[Results of irradiation using different radiation sources in the treatment of malignant parotid tumors].

Patients with malignant parotid tumors were treated at two different centres with slightly ionizing radiation (110 patients) and neutron therapy (15 patients). The treatment results are compared. The principles of comparison applied in this analysis are identical. The histologic classification was made according to the W.H.O. recommendation of 1972 and the staging according to Becske. Local tumor control is the criterion applied in the comparative assessment of both methods. The analysis has shown that there is no significant difference in the treatment results within the individual stages of tumor's advancement after application of an orthovolt therapy and a telecobalt therapy. Patients treated in early stages (stages I and III) had a substantially higher rate of local tumor control (72%) than patients in advanced stages (46%, stages III and IV). A more favorable treatment results was achieved by neutron therapy in all stages of tumor's advancement. In this case the rate of patients with local tumor control (87%) was significantly higher than after radiotherapy with slightly ionizing radiation (46%).

[Testing of the renal capsule technic in normal immunocompetent mice].

We have examined the possibility of transplanting syngeneic and xenogeneic tumor material below the renal capsule of normal, immunocompetent mice as a chemotherapeutic test model. Whereas the Lewis lung carcinoma in the syngeneic system was appropriated for the 6-day-test, only 56% of spontaneous C3H mammary carcinomas showed an increase in the size of the transplanted material. An individual sensitivity to the cytostatic agents as an expression of biological variability could be detected for primary C3H mammary carcinomas. Both, xenogeneic lines of human origin, established in nude mice, and fresh resection material from the clinic showed on the normal, immunocompetent mouse no time-dependent, reproducible growth of the transplanted material in control animals. However, growth is considered as a precondition for estimating the effect of therapy. The normal, immunocompetent mouse was in our experiments not suitable for application of the renal capsule assay in the 6-day-test as a recipient for xenogeneic transplantation.

Establishment of a tumor cell line and its use for an agar-free colony-forming assay.

The establishment of a spontaneously transformed tumorigenic human fibrosarcoma line derived from the ovary of a breast cancer patient (NO-15) is described. This tumor cell line has been characterized by tumor growth in nude mice. The cells form colonies without the usually recommended agar technique and are sensitive to cis-dichlorodiammineplatinum(II) (cis-DDP), daunorubicin (DR) and bleomycin (BLM), three well-known chemotherapeutic agents. The cells were incubated in the presence of drugs using three different methods. These agents proved to be noncytotoxic to monolayers treated for three hours. Conversely dose-dependent cytotoxicity was seen in suspension cultures treated for three hours. Optimal correlation between the three methods for the evaluation of cytotoxicity was seen in monolayers exposed to these agents for 24 hours. The results of three different methods to evaluate drug efficacy (number of colonies, growth rate and 3H-thymidine incorporation) were in good correlation. The incorporation of the radiolabeled tracer into cellular DNA is advantageous for its rapidity and quantitative reproducibility.

DNA distributions in human normal, precancerous and cancerous breast tissue. I. Ploidy and cell cycle distribution.

DNA distributions were recorded flow cytometrically in 19 normal, 23 precancerous tissues with epithelial proliferation and cell atypia (P1), 7 carcinomata lobularia in situ and intraductale carcinomas (P2) and in 44 invasive carcinomas of the human breast. The results were compared to histological findings, tumor staging and grading, lymph node involvement and age of the patients. DNA diploidy was found in all normal, 21 P1 tissues, 6 P2 tissues and 12 carcinomas; DNA aneuploidy in 2 P1 tissues, 1 P2 tissue and 32 carcinomas. Lobular carcinomas were preferentially involved in the DNA diploid group, duct carcinomas in the DNA aneuploid group. Cell cycle distribution analysis resulted in significantly higher percentages of cells in S- and G2M phase in DNA aneuploid carcinomas compared to normal and precancerous (P1, P2) tissues. The percentage of cells in G2M phase was significantly higher in DNA aneuploid carcinomas originating from patients younger than 50 years compared to patients older than 50 years. Correlations between the DNA-index, cell cycle distribution, lymph node involvement, tumour staging or histological type were not evident.

[Ultrasound-guided fine needle biopsy in tumor diagnosis].

Ultrasound guided fine needle biopsy led in 92 of 100 patients with suspected tumours to correct diagnostic results which determined the diagnostic and therapeutic management. Ultrasound-tomography may thus in connection with fine needle biopsy enlarge and simplify tumour diagnosis.

Studies on assay of estradiol binding proteins by cytochemical and immunocytochemical techniques in human breast tumors and other tissues.

Estradiol binding proteins in frozen sections of human breast tumors, ovarian carcinomas, hypernephromas, rat uteri and cow udders were demonstrated by fluorescent labeled estradiol bovine serum albumin conjugate (cytochemical or direct method). The specificity of the binding of estradiol conjugate to tissue sections was tried to approach by addition of various steroid or non-steroidal competitors. The estradiol conjugate binding sites were practically not present in sections of human spleen, cross muscle and colon muscle.

Monoclonal antibodies against antigens of the human mammary carcinoma cell line MCF-7.

Mouse monoclonal antibodies against the human mammary carcinoma cell line, MCF-7, were produced and tested against a panel of cell lines. They recognize a variety of intra- and extracellular antigens. Four of them did not react with the human rhabdomyosarcoma cell line A-204 used as a discriminating second target during initial screening. One of these, 45-B/B3, is specific for epithelia, and probably recognizes a cytokeratin. This antibody clearly differentiates carcinomas from sarcomas and lymphomas, and epithelial cells in culture from fibroblasts. It is not species-restricted.

High risk breast cancer: long-term surgical adjuvant therapy based on predictive tests - preliminary report.

A surgical adjuvant therapy trial was started in 1974, in locoregionally advanced breast cancer. The adjuvant hormonal and cytostatic therapeutic regimen was administered according to the results of predictive tests: estrogen receptor (ER) assay and oncobiogram (group I); in the control group (0) no adjuvant therapy was given until relapse, but tests were performed like in group I. Preliminary results. In a subgroup with adjuvant chemotherapy, using one or more substances according to oncobiogram, with or without hormonal treatment, the treated women did highly-significantly better, compared with the untreated group. No advantage was seen from ovariectomy and/or anabolic steroids, either with or without Cytoxan (Test results: ER +, in vitro not growing tumours). Some doubt exists about the results: resistant, especially in testing cytostatics. This doubt and the rate of tumours, not growing in vitro, limit the use of predictive tests, concerning cytostatics. Further, one has to be cautious in the use of additive hormonal treatment in surgical adjuvant therapy: in some cases stimulation can not be excluded.

[Application of the serological clostridium assay in tumour diagnostics (author's transl)].

The suitability for serologic tumour diagnostics of the non-oncolysing strain Clostridium butyricum CNRZ 528 has been investigated. By introducing the transplantable Brown Pearce tumour of the rabbit and spontaneous tumours of the dog as test models the antibody production against clostridial rods--indicating malignant growth as had been proved with the Möse strain Cl. oncolyticum M55--has been evaluated. It could be established--utilizing the complement fixation test--that one serological method alone is not sufficient for obtaining clear-cut test results in anyone case. It is to be recommended that the indirect hemagglutination and the complement fixation tests ought to be combined. Our studies on specific clostridial antigens and on optimization of techniques will be continued.

[The adaption growth of the heart in the higher (experimental autoradiographic researches in rats) (author's transl)].

There was examined the influence of the age on the development of a compensatory hypertrophy of the heart in 4 to 16 month old masculin Wistar-rats autoradiographically. The valuation took place at various times after stenosing of the aorta and application of 3H-thymidine separatly in both ventricular walls by counting of the nuclei of the marked muscle- and connective tissue cells of the heart and their mitosis. [Learning and chronobiological regulation following experimental damage to the coronary-artery wall of albino rats].

Forty male albino rats were used to study the relationship between experimentally (concentrated hydrochloric acid) produced local structural changes of the coronary artery wall and central nervous information processing and chronobiological events, respectively. The alterations of the coronary arteries caused by this treatment did not produce infarction but initiated on the whole length of the arterial branch proliferative processes that are complete within 12 weeks following the intervention. Central nervous information processing and chronoregulatory processes showed, immediately after exposure to hydrochloric acid, pronounced malfunctions followed by a stage of hyperreactivity (4 weeks following the damage). At 8 weeks from the intervention, the parameters of information processing and chronobiological events of the CNS again equalled the control values. Since the central nervous functions were back to normal earlier than the structural alterations, the results are interpreted as reflecting CNS-controlled interactions between the complex of visceral afference and efference, the regulation of which is determined by the CNS after coronary damage in such a way as to restore the adaptational capacity of the organism by a predominance of sanogenetic processes.

[Tying with thread of the rat coronary arteries, a method for the production of experimental coronary stenosis].

A method is described for producing a localized coronary artery stenosis by tieing two threads around the Ramus descendens of the left coronary artery, which produces progressive cell proliferation of the vascular wall. Studies on 102 male Wistar rats (70 experimental and 32 sham-operated control animals) have shown coronary artery stenosis narrowing the lumen by more than 50% (up to and exceeding 90%) to develop after 7 days in 30% of the animals, after 14 days in 75%, and after 21 days (postoperatively) in 100%. In about one-third of the cases subtotal infarction in the supply area of the left coronary artery occured as a result of post-operative damage to the coronary artery with obturating thrombosis. The induction of effective coronary stenosis limiting the coronary reserve offers possibilities for further experimental studies on the role of relative coronary insufficiency in the pathogenesis of myocardial infarction.

[Coronary flow measurements on the isolated rat heart following experimental coronary artery stenosis].

A stenosing proliferation of the vascular wall was produced by exposing the Ramus descendens of the rat's left coronary artery to continued mechanical stimulation. Two weeks after the operation the hearts were perfused according to a modified Langendorff method, and the flow rate was measured under aerobic conditions and following short-time anoxia. Under either experimental conditions, the flow rate proved to be significantly decreased.
