A rapid and reliable sequential staining method for bovine muscle.

Various different combinations of histochemical stains at differing pH levels on single sections of bovine skeletal muscle were evaluated. The staining sequence that was the most consistent and reliable of those studied included an initial NADH-TR (reduced nicotinamide adenine dinucleotide-tetrazolium reductase) followed by an acid ATPase stain at pH 4.15. This staining combination resulted in easily discernable type I, II A, and II myofibers. Thus, a marked saving of expended time, labor and materials was realized.

Rectovaginal constriction in Jersey cattle: genetics and breed dynamics.

Data from farmer-owned herds and from experimental matings supported monofactorial recessive inheritance of rectovaginal constriction in US Jersey cattle. Kempthorne's population genetics model of a recessive trait involving only male selection was extended to include mutation and converted to selection of females only. Computer analyses with that model estimate slow decline in the frequency of the gene for rectovaginal constriction. Practical dynamics of the disorder in a breed registering 50,000 females and 2,000 males annually are given for current conditions and after 500 generations of selection.

Use of multiallelic human DNA probes to detect polymorphisms in the porcine genome.

Two human variable-number tandem repeat probes, pYNH24 and pYNA23, were examined for their possible ability to detect polymorphisms in the porcine genome. Useful DNA polymorphisms were detected in the porcine species, using both probes. In addition, results of Southern blot analysis of these markers in family studies indicated that the genome fragments obey mendelian laws of inheritance.
