31P-NMR saturation transfer studies of aerobic Escherichia coli cells.

31P-NMR measurements of saturation transfer have been used to measure the flux between Pi and ATP in Escherichia coli cells respiring on an endogenous carbon source. Measurements were made in the wild type and in cells genetically modified to give a 5-fold higher concentration of the F1F0-ATP synthase. The flux in the two cell types was not significantly different. This, together with studies using inhibitors specific for the glycolytic enzyme, glyceraldehyde-3-phosphate dehydrogenase and the ATP synthase, suggests that the observed flux arises predominantly from glycolytic rather than ATP synthase activity. Although this conclusion is in disagreement with previous experiments on E. coli, it is in agreement with recent experiments on yeast.

The impact of alcohol ingestion on the orthopaedic and accident service.

There is a well recognized association between the ingestion of alcohol and trauma. The incidence of orthopaedic injuries related to alcohol ingestion and requiring admission has not previously been reported. In a pilot study from the Royal Liverpool Hospital we found that in 22.4% of patients requiring admission, alcohol had been consumed prior to the accident. The incidence of spinal and ankle fractures was higher in the inebriated patients and the circumstances of their injuries differed from those who were sober. Injuries following drinking occurred mainly in young and middle aged males. We discuss simple methods of recognising problem drinking among accident and orthopaedic patients.

Metabolic effects of interleukin 3 on 32D cl23 cells analyzed by NMR.

31P NMR of living 32D cl23 cells and 1H NMR of cell extracts were used to study the metabolic effects of interleukin 3 (IL3). When IL3 was removed from 32D cl23 for 9-10 hours 31P spectra showed a decrease in sugar phosphate, gamma ATP/ADP, alpha ATP/ADP/NAD, and beta ATP resonances which declined progressively over a time period of up to 16 hours. By comparison, ATP measurements using the luciferin/luciferase method resulted in the decline of ATP levels from 12 hours in the absence of IL3. At this time, viability of the cells was unaffected. For 1H NMR experiments cells were grown in the presence and absence of IL3 for 4 and 24 hours, after which acid cell extracts were prepared. These spectra revealed a four-fold decrease in lactate 4 hours post-IL3 removal. Alanine levels were unchanged but glycine was elevated 1.5-fold whilst various other amino acids were elevated slightly. After 24 hours without IL3, only 22% of cells were viable which was reflected in a general decline of most resonance intensities. These findings suggest that IL3 exerts its effect primarily on glucose metabolism and has a delayed secondary effect on maintenance of ATP levels in the cell. We have demonstrated the applicability of high resolution 1H and 31P NMR to the study of cellular metabolism in hemopoietic cells.

Lack of sampling site variation in chorion villus biopsy as assessed by DNA, enzymatic, morphological and cytogenetical analyses.

A study of villus samples from eight random sites on five electively aborted chorion sacs was performed to determine any significant differences in yield, quality and composition of DNA, iduronate sulphate sulphatase activity and karyotype status. The villi were also examined for their histological characteristics (e.g. stem, intermediate or terminal villi) and for HCG and BGP immuno-reactivities. The overall findings indicated no significant site to site variations in any of the parameters studied. It is therefore proposed that any villus should be equally suitable for prenatal diagnosis.

Acetabular protrusion and the Monk Duopleet prosthesis in subcapital femoral neck fractures.

A total of 166 patients with acute, displaced subcapital fractures of the femoral neck were treated over a 5-year period with the Monk Duopleet bi-articular prosthesis. The average age at operation was 77.8 years. 81 patients were available for review at an average of 37.1 months after surgery. Radiographic evidence of acetabular protrusion was found in 3 patients (3.7 per cent). This incidence of protrusion is considerably lower than that normally reported for one-piece prostheses such as the Austin Moore or Thompson. The causes of acetabular protrusion are discussed.

Refracture of the patella.

Mildly displaced fractures of the patella, with an intact extensor mechanism, are widely treated in a plaster cylinder. This may lead to disuse osteoporosis of the patella and a subsequent refracture, which may be at a different site. A suggested method of splintage is to use a removable back splint for a period of 1-2 weeks.

The spontaneous release of transmitter from insect nerve terminals as predicted by the negative binomial theorem.

1. In the retractor unguis muscles of Periplaneta americana and Blaberus giganteus, the rate of spontaneously occurring miniature potentials from nerve-muscle synapses associated with the ;white' muscle fibres was greater than that observed in the ;red' muscle fibres.2. In 5% of the intracellular recordings, particularly from the ;white' fibres, short ;bursts' of relatively high (20-100 sec(-1)) rates of m.e.p.p.s occurred infrequently in an unpredictable manner. This bursting phenomenon was not observed in extracellular recordings of the miniature potential discharge.3. Rigorous statistical analyses indicated that the spontaneous mode of transmitter release at cockroach retractor unguis muscle fibres was predictable by the negative binomial theorem with a 90% reliability.4. Perfusion of nerve-muscle preparations with high Mg(2+) salines resulted in low spontaneous discharge rates which were characterized by a negative binomial distribution.5. The fit of the data to the negative binomial theorem implied some mutual interaction between those processes responsible for the spontaneous release of transmitter from cockroach nerve terminals.
