On the basis of performed tests, we observed the change of pattern of pyruvate kinase isoenzyme in a malignant testicular tumor (only isoenzyme K) and the same isoenzyme in the tissue of other testis not being tumorously changed. In control group pyruvate kinase isoenzyme K and M was observed. Similarly, another pattern of aldolase isoenzymes in tumorous tissue was stated (AC) comparing with control group (arrangement AC, BC). The changes referred also to the activity of both examined enzymes in tumorous tissue and in the tissue of testis not being tumorously changed. The authors suggest the existence of a factor with depressor characteristics, which is produced by neoplasm and changes the function of genes that are responsible for synthesis examined isoenzymes in the tissue not being tumorously changed.
The authors have demonstrated the statistically significant decrease of aldolase and pyruvate kinase activity in erythrocytes in a group of men with hypogonadism and with a significant defect of gonad endocrinologic function. A similar phenomenon appeared in erythrocytes in a group of sexually mature rabbits after castration. In the course of substitutional treatment with testosterone esters given intramuscularly, the increase of activity characteristic for both examined glycolytic enzymes was observed in both experimental groups, i.e. the patients and the animals. The pyruvate kinase increase, however, was not statistically significant in the examined group of men, which resulted from the small size of the group. The authors suggest the existence of testosterone influence on regular activity of bone marrow glycolytic enzymes, and consequently on the mature erythrocytes. The decrease of aldolase and pyruvate kinase activity in erythrocytes of patients with male hypogonadism may decrease the erythrocytes valence. This ist another biochemical argument for the need of conducting long-term substitutional therapy in cases of male hypogonadism.
Adrenal function was studied in patients with chronic myeloid leukaemia treated in the past or presently with Busulphan. Adrenocortical function was determined by means of 24-hour profile of 11-hydroxysteroids (11-OHCS) in plasma, and urinary 24-hour 17 hydrocorticosteroids (17-OHCS) and 17-ketosteroids (17-KS). The adrenomedullary function was determined measuring VMA level in 24-hour urine. In most patients normal 24-hour 11-OHCS profiles and 24-levels of 17-OHCS, 17-KS and VMA. Only in some cases these levels were raised. This rise was observed more frequently in patients with blastic crisis of myeloid leukaemia which may indicate that the adrenal reserve was maintained in these patients. The investigations failed to show that the disease itself or Busulphan treatment impaired adrenal function. The possibility of a direct effect of Busulphan on melanin metabolism in the organism is discussed.
