Expression patterns of chondrocyte genes cloned by differential display in tibial dyschondroplasia.

Tibial dyschondroplasia (TD) appears to involve a failure of the growth plate chondrocytes within growing long bones to differentiate fully to the hypertrophic stage, resulting in a mass of prehypertrophic chondrocytes which form the avascular TD lesion. Many biochemical and molecular markers of chondrocyte hypertrophy are absent from the lesion, or show reduced expression, but the cause of the disorder remains to be identified. As differentiation to the hypertrophic state is impaired in TD, we hypothesised that chondrocyte genes that are differentially expressed in the growth plate should show altered expression in TD. Using differential display, four genes, B-cadherin, EF2, HT7 and Ex-FABP were cloned from chondrocytes stimulated to differentiate to the hypertrophic stage in vitro, and their differential expression confirmed in vivo. Using semi-quantitative RT-PCR, the expression patterns of these genes were compared in chondrocytes from normal and TD growth plates. Surprisingly, none of these genes showed the pattern of expression that might be expected in TD lesion chondrocytes, and two of them, B-cadherin and Ex-FABP, were upregulated in the lesion. This indicates that the TD phenotype does not merely reflect the absence of hypertrophic marker genes, but may be influenced by more complex developmental mechanisms/defects than previously thought.

Identification of a family of noncanonical ubiquitin-conjugating enzymes structurally related to yeast UBC6.

Ubiquitin-conjugating enzymes (UBCs) selectively target proteins for proteasomal degradation by the covalent attachment of ubiquitin moieties. Yeast UBC6 is unusual in having an active site distinct from all other UBCs and in possessing a transmembrane domain that anchors it to the cytoplasmic surface of the endoplasmic reticulum. During a differential display analysis on chick growth plate chondrocytes we isolated a cDNA encoding a noncanonical ubiquitin-conjugating enzyme (NCUBE1) structurally similar to yeast UBC6. Chick NCUBE1 transcripts were detected in all tissues examined and decreased threefold during chondrocyte terminal differentiation. Database searches identified other related proteins; the human and mouse orthologues of NCUBE1, a second human homologue of yeast UBC6 (NCUBE2), and related proteins from S. pombe, C. elegans, and P. mariana. Together with yeast UBC6 these proteins constitute a distinct family of UBCs sharing a conserved noncanonical active site sequence and a C-terminal transmembrane domain. By analogy with yeast UBC6 they are likely to be localised to the endoplasmic reticulum where they may be involved in targeting retrotranslocated, ER-associated proteins for proteasomal degradation.

Shame and suicide: a case study.

Building on a recent paper by Lester (1997) on shame and suicide, a case is presented of a woman who made a serious attempt to kill herself, motivated in large part by shame. The therapeutic strategies employed are described, and the case brought to a successful resolution.

Murder in Russia.

Ecological studies of regional suicide rates in Russian oblasts and krais and American states revealed that the clustering of social indicators and the social correlates of regional homicide rates were different in the two nations. However, indices of social distress (unemployment and illegitimate births) predicted homicide rates in both nations. The study pointed out the importance of replicating research in several nations in order to explore the generality of research findings and theories.

Comparing correlations over time and space.

Correlations between the suicide and birth rates in Canadian provinces from 1970 to 1990, estimated using three different methods (over time, over space, and mixed) were small but negative.

Controlling the environment to prevent suicide: international perspectives.

BACKGROUND: Suicide and suicidal behaviour are multifaceted events requiring complex solutions. Controlling the environment is a neglected solution, despite strong support for this approach from the World Health Organization (WHO). METHOD: To discuss this approach from a global view, this review is written by authors from various cultures: American, Australian, Canadian, Chinese, Cuban, Dutch, Indian, Irish, Japanese, Lithuanian, Native North American, Russian, and South African. RESULTS: We examine gun control to illustrate the environmental control approach; however, the worldwide diversity of suicide methods calls for diverse responses. Further, controlling the environment encompasses more than restricting the means of suicide, which we illustrate with examples of toned-down media reports and restricted medicine availability. CONCLUSIONS: Controlling the environment may be a viable strategy for preventing suicide, although research shows that few clinicians implement such approaches.

Predicting anxiety in competitive sports.

The relationship between attitudes toward winning and competitive anxiety was explored in 59 undergraduate students competing on four different New Jersey-based, NCAA Division III sports teams. In a backward multiple regression analysis, cognitive anxiety was predicted significantly only by self-confidence, while somatic anxiety was predicted significantly by self-confidence and the rated importance of the competition.

Correlates of rates of suicide and homicide committed by different methods.

The social correlates of suicide rates by each method over time in the USA from 1950 to 1985 were very different, suggesting that suicides by different methods may not be sociologically identical behaviors. Homicide rates by each method, however, showed a more consistent pattern of social correlates.

Cause of Death↗

Executions as a deterrent to homicide.

For the period 1977-1992, the number of homicides in a state tended to decrease more often after a year of no executions than after a year with one or more executions, the opposite of a deterrent effect.

The pace of life and suicide.

The pace of life was not associated with the suicide rate in 25 nations of the world in the early 1990s.

Contacting the dead and national character.

In a sample of 11 European nations, the proportion of residents who reported experiencing contact with the dead was positively associated with Lynn and Hampson's measure of national estimates of neuroticism.
