjects also released more ATP in response to collagen stimulation. Since no age-related changes are commonly found in the platelet count, the observed increase in platelet sensitivity to aggregation agents cannot be regarded as a mere compensatory reaction. An attempt is made to relate altered platelet function to peculiarities of lipid metabolism associated with ageing.
