Nucleotide sequence analysis of a 30-kb region of the bovine herpesvirus 1 genome which exhibits a colinear gene arrangement with the UL21 to UL4 genes of herpes simplex virus.

We report the nucleotide sequence of the 19-kb HindIII fragment B of bovine herpesvirus 1 (BHV-1) DNA and adjacent parts of the HindIII A and L fragments, which together span a still completely uncharted 30-kb region located between the glycoprotein H gene and the right end of the unique long segment. The analysis revealed 17 complete open reading frames (ORFs) and 2 ORFs that were interrupted by potential splice donor and acceptor sites. All of these ORFs exhibited strong amino acid sequence homology to the gene products of other alphaherpesviruses. The BHV-1 ORFs were arranged colinearly with the prototype sequence of herpes simplex virus 1 in the range of the UL21 to UL4 genes. Colinearity was also observed with the genes of betaherpesviruses and gamma herpesviruses, although not all ORFs exhibited clear sequence homology. The possible functions of the proteins encoded within the sequenced region are assessed and features found are discussed. Unexpected findings include the following: high amino acid sequence conservation among alphaherpesviruses despite large differences in G + C content, ranging from 45% for varicella zoster virus to 72% for BHV-1; high similarity with other UL20 proteins at the predicted structural level in spite of relatively low amino acid homology; and a 2-kb open reading frame overlapping UL19 in the opposite sense and exhibiting high amino acid similarity to the same area of pseudorabies virus.

The New York High-Risk Project. Psychoses and cluster A personality disorders in offspring of schizophrenic parents at 23 years of follow-up.

BACKGROUND: We herein present lifetime prevalence rates of psychoses and DSM-III-R cluster A personality disorders in sample A of the New York High-Risk Project, a prospective study following offspring of parents with schizophrenia (HRSz subjects) and affective illness (HRAff subjects) and of psychiatrically normal parents (NC subjects) from midchildhood to adulthood. METHODS: We interviewed the offspring in adulthood with the Schedule for Affective Disorders and Schizophrenia, Lifetime Version, for Axis I disorders and the Personality Disorder Examination for Axis II disorders. RESULTS: Lifetime prevalence rates (+/- SE) of schizophrenia and unspecified psychosis were 11.1% +/- 4.3% and 5.6% +/- 3.1%, respectively, in the HRSz group and 0% in the HRAff and NC groups. Rates of schizoaffective disorder subclassified as mainly schizophrenic, however, were highest in the HRAff group. Rates of psychotic affective disorders did not differ between the HRSz and other groups. Age-corrected morbidity risks were similar to lifetime prevalence rates. Rates of the three cluster A personality disorders did not differ among the groups, but the combined rate was greater in the HRSz and HRAff groups than in the NC group. CONCLUSIONS: Our data strongly support a specific familial liability to narrowly defined schizophrenia that is not shared by families of probands with affective disorder. Schizoaffective disorder and cluster A personality disorders, however, occur in families of both schizophrenic probands and probands with affective disorder. Psychotic affective disorders, which are not increased in HRSz subjects, do not appear to be an expression of the liability to schizophrenia.

Assessment of the magnitude of anxiety in adults undergoing first oxygen treatment in a hyperbaric chamber.

The purpose of this study was to determine the magnitude and specific foci of anxiety experienced by patients receiving their initial hyperbaric treatment. The Spielberger State-Trait Inventory questionnaire and a personal interview were administered to 24 patients to determine anxiety levels. Upon arrival at the hyperbaric clinic and prior to any type of health teaching, the State and Trait questionnaire was administered to the patients. One hour prior to the treatment, the State questionnaire was repeated to determine any significant changes in anxiety levels. No significance was found in anxiety levels between pre-health and pre-treatment. The Trait questionnaire revealed that this specific group of patients, regardless of the circumstances, felt they were not very anxious individuals. Post-treatment, the State questionnaire was administered again and these anxiety levels did differ significantly from the pre-health and pre-treatment levels. The findings indicate the need to continue to validate clients' perceived needs during hyperbaric treatment, which will help direct future nursing practice.

The New York High-Risk Project: anhedonia, attentional deviance, and psychopathology.

In the New York High-Risk Project (NYHRP) we followed subjects at risk for schizophrenic or affective disorders and low-risk controls from childhood to adulthood, with the goal of identifying early predictors of later schizophrenia-related psychopathology. In this article, we focus on two potential predictors: the Physical Anhedonia Scale administered in adolescence and the Attention Deviance Index obtained in childhood. Subjects of this report are 161 members of the NYHRP's first sample (sample A), who had scores on both attention and anhedonia and had followup clinical assessments in adulthood. We used a path analysis model and several separate regression analyses to examine the relationships of the parent diagnostic groups, attentional dysfunction, and anhedonia to each other and to each of three psychopathological outcomes: schizophrenia and schizophrenia-related psychoses, major affective disorder, and social isolation in nonpsychotic subjects. Subject groups did not differ in anhedonia scores but did differ in childhood attentional dysfunction, psychosis, and social isolation, all of which are more common in subjects at risk for schizophrenia. In these subjects at risk for schizophrenia, but not in the other two groups of subjects, childhood attentional dysfunction is related to anhedonia, social isolation, and possibly nonparanoid psychosis. Anhedonia is associated with social isolation and with psychosis in females. Several other gender effects are also noted.

Characterization of dexamethasone-induced reactivation of latent bovine herpesvirus 1.

Synchronous reactivation of bovine herpesvirus type 1 in all latently infected rabbits was achieved following a single intravenous dose of dexamethasone. Reactivated latent virus was first present in ocular secretions between 48 and 72 h post-dexamethasone treatment (PT). Cell-free infectious virus, viral-antigen-containing neurons, and pathologic changes were detectable in trigeminal ganglia (TG) by 48 h PT. A shift from the viral transcriptional pattern characteristic of the latent state (latency-related RNA [LR RNA]) to one typical of that seen during acute infection was detected in a small number of neurons in latently infected TG between 15 and 18 h PT, with viral DNA first detectable by in situ hybridization at 18 to 21 h PT. The number of LR RNA-containing neurons in latently infected TG decreased significantly at 24 and 48 h PT but returned to near-normal levels by 72 h PT. Correlation of this decrease with viral reactivation suggests that altered regulation of LR RNA transcription is a significant event in the process of viral reactivation.

Early life precursors of psychiatric outcomes in adulthood in subjects at risk for schizophrenia or affective disorders.

In the New York High-Risk Project (NYHRP), 186 offspring of schizophrenic, affectively ill, and psychiatrically normal parents have been followed prospectively from 1971-72 to the average age of 27 years in 1990 with the goal of identifying early precursors of later psychopathology. In this report, we use path analyses to examine the relationship of several life-history variables to three pathological outcomes in the offspring: namely, psychosis, psychiatric hospitalization, and psychological dysfunction. The chief direct relationship with these outcomes is the effect of having a schizophrenic parent. The latter effect is also mediated indirectly by IQ.

Characterization of the latency-related transcriptionally active region of the bovine herpesvirus 1 genome.

Approximate 5' and 3' ends of the bovine herpesvirus 1 (BHV-1) latency-related RNA (LR RNA) were mapped in rabbit trigeminal ganglia (TG) by in situ hybridization. The data provide a size estimate of 0.77 to 1.16 kb for the LR RNA. An LR RNA mapping to a similar location was also detected in TG of cattle latently infected with BHV-1. The BHV-1 LR region is transcriptionally active in bovine cell cultures lytically infected with BHV-1. A 1.15-kb transcript, present at early and late times postinfection, of the same sense and approximate size that seen in latently infected TG overlaps a 2.9-kb immediate-early and a 2.6-kb early and late transcription unit present on the complementary strand. Sequence analysis of the LR RNA sense strand indicates the presence of a potential polymerase II promoter in close proximity to the 5' terminus of the LR RNA and two open reading frames within its map positions. The complementary strand contains the 3' portion of a large open reading frame that almost completely overlaps the map position of the LR RNA present on the opposite strand.

Analysis of the transcriptional promoter which regulates the latency-related transcript of bovine herpesvirus 1.

As a transcriptional promoter in primary cultures of sensory ganglionic neurons, DNA sequences near the 5' terminus of the latency-related (LR) gene of bovine herpesvirus 1 were at least 10 times more efficient than the simian virus 40 early promoter-enhancer. In contrast, as a promoter in bovine, rodent, or monkey cells, the LR promoter was approximately six times less efficient than the simian virus 40 early promoter-enhancer. The LR promoter had strict orientation preferences in neurons and all other mammalian cell lines tested. Removal of a 146-base-pair XhoI fragment from the LR promoter resulted in stimulation of LR promoter activity in bovine cells but not rabbit neurons, monkey fibroblasts, or rodent cells. LR promoter activity in bovine cells is stimulated by bovine herpesvirus 1 lytic infection, suggesting that viral gene products or virus-induced factors positively regulate the expression of the LR gene. A synthetic glucocorticoid, dexamethasone, repressed LR promoter activity in bovine cells. These results imply that a variety of factors can influence the expression of the LR gene during latent infections of neurons as well as during the lytic infection cycle.

In search of closure: subjective contour illusions, Gestalt completion tests, and implications.

This study investigates the construct validity of perceptual closure tests (CTs), and isolates a common processing demand from the right-hemisphere. Sixty-seven patients with focal unilateral lesions (34 right side, 33 left side), and 80 control subjects participated. Multivariate analyses indicated that there was substantial age-related variance in all CTs, while sex was variably significant; CTs are not uniform in their discriminating ability, and hence factorially complex; and their specifically right hemisphere-sensitive dimension was subjective contour illusions, and interestingly unrelated to facial discrimination ability. The methodological and theoretical implications are discussed.

Impact of weighing procedures and variation in protein degradation rate on measured performance of growing lambs and cattle.

A lamb growth trial was conducted to evaluate soybean meal and blood meal and to compare two experimental designs (a completely randomized design and a switchback design). A larger amount of variation was observed with the switchback than with the randomized design. A larger number of animals, longer treatment period or increased number of weighings may be needed to overcome end point weighing errors and decrease the variation in the switchback design. In both designs, there was a significantly greater conversion of protein to gain for blood meal (BM)-supplemented lambs than for soybean meal (SBM)-supplemented lambs. The relative value of BM compared with SBM was 251% (completely randomized design) or 296% (switchback design). Weighing steers 2 or 3 d at the beginning and end of the trial reduced variation and increased the probability of detecting differences in daily gain, gain/feed and protein efficiency when compared with a standard weighing method (once at the beginning and once at the end of the trial). Regressing 11 weekly weights on an average of one, two or three initial and final weights also reduced variation and increased the probability of detecting treatment differences. Regressions of weekly weights were more useful as the number of initial and final weights was decreased. Daily gain and gain/feed increased linearly (P less than .05) with protein level when BM or SBM was fed. A nonlinear model was used to predict protein requirement, maximum gain and relative protein values. There was a range in relative values for BM of 255 to 348% depending on the method of analysis used.

The children of psychiatrically disturbed parents: differences as a function of the sex of the sick parent.

To examine the effects of the sex of psychiatrically disturbed parent on the offspring, we compared children of a schizophrenic parent, a psychiatrically disturbed but not schizophrenic parent, and a normal parent using behavioral, attentional, and neurological indices. Results of a discriminant analysis indicate that children of psychiatrically disturbed mothers have an increased sensitivity to tactile stimulation, possibly reflecting more labile autonomic nervous system functioning. In addition, children with a schizophrenic parent of either sex show evidence of neurological motor dysfunction.
