Isolation and characterization of a monoclonal antibody specific for fibrinogen and fibrin of human origin.

A hybridoma cell line secreting a monoclonal antibody directed against human fibrinogen has been isolated. The antigenic determinant recognized is present on both the fibrinogen and fibrin molecules but is apparently absent from the D and E fragments and from fibrinopeptides A and B. The antibody seems to recognize a conformational structure present in native fibrinogen and in which several or possibly all the fibrinogen polypeptide chains may participate.

[Action of digoxin on systolic and diastolic time intervals. An echocardiographic study in infants].

The effects of digoxin on systolic and diastolic time intervals were studied in 25 children and infants the majority of whom had congenital heart disease by M mode echocardiography. The recordings were performed before and after the administration of digoxin. Serum digoxin levels were measured to confirm therapeutic dosage. After digoxin, the right and left ventricular pre-ejection periods, the duration of the corrected electromechanical systole and the Weissler indices decreased, and the isovolumic relaxation periods increased. The ventricular ejection times were unchanged except for the corrected right ventricular ejection time which was only slightly decreased. Our results concerning left ventricular systolic time intervals are in agreement with other studies in children. As no other studies of the effects of digoxin on the right ventricular systolic time intervals, or of the right and left isovolumic relaxation time are available, confirmatory studies are required. The decrease in the right and left pre-ejection periods, electromechanical systole and the Weissler indices, is interpreted as being related to the positive inotropic effect of digoxin whilst the increase in isovolumic relaxation reflects only a decrease in preload. This study allows a better understanding of the effects of digoxin on the different phases of the cardiac cycle and a better appreciation of its action potential.

Identification of a viral antigen recognized by H-2-restricted cytolytic T lymphocytes on a murine leukemia virus-induced tumor.

Monoclonal antibodies were produced against protein p30, a structural protein of murine leukemia viruses (MuLV) coded by the gag gene of MuLV. Three monoclonal antibodies of different isotypes (i.e., IgG-1, IgG-2a, and IgG-2b) were chosen for extensive analysis. These three antibodies bound to mouse tumor cells induced by Friend, Moloney, Rauscher, and Gross MuLV, but not to noninfected normal mouse spleen cells. The ability of these monoclonal antibodies to inhibit cytolytic T lymphocyte (CTL) activity by masking the antigens recognized by CTL on the target cell surface was studied in various CTL systems. It was found that the only CTL that were consistently inhibited in their lytic activity came from BALB.B (H-2b) mice immunized against syngeneic Gross MuLV-induced B.GV cells. These results thus showed that a subpopulation of BALB.B anti-Gross MuLV CTL recognized a Gross MuLV gag gene product expressed on the surface of B.GV cells.

[Quantitative study of the genetics of haptoglobin levels].

A quantitative study, based on several African and Pyrenean populations led to the estimate of the effect of some factors on haptoglobin rate: it shows an influence of age and electrophoretic phenotype, but no apparent effect of sex. Moreover, this study led to the conclusion that there is heritability of haptoglobin rate.

[C-type virus particles in lymphocytes cultured from cows with persistant lymphocytosis. Preliminary results on French cattle stock].

C type virus particles were shown in cultures of lymphocytes originating from cows with persistent lymphocytosis. These observations on French cattle are in complete accordance with data previously obtained in the USA. Viral replication is highly stimulated by the addition of phytohemagglutinin in the culture medium. Several morphological and biological properties, specific to the bovine virus differentiate this virus from C type viruses of other species.

[Young physicians and their private practice].

A postal survey was carried out in Spring 1999 on a random sample of 3623 physicians having settled up their private practice during the first half of the 90's. The number of replies amounted to more than 1800. The proportion who "bought the right to be introduced to the clientele" have raised during the recent years. Those who bought the "right" succeeded to obtain more rapidly a "sufficient" number of patients. In general, the financial situation of French young practitioners was good during the 90's. The proportion of failures was not higher than 3%. France has been characterized by large gaps between its geographical areas as concerns medical staffing, a feature potentially harmful to equity in access to health services. In view of designing sound policies in the matter, medical practitioners were asked if they accepted to change their installation site in case an allowance of 50,000 Francs (about 76,000 Euros) was granted. Less than 4% replied "yes" and 18% "perhaps" to the question. These figures concerned changes of site within the same region. The prospects of migrating to an other region gathered even less enthusiasm. It is evident that policies aimed at reshaping geographic medical staffing should be multi-faceted and not grounded on simple financial allowances.

[The demographic future of the pharmacy profession (1990-2020)].

Training of pharmacists in France is regulated by a numerous clausus system. However, forecasting the number of active pharmacists in the future is not an easy task for various reasons: (i) after graduation, certain pharmacists do not become occupationally active; (ii) there are many types of pharmaceutical occupations, a pharmacist may leave an occupation for an other or enter an occupation long time after graduation; (iii) for the time being, the dynamics governing retirement, mortality and other occupation leaving processes have not been observed and analyzed. Based on certain sets of reasonable assumptions, the Centre de Sociologie et de Démographie Médicales has attempted to design scenarios for future development of French pharmaceutical manpower. If the student annual intake is kept at its current level (2,250), the number of active pharmacists registered with the Ordre des Pharmaciens would rose to 60,400-63,000 in the year 2000, and to 65,400-71,300 in the year 2010, from less than 53,000 in 1990. If the numerous clausus is immediately reduced to 1,500 (i.e. a cut of one third from the current level), the number of registered pharmacists would be 57,000-58,300 in 2000 and 57,500-61,000 ten years later. All these calculations are carried out with techniques of demographic projection. It remains to know what set of figures would be the most suitable to the future need of the nation regarding pharmaceutical manpower.
