Distribution of 125J-labeled leucopyrokinin (LPK) in rats.

The distribution in parts of the brain and internal organs of 125J-labeled leucopyrokinin (LPK), an insect myotropic peptide injected into the lateral brain ventricle was determined in rats. A high accumulation of this peptide in adrenals and in the hypothalamus and hippocampus of the brain was found. A lesser but significant LPK accumulation in other internal organs and parts of the brain was also observed. The significance of this effect for the biological activity of LPK in rats is discussed.

The effect of the leucopyrokinin analogue: [2-8]-leucopyrokinin on central opioid receptors in rats.

The antinociceptive effect of intracerebroventricular injections of [2-8]-leucopyrokinin (LPK), a truncated leucopyrokinin analogue, was determined in rats, by means of a tail immersion test. We found a significant antinociceptive effect of three i.c.v. doses of [2-8]-LPK: 1, 5 and 10 nmol. Pre-treating animals with naloxone hydrochloride (1 mg/kg i.p.) completely blocked the effect of two high doses of [2-8]-LPK. To determine the sub-types of opioid receptors involved in [2-8]-leucopyrokinin-induced analgesia we injected specific blockers of mu-, delta- and kappa-receptors namely, beta-funaltrexamine hydrochloride, naltrindole hydrochloride and nor-binaltorphimine dihydrochloride, respectively, prior to [2-8]-leucopyrokinin at equimolar doses. We conclude that the antinociceptive effect of [2-8]-leucopyrokinin is mediated mainly by central mu- and delta-opioid receptors.

Synthesis and biological evaluation of selected insect neuropeptide analogs modified by D- or L-phenylglycine derivatives.

Novel analogs, modified by L- or D- phenylglycine and p-substituted derivatives, of the neuromodulator proctolin (Arg-Tyr-Leu-Pro-Thr) and of the Trypsin Modulating Oostatic Factor from the gray flesh fly Neobellieria bullata (Neb-TMOF-Asn-Pro-Thr-Asn-Leu-His) were synthesized and checked for activity. Proctolin analogs were modified at position 2: Arg-Phg-Leu-Pro-Thr (I), Arg-D-Phg-Leu-Pro-Thr (II), Arg-Phg(p-OH)-Leu-Pro-Thr (III), Arg-D-Phg(p-OH)-Leu-Pro-Thr (IV), Arg-Phg(p-NO2)-Leu-Pro-Thr (V) Arg-D-Phg(p-NO2)-Leu-Pro-Thr (VI), Arg-Phg(p-NH2)-Leu-Pro-Thr (VII), Arg-D-Phg(p-NH2)-Leu-Pro-Thr (VIII), Arg-Phg(p-N,N-di-Me)-Leu-Pro-Thr (IX), Arg-D-Phg(pp-N,N-di-Me)-Leu-Pro-Thr (X) while analogs of Neb-TMOF underwent modifications at position 6: Asn-Pro-Thr-Asn-Leu-Phg(p-NO2) (XI), Asn-Pro-Thr-Asn-Leu-D-Phg(p-NO2) (XII), Asn-Pro-Thr-Asn-Leu-Phg(p-NH2) (XIII), Asn-Pro-Thr-Asn-Leu-D-Phg(p-NH2) (XIV), Asn-Pro-Thr-Asn-Leu-Phg(p-N,N-di-Me) (XV), Asn-Pro-Thr-Asn-Leu-D-Phg(p-N,N-di-Me) (XVI). Earlier studies on proctolin demonstrated that the presence of the -CH2- group between C-alpha and the phenyl ring at position 2 of the peptide chain is important for the myotropic activity. Based on these results, we replaced Tyr at position 2 by different phenylglycine derivatives, lacking the methylene group at the side chain. Myotropic activity of the proctolin analogs was assayed in vitro on the semi-isolated heart of the mealworm Tenebrio molitor and on the foregut of the locust Schistocerca gregaria. All analogs (I-X) were practically inactive. For Neb-TMOF, it was previously demonstrated that the exchange of His-6 by p-substituted Phe-derivatives, especially by Phe(p-NH2), an amino acid containing a basic function, results into analogs which inhibit trypsin biosynthesis in the gray fleshfly. For this reason these new Neb-TMOF analogs with L- or D-phenylglycine p-substituted derivatives at position 6, were developed and tested (in vivo) in the trypsin biosynthesis assay of the gray fleshfly N. bullata. Only analogs XV and XVI slightly inhibited trypsin biosynthesis in the midgut. Because more than 50% of the injected animals died and none of the surviving animals ate much of the liver meal, the lower trypsin level in the gut might be a indirect effect. Other peptides (XI-XIV) had no effect on the level of trypsin biosynthesis in the midgut.

Insect neuropeptide proctolin and its analogues. An overview of the present literature.

In the present paper, the literature describing synthetic, biological and conformational studies on the insect neuropeptide proctolin (H-Arg-Tyr-Leu-Pro-Thr-OH) and its analogues is summarized. The paper covers proctolin and its 80 analogues modified in positions 1-5, a cycloanalogue and analogues with a truncated or elongated peptide chain. These peptides were bioassayed by different methods, e.g. studies of myotropic activities in several insect species in vitro and behaviour in rats in vivo. Based on these data structure-activity relationships are discussed.

Selective activity of a proctolin analogue reveals the existence of two receptor subtypes.

1. The neuropeptide proctolin (Arg-Tyr-Leu-Pro-Thr) both potentiates neurally evoked contractions and causes contractures of insect skeletal muscle. In the hindleg extensor tibiae muscle of the locust, Schistocerca gregaria, the proctolin analogue [Afb (p-NO2)2]-proctolin is also able to potentiate neurally evoked contractions but is approximately 1,000-fold less effective in evoking contractures. 2. Proctolin and [Afb (p-NO2)2]-proctolin are equipotent in their ability to elevate the second-messenger inositol trisphosphate in isolated extensor tibiae muscle fiber membranes. 3. [Afb (p-NO2)2]-proctolin is approximately 1,000-fold less effective than proctolin in reducing the resting potassium conductance (GK) in extensor tibiae fibers. 4. We conclude that the action of proctolin on the extensor tibiae muscle is mediated by at least two receptor subtypes and that [Afb (p-NO2)2]-proctolin acts selectively on the receptor that potentiates neurally evoked contractions.

Analysis of peptidic epitopes recognized by the three monoclonal antibodies specific for the same region of glycophorin A but showing different properties.

Analysis of epitopes for the three monoclonal antibodies (GPA105, GPA33, OSK4-1) against glycophorin A (GPA) was performed with the use of proteolytic fragments of GPA, the synthetic nonapeptide with the sequence of amino acid residues 35-43 of GPA, and a series of peptides synthesized on plastic pins. The antibodies were specific for a short peptide sequence RAHE (a.a. 39-42 of GPA, MAbs GPA105 and OSK4-1) or RAHEV (a.a. 39-43 of GPA, MAb GPA33). Despite recognizing the same fragment of GPA, the three antibodies showed differences in fine specificity and in response to antigen desialylation. Reactions with single replacement analogs of the RAHEV sequence showed that immunodominant (unreplaceable) residues for the MAbs GPA33 and OSK4-1 were His and Glu, respectively, whereas no such residue was found for the MAb GPA105. Desialylation of the antigen gave strong enhancement of reactivity with the MAb GPA33, moderate--with the MAb GPA105, and weak or no enhancement of reaction with the MAb OSK4-1. The results showed that monoclonal antibodies directed against the same fragment of the polypeptide chain of densely glycosylated antigen may recognize different subsites which are masked at different degree by sialic acid residues.

Insect peptide hormones, an overview of the present literature.

A comprehensive overview of the recent state of the art of insect peptide hormones with chemical structures is presented. An increased interest in insect neuropeptides and dynamic development of that research area has been influenced by a rapid improvement of instrumentation necessary for isolation and structural characterization. Several research teams have studied the relationships between biological properties of insect and vertebrate peptide hormones. Thus hormones from the AKH family can be considered glucagon counterparts, whereas the myotropic hormones such as proctolin and Lem-PK (LPK) are a substance P equivalent. Insect melanization hormones Bom-MRCH in their structural characteristics and properties resemble those of mammal MSH, and leucosulfakinins Lem-SK-I and -II show some similarities with gastrin II and cholecystokinin. Bombyxin-II (Bom-PTTH-II) reveals a structural homology with human insulin and similar biological properties to adenocorticotropic mammal hormone. Allatostatin (Dip-JHS-I) may be compared to somatostatin as it can be inferred from the observations that this peptide modulates JH secretion in cockroach, Blattella germanica. Determination of the primary structure of eclosion hormones Mas-EH and Bom-EH-II as well as the amino acid sequence of allatotropin and allatostatin is a significant contribution to the understanding of the molecular mechanisms of metamorphosis and insect development.

[Hyp3]-tuftsin ([Hyp3]-TU) synthesis and biological activity.

[Hyp3]-tuftsin (Thr-Lys-Hyp-Arg) has been synthesized by the liquid-phase method. In biological investigations performed on rats antinociceptive and diuretic effects have been determined. It has been suggested that the presence of hydroxyl substituent in pyrrolidine ring of proline slightly modifies antinociceptive TU effect and is responsible for the increased diuretic [Hyp3]-TU activity.

Role of guanidine group at the N-terminal proctolin chain in cardioexcitatory effects in insects.

Six proctolin analogues (I-VI) modified in position 1 of the peptide chain by the following amino acids: homo-Arg, Gac, Gav, Gap, Phe (p-guanidino) and Orn, were synthesized by conventional liquid phase method. The myotropic activity of the obtained peptides was investigated in cardioexcitatory test on two insect species, cockroach, Periplaneta americana L., and yellow mealworm, Tenebrio molitor.

Further studies on proctolin analogues modified in position 2 of the peptide chain and their influence on heart-beat frequency of insects.

Seven proctolin analogues (I-VII) modified in position 2 of the peptide chain by Phe (p-guanidino) (I), Phe (p-OEt) (II), Tyr (3'-NH2) (III), Tyr (3'-NO2) (IV), Afb (p-OH) (V) (Afb = 3-amino-4-phenyl-L-butyric acid), Afb (p-NH2) (VI), Afb (p-NO2) (VII), and the tetrapeptide Tyr (3'-NH2)-Leu-Pro-Thr (VIII) were synthesized by the classic liquid-phase method. The biological effects of the peptides were investigated in cardioexcitatory tests on two insect species, the cockroach Periplaneta americana L., and the yellow mealworm, Tenebrio molitor L. Within physiological concentrations (10(-9)-10(-7) M) peptides II, III, and IV stimulated the heart action of P. americana like proctolin itself. Under identical conditions, in the case of T. molitor, only peptide III showed cardiostimulatory properties, whereas other compounds (including II and IV) were inactive at concentrations up to 10(-7) M. Results reported here reflect, with reference to the analogues I-VII, selective recognition of receptors on myocardium of both insect species. The tetrapeptide VIII revealed a weak deacceleratory effect on P. americana and T. molitor heart action.

Tuftsin a natural peptide with antiviral activity--structural basis of its action.

The influence of tuftsin--the natural phagocytosis stimulating tetrapeptide and its elongated analogues--on the mortality of encephalomyocarditis virus-infected NMRI mice was investigated. It seems that amino acids elongation in the parent peptide chain does not result in significant increase of antiviral activity of the compounds tested.

The influence of synthetic tuftsin and its analogs on the function of granulocytes of children with acute lymphoblastic leukemia.

The effect of tuftsin, its synthetic analogs and arginine on phagocytosis of Staphylococcus aureus by granulocytes from leukemic children was investigated in vitro. The high stimulatory effect of tuftsin and arginine was shown. The decrease of phagocytosis of PMN from healthy subjects after preincubation with its analogs and arginine was observed, suggesting the regulatory effect of these peptides.

Proteolytic enzymes in peptide synthesis.

In this paper, the literature of the past few years on the application of the proteolytic enzymes in peptide synthesis is summarized. The principle is sound and peptide synthesis for commercial purposes appears feasible. The exclusion of water or its use at a low concentration in immiscible or in biphasic media ensures controlled hydrolysis. The use of proteolytic enzymes attached to a solid support has proved a convenient method of synthesis. Synthesis of peptides with some enzymes occurs by bond exchange but with others the enzymes can extract a water molecule from a free carboxy and an unprotected group so as to create an amide bond.
