POLYPEPTIDE CHAINS OF ANTIBODY: EFFECTIVE BINDING SITES REQUIRE SPECIFICITY IN COMBINATION.

Separated chains from antibody of different rabbits do not always give effective binding sites although combination of the chains does occur. There is a specificity of combination such that only chains from the same (or perhaps a related) rabbit form effective binding sites. There appears to be preferential combination between those chains which yield effective sites.

ANTIBODIES AGAINST THE COMPONENT POLYPEPTIDE CHAINS OF BOVINE INSULIN.

Antibodies were produced in guinea pigs against A and B chains of bovine insulin; the chains were prepared by cleavage of all the disulfide bonds with sulfite. Antibodies against A chain reacted with A chain, but not with B chain or native insulin. Antibodies against B chain reacted strongly with both B chain and insulin, but not with A chain. Antibodies against insulin reacted strongly with insulin, weakly with B chain, and not at all with A chain. The results indicate that the insulin-reactive antibodies in antiserums to B chain and to insulin differ and are directed to differing individual antigenic regions of the insulin molecule.

ANTIGEN-BINDING ACTIVITY OF 6S SUBUNITS OF BETA-2-MACROGLOBULIN ANTIBODY.

Direct evidence is provided for the antigen binding activity of the 6S subunits formed by reduction and alkylation of rabbit beta2-macroglobulin antibody. Binding activity of the subunits was clearly demonstrated by radioimmunoelectrophoresis with a preparation of purified rabbit antibody against the p-azobenzenearsonate group, which contained 40 percent of beta2M-antibody. The precipitate arc formed between the subunits and sheep antiserum against rabbit macroglobulin was shown to bind radioactive antigen specifically.

Antibody combining site: the B polypeptide chain.

The antibody molecule consists of several polypeptide chains. Peptides, which appear to have been derived from the binding region of the rabbitantibody molecule directed against pazobenzenearsonate, have been isolated. The particular polypeptide chain from which these peptides are derived has now been identified as the B chain described by Fleischman, Pain, and Porter.
