[The effect of a nerve growth factor from different biological sources on the spontaneous maturation of mouse oocytes and on the parthenogenetic activation of pronucleus formation].

The paracrine role of the nerve growth factor (NGF) in oocyte maturation is analysed. It is shown that the addition of mouse beta-NGF to the serum-free culture medium in doses of 100, 10 and 1 ng/ml induces reinitiation of meiosis in mouse oocytes after gonadotrophic stimulation. It is identified that oocytes without cumulus cells demonstrate a 2-3 fold increase in the rate of parthenogenetic pronucleus formation in comparison with the control. Similar results are obtained using bovine beta-NGF. It is supposed that the oocyte surface has a receptors to beta-NGF.

[The effect of gamma irradiation on the structure and enzymatic activity of the nuclear membrane of the liver in pregnant rats and their embryos].

Morphological and biochemical investigations of pregnant rats and embryo liver cell nuclei after in vivo irradiation in the doses of 1 and 2 Gr revealed their high radiosensitivity at all stages of gestation and embryonal development. At damaging effect of radiation, we managed to observe sharp accumulation of products of lipid peroxide oxidation and suppression of the activities of such enzymes as cytochrome-c-oxidase, NAD.N-cytochrome-c-reductase, ATPase and RNAase in liver nuclei of pregnant rats and embryos. The changes of such a kind are shown to intensify with the increasing of irradiation doses. The most profound inhibition of the activities of these enzymes in liver nuclei of embryos irradiated in utero was observed during the period of organogenesis (the 13th day of the development) and in fetal period of embryogenesis (the 17th day of the development), as well as at the 13th and 17th day of gestation. The morphological data also demonstrate the high level of cell nucleus sensitivity to the action of radiation during gestation and embryogenesis.

[Computer analysis and comparison of neurotoxin structures from snake and the anemone Radianthus macrodactylus].

Evolutionary trees of proteins analysed by means of computer comparative methods were reconstructed and the level of the structure and functional relationship was revealed. The distance between the percent amino acid contents was used to classify a group of toxins from the sea anemone Radianthus macrodactylus, which proved to belong to long neurotoxins. The scores were calculated by the pattern recognition method. By the use of the distance values, ten phospholipases A2 were divided into two groups, containing representatives of the most closely related organisms.

[RNA synthesis and transport in the rat liver under the effects of pesticide cotoran (fluometuron)].

The experiments on the investigation of pesticide cotoran-effect on RNA synthesis and transport were carried out. Cotoran was shown to destroy considerably the processes of RNA biosynthesis in rat liver, that results in the decrease of RNA transport from nuclei into cytoplasm. By special experiments it was established that functional activity and the integrity of nuclear membrane (according to the alteration in the activity of nuclear membrane enzyme Mg2-dependent ATP-ase) was not destroyed.

[Effects of pesticide fluometuron (cotoran) on template synthesis of RNA].

The experiments on the investigation of pesticide fluometuron (cotoran) influence on nuclease sensitivity and template activity of rat liver chromatin were carried out. Cotoran was found to bind specifically with non-histone proteins of chromatin. It was shown that this pesticide considerably decreases template activity of chromatin and its sensitivity to the action of nucleases. It suggests, that certain conformation changes occur in chromatin upon the action of cotoran.

[Effects of peginol C-2000 on respiration and oxidative phosphorylation in liver mitochondria of rats poisoned with butylcaptax].

The data on the effect of synthetic antioxidant Paginol C-2000 on respiration and oxidative phosphorylation in liver mitochondria of rats primed with butylcaptax are given. It is shown that intoxication of organism with butylcaptax in the dose of 1/10 LD50 during 5 days resulted in suppression of respiration and oxidative phosphorylation in rat liver mitochondria. It is found, that administration of Raginol C-2000 into organisms of primed animals leads to the reduction of succinate pathway of oxidation, that, in its turn, results in the normalization of functioning level of ATP-synthesizing system of mitochondria.

[The translocation and specific binding of the pesticide fluometuron (cotoran) with the proteins of rat liver nuclei].

Intracellular distribution of labeled cotoran was studied. 3H-cotoran was shown to penetrate through the nuclear membrane to accumulate uneventfully in the intranuclear components. An insignificant amount of 3H-cotoran was associated with the nucleoplasm and the outer nuclear membrane. At the same time, essential radioactivity was observed in the proteins of the nuclear matrix (up to 30%) and in non-histone proteins of chromatin (up to 60%). Acception of 3H-cotoran on metaphase chromosomes of cultured cells as well as specificity of cotoran binding with non-histone proteins of chromatin in vivo and in vitro was studied by radioautography. It was revealed that cotoran was translocated into the interphase nuclei to be accepted by metaphase chromosomes of the HeLa line cells and fibroblasts in human embryo, and specifically, in receptor-like manner, bound to chromatin proteins.

[Isolation and characteristics of a nerve growth factor from the venom of the Central Asiatic viper Echis multisquamatus].

The nerve growth factor (NGF) was isolated from the Echis multisquamatus venom by ultrafiltration on PM-10 filter, chromatography on TSK-55 gel, ion-exchange chromatography on CM-cellulose and gel filtration on Sephadex G-75. The protein exhibited a marked nerve growth activity within the concentration range of 10-15 ng/ml in cultures of chicken embryo spinal ganglia. The molecular mass of NGF is equal to 33,000-37,000 Da according to Sephadex G-75 gel filtration data; however, according to SDS electrophoresis data its Mr is 13,000 Da. Isoelectrofocusing data suggest that the pI of the isolated factor lies in the region of 7.0-7.2; sugar content is 1-2%.

[Electron immunocytochemistry of lymphocyte surface immunoglobulins in the bursa of Fabricius in the pre- and early postnatal ontogeny of chickens].

The appearance and localization of surface immunoglobulins on B-lymphocytes of the bursa of Fabricius of developing chick embryos and chicks were studied by immuno-electron-cytochemical peroxidase-antiperoxidase (PAP) method. Ultrastructural characteristics of marked and unmarked lymphocytes of the bursa of Fabricius was shown. The increase in the density of reaction product on PAP-complex in the course of lymphocytes differentiation was established.

[Effect of thyroxine in vitro on the transmembrane potential of rat liver mitochondria].

It has been shown in in vitro experiments that a certain latent period after the addition of thyroxine (T4) and triiodothyronine (T3) was necessary for the manifestation of their effects on transmembrane potential (TMP) of the rat liver mitochondria. The duration of the lag-period decreased upon an increase in the concentrations of these hormones, and T4 at a dose of 2.10(-4) M produced a fall in TMP immediately after its addition. The rate of TMP fall was in proportion with the concentrations of thyroid hormones introduced into the cell, with T3 30-40% more effective than T4. It was established that the action of I2 resembled that of thyroid hormones, namely, a fall in TMP, mitochondrial swelling, activation of transhydrogenase Kl was ineffective. It is suggested that the appearance of the lag-period upon the action of thyroid hormones might be explained by the period of time necessary for the formation of the active iodine forms, as well as by the formation of fatty acids (donators of H+) by mitochondrial phospholipases. All these factors lead to TMP fall resulting in decreased formation of sufficient ATP quantities in mitochondria.

[Action of thyroxine on the cytochrome content in the brain and liver mitochondria during the postnatal development of rats].

The content of cytochromes c + c1, b and a in brain and liver mitochondria in 7-day-old rats reaches the level seen in adult animals. Administration of L-T4 in a dose of 0.7 micrograms/g rat bw for 4 days before sacrifice results in activation of cytochrome synthesis in both test organs within the first week of the suckling rats' life. On the 20th day of the postnatal period the effect of T4 is seen only in the liver while the brain tissue turns out indifferent to the thyroid hormone. Thus, T4 activates cytochrome biosynthesis in brain mitochondria during the first week of the rats' life, that leads to the acceleration of the functional activity and higher differentiation of the developing brain mitochondria.

[Effect of thyroxine on the activity of a series of mitochondrial enzymes during the process of development].

The activity of alpha-glycerophosphate dehydrogenase and succinate dehydrogenase suffers almost no changes during the chick embryogenesis, whereas the activity of cytochrome oxidase increases by 20--25%. The activity of these enzymes in the embryos exceeded that in the adult birds. The injection of thyroxine during embryogenesis did not affect the activity of the enzymes under study but in chickens stimulated the activity of mitochondrial enzymes. The increase of activity of alpha-glycerophosphate dehydrogenase, cytochrome oxidase and succinate dehydrogenase was also noted in young rats.

[Participation of thyroid hormones in the regulation of the genetic activity of normal and transformed human cells].

It is shown that 125J-thyroid hormones are localized on the structures of the interphasic nucleus and metaphasic chromosomes of cultured fibroblasts of 8--10-day human embryos. At the same time, the labelled thyroid hormones, though localizing in the interphasic nuclei of HeLa cells, are not accepted, unlike normal cells, by their metaphasic chromosomes. It is suggested that during transformation the acceptor ares of the HeLa cells genome lost their property to bind their own receptor complexes with thyroid hormones.
