The structure of the ferric siderophore binding protein FhuD complexed with gallichrome.

Siderophore binding proteins play a key role in the uptake of iron in many gram-positive and gram-negative bacteria. FhuD is a soluble periplasmic binding protein that transports ferrichrome and other hydroxamate siderophores. The crystal structure of FhuD from Escherichia coli in complex with the ferrichrome homolog gallichrome has been determined at 1.9 ¿ resolution, the first structure of a periplasmic binding protein involved in the uptake of siderophores. Gallichrome is held in a shallow pocket lined with aromatic groups; Arg and Tyr side chains interact directly with the hydroxamate moieties of the siderophore. FhuD possesses a novel fold, suggesting that its mechanisms of ligand binding and release are different from other structurally characterized periplasmic ligand binding proteins.

A memory retraining program for nontoxic chronic alcoholics.

This study assessed the effectiveness of a memory rehabilitation program for fifty-nine alcoholics, using a pretest/posttest control group design. The Dooks Memory Test evaluated the different facets of memory, and the Trailmaking Test A and B measured attention and concentration. An analysis of covariance yielded a significant main effect for the Memory Retraining Program (p less than 0.001). The covariate for the composite memory scores was also significantly (p less than 0.025). There was also a significant effect of Age (p less than 0.025) and of IQ p less than 0.002. There were no interaction effects between age and treatment, or IQ and treatment, and no other independent variables (education, duration, days since last drink, depression) were significant. The results point to the potential effectiveness of a memory retraining program for a variety of educational levels, age groups and IQ scores.

Disturbances of attention in alcohol withdrawal syndrome.

Disturbances of attention have often been noted clinically in the alcohol withdrawal syndrome, but no studies to date have systematically explored this phenomenon. The present study examined attentional impairment in 20 tranquilizer-free alcohol withdrawal patients by using continuous performance tests assessing concentration in both an auditory and a visual mode. It was found that withdrawal patients generally evidenced poorer auditory than visual concentration. To investigate the relationship between the time-course of withdrawal and attentional impairment, the alcoholics were divided into an Early "Detox" and a Late "Detox" group, predicated on the reported amount of time since last alcohol consumption. Contrary to expectation, those alcoholics with a longer time-lapse since cessation exhibited poorer attentional performance than those with a shorter time-lapse. The possibility of an independent course for mental symptoms apart from physiological withdrawal signs was discussed.
