Topographical mapping of GnRH receptors on dispersed mouse pituitary cells by backscattered electron imaging.

Mouse anterior pituitary cells cultured for 2 days were stimulated with one of three biotinylated-GnRH probes ([biotinyl-Lys6]-[D-Lys6]GnRH, [biotinyl-Ser4]-[D-Lys6]GnRH, [biotinyl-Ser4]-[D-Trp6, des-Gly10]GnRH) in the cold (4 degrees C) for 1 hr. These cells were subsequently fixed and an avidin-gold complex was conjugated to the bound GnRH. After a second fixation, the gold label was silver-enhanced for viewing with a scanning electron microscope. Gonadotropes were identified as a result of the labeling procedure, measured for size, and the number of GnRH receptor sites counted. Gonadotropes ranged from 3 to 13 microns in diameter and contained from 23.2 +/- 3.3 to 338.4 +/- 25.2 sites per cell depending upon the size of the cell and the ligand employed. The methods described should be applicable for studying the topographical distribution of a variety of cellular receptors.

Incidence of Campylobacter species in animal handlers.

From the stool samples collected from diarrhoeic and healthy handlers of domestic animals different Campylobacter species were isolated in 16.6 and 21.1 per cent respectively. Both biotypes 1 and 2 were identified. All the 10 strains of C. jejuni were found to be enterotoxigenic or potentially enterotoxigenic in the ligated ileal loop of albino rats.

Levels of alpha-inhibin in aging female mice.

alpha-inhibin was immunocytochemically localized in granulosa cells of different stages of developing follicles, freshly formed corpora lutea, and scattered interstitial cells (pigmented or ceroid cells) in ovaries of 6-, 14-, and 23-25-mo-old C57BL/6NNia mice. Developing follicles exhibited the greatest amount of staining. Quantitation of the stain using an image analysis system indicated the staining intensity within ovarian follicles of 14-mo-old mice was greater than that in 23-25-mo-old mice. The levels of plasma alpha-inhibin and estradiol (E2) decreased with age. The number of follicles present in ovaries of middle-aged mice was comparable to those of 6-mo-old mice, yet plasma levels of FSH were significantly higher than those of 6-mo-old mice. This may be due to an age-related loss in the sensitivity of the hypothalamus and/or pituitary of middle-aged mice to ovarian hormones. In contrast, ovaries of 23-25-mo-old mice contained few antral follicles and consequently produced little alpha-inhibin. There appeared to be little negative feedback regulation of FSH secretion in 23-25-mo-old mice as a result of age-related ovarian impairments. This study supports an earlier hypothesis from our laboratory [Biol Reprod 1985; 32:989-997] that the primary defect(s) limiting age-related reproductive performance in mice appears to reside within the hypothalamo-hypophyseal axis, whereas secondary defects arise from the ovary.

Enterotoxigenicity of human and animal isolates of Campylobacter jejuni in ligated rat ileal loops.

Human and animal isolates of Campylobacter jejuni were tested for enterotoxigenicity in ligated loop of Charles-Foster rats. Of 21 isolates, 13 were proved to be toxin-positive in the initial experiments. However, the remaining 8 required one to three passages through the rat ileal loop before showing the capacity to produce enterotoxin. All isolates caused fluid accumulation comparable with the amount produced by the standard toxigenic strain of Vibrio cholerae 01 1naba 569B. The isolates of C. jejuni from human and animal sources did not show any quantitative difference in their capacity to produce enterotoxin.

Effect of stress on the catecholamine content of the adrenal gland of intact and bursectomized chicks.

1. Bursa-intact and bursectomized chicks were exposed to cold-wet immobilization (CWI) stress for 1.5 min. The catecholamines (CA) from the adrenal gland were measured spectrofluorometrically 5, 15, 30 and 60 min after stress. 2. In bursa-intact chicks, the CWI stress caused decrease of both norepinephrine and epinephrine from the adrenal glands 5 min after stress. Resynthesis of epinephrine exceeded the control value 30 and 60 min after stress. 3. In bursectomized chicks, the CWI stress brought about a decrease of norepinephrine 15 and 30 min and of epinephrine 30 and 60 min after stress. 4. The findings suggest that bursa facilitates early (5 min) release of CA and also helps in quick resynthesis of epinephrine during stress.

Effect of progesterone (norethindrone) only oral contraceptive on mouse uterus--a SEM study.

Impact of progesterone (norethindrone) only oral contraceptive (MICRONOR) on mouse uterus was investigated by SEM. The three dimensional SEM photographs revealed that, flat wave like mucous folds of the uterus (control) become more thick and highly convoluted after 50 days of MICRONOR feeding. The uteri became highly secretory as indicated by increased thickness of nonciliated secretory cell population with their increased microvilli. Low doses of norethindrone in combination with circulating estrogen, instead of causing regression, maintained a steady and highly secretory state which may interfere implantation/gamete transport.

Outbreak of cholera in Berasia block of Bhopal District in Madhya Pradesh.

Bhopal district, Madhya Pradesh has been considered to be free of cholera. In an outbreak of gastro-enteritis in Berasia block of Bhopal district, Cholera vibrio Inaba and Ogawa were isolated from 17.64 per cent of stool samples collected. Extreme insanitary conditions, large aggregation of uncontrolled population and probably consumption of infected ice candies were responsible for the outbreak of cholera.

The effects of slip velocity at a membrane surface on blood flow in the microcirculation.

Closed-form solutions are presented for blood flow in the microcirculation by taking into account the influence of slip velocity at the membrane surface. In this study, the convective inertia force is neglected in comparison with that of blood viscosity on the basis of the smallness of the Reynolds number of the flow in microcirculation. The permeability property of the blood vessel is based on the well known Starling's hypothesis. The effects of slip coefficient on the velocity and pressure fields are clearly depicted.

Sporotrichosis in India: first authentic case report from the north-western region and a critical literature review.

The first case of sporotrichosis from north-western region is reported along with a critical review of the work done on this disease in India. The patient, a 55-year-old housewife with multiple lymphocutaneous lesions on her left arm, had never travelled to north-eastern India where sporotrichosis occurs endemically. She was a resident of Ranikhet, District Nainital, Uttar Pradesh (U.P.), situated about 235 km north-east of Delhi. The diagnosis was established by demonstration of Sporothrix schenckii through direct microscopy and culture of pus collected from multiple suppurative lesions, verification of the dimorphic character of the fungus in vitro and its pathogenicity to white mice. She was treated successfully with oral administration of potassium iodide for 6 weeks.

Magnum morphogenesis during the natural development of the quail oviduct: analysis of egg white proteins and progesterone receptor concentration.

The histological development of the quail oviduct and the changes in concentrations of progesterone receptor, ovalbumin, conalbumin, ovomucoid and ovoglycocomponents are analyzed during the period spanning 7-35 days of age. The initiation of luminal epithelial cell proliferation is the first event of magnum growth. The epithelial cells begin to evaginate into subepithelial stroma and form tubular glands. Meanwhile, luminal epithelium starts cellular pleomorphism through ciliogenesis. No egg white proteins are detectable in the developing glands; at the same time, the concentration of the progesterone receptor increases from about 5500 sites/cell to 30,300 sites/cell. Tubular gland cells then begin to synthetize and accumulate egg white proteins, mucous cells differentiate in the luminal epithelium, and the cell proliferation decreases and finally stops. Compared with earlier studies dealing with the blood levels of estrogen and progesterone in developing quails during the same period, and the cellular changes induced in the oviducts of ovariectomized and ovariectomized-hypophysectomized quail by exogenous steroids, these results distinguish between the cellular responses that are physiologically controlled by estradiol and other responses that have multihormonal regulation.

Effect of couple stresses on the unsteady convective diffusion in fluid flow through a channel.

A generalized dispersion model is used to obtain exact solution for unsteady convective diffusion in the presence of couple stresses. The effect of the couple stress parameter 'a' on the most dominant dispersion coefficient is clearly depicted. The dimensionless mean concentration distribution is obtained as a function of dimensionless axial distance, time and 'a'. The results for 'pure convection' are also reported. It is shown that the effect of couple stress is predominant only for small values of 'a' and when a----infinity the flow characteristics tend to their equivalents in Newtonian theory. The results of Taylor's dispersion model are recovered as a particular case in the limit tau----infinity.

Blood Physiological Phenomena

Report of a study into skin sensitivity and into scar diameter after vaccination with BCG in 5,000 new-borns in Cakovec, Yugoslavia.

To compare the skin allergy producing potency of batches of BCG from three different production centers, 5,066 new-borns were vaccinated with one of there batches (one from the Netherlands, one from Yugoslavia and one from Denmark). Six, twelve and twenty-one months after vaccination tuberculin testing was done and the diameter of scars was measured in subgroups. Skin allergy through BCG-vaccination was registered to be highest after 6 months and appeared to wane rapidly afterwards. There was a definite difference in the skin allergy potency between the three vaccines. The difference in scar diameter was less pronounced. In 5,066 vaccinated new-borns 137 (2.7%) enlarged axillar glands were registered. There appeared to be a positive correlation between the mean Mx-induration 6 months after vaccination and the percentage of enlarged glands. No serious complications have been registered so far.

Micro-indentation hardness studies on human bones.

Bones from different portions of human skeleton are polished and cut to suitable sizes and then subjected to micro-indentation at various loads using Vicker's diamond pyramidal indenter. The Vicker's hardness number is found to differ slightly from portion to portion of the skeleton. Interferometric studies of the indentation reveal that the applied stress of indentation causes a 'pile-up' of material near the corners of the pyramidal indents.
