Dynamics of serum markers in the different course of hepatitis B virus (HBV) infection.

The presence of serum markers of hepatitis B virus (HBV) infection in different stage of illness has a specific pathogenic and diagnostic significance. Based on the frequency of the appearance of HBV markers in patients' sera at different stages of the illness, we attempted a grouping possibly helpful for differentiation, epidemiologic and prognostic evaluation of hepatitis. The significance for chronic disease development of the dynamics of HBV marker levels in the serum is discussed.

Effect of human leukocyte interferon on HBsAg production in PLC/PRF/5 human hepatoma cell line.

We describe the effect of the natural human interferon alfa (HuIFN alfa) on HBsAg production in PLC/PRF/5 cells. Decreased HBsAg production was regularly demonstrated 48 hr after the treatment of cells with 3-300 IU of HuIFN alfa/ml; it lasted for further 24-48 hr depending from the amount of interferon (IFN) added. We found no differences in the sensitivity to HuIFN alfa of non-confluent cells as compared to cells which had reached confluency. The decrease of the extracellular HBsAg excretion seems to be due to inhibition of intracellular HBsAg formation.

Effect of influenza virus on the interaction of cytoskeleton with cell membranes.

The effect of influenza virus on the organization of cytoskeleton (CS) associated with cell surface plasma membranes (PM) of chick embryo cells (CEC) was investigated. The CS isolated from virus-treated PM contained more proteins and more actin than the CS isolated from control membranes. Virus particles were found to be associated with this CS, although the purified virus alone, treated in the same manner, did not sediment at low speed in the residue insoluble in Triton X-100. Autophosphorylation of CS, especially of the polypeptides 24 K, 45 K, 65 K and 105-110 K, was increased in CS prepared from virus-treated membranes. Superprecipitation of the CS prepared from CEC was stimulated by addition of the virus. Electron microscopy of the Triton X-100 insoluble residue from virus-treated PM showed more structures similar to microfilament bundles than Triton X-100 insoluble residue from control membranes. Based on these results we suggest that already in early stages the virus infection leads to a reorganization of membrane-associated CS.

Effect of influenza virus on protein phosphorylation in isolated membranes of chick embryo cells.

Membranes isolated from chick embryo cells (CEC) were found to contain an endogenous protein kinase that phosphorylated endogenous proteins. 32P incorporation into membrane proteins was analysed by sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE) and detected by autoradiography. Membrane phosphorylation in the presence of physiological saline (PM-phys) and in the presence of influenza virus (PM-V) were compared. Under short-time incubation (less than 1 min) with gamma-32P ATP there was practically no difference between PM-phys and PM-V in 32P incorporation. Under prolonged incubation (3-15 min), gradual dephosphorylation of the phosphoprotein moving in SDS-PAGE in the zone of relative molecular mass (Mr) of 60 kDa (phosphoprotein P60) was observed in PM-phys whereas in PM-V, phosphorylation of P60 increased with the time of incubation with gamma-32P ATP. Dephosphorylation of P60 in PM-phys was inhibited by 2.5 mmol/l EGTA as well as by 100 mumol/l chlorpromazine (CPZ) and stimulated by calmodulin (CaM) in the presence of Ca2+. Responsible for the dephosphorylation was probably the endogenous Ca2+ and/or CaM-dependent membrane protein phosphatase. It was inhibited by influenza virus (even in the presence of Ca2+ and CaM). The possible role of P60 phosphorylation in the first step of virus infection is discussed.

Hepatitis B and herpes viral components in the cerebrospinal fluid of chronic schizophrenic and senile demented patients.

Cerebrospinal fluids (CSF) or sera or both from 57 chronic schizophrenics and 18 senile demented patients were examined by various tests for HBsAg. both CSF and serum were positive in 2 schizophrenics while in six HBsAg was detected in the CSF only. Elevated values of circulating immune complexes were found in positive patients. Most CSF positive for HBsAg also contained neutralizing antibodies to Herpesvirus hominis type 1. Ultramicroscopic structures similar to hepatitis B virus (HBV) components and herpesviral particles were visualized in the CSF of one patient on electron microscope (EM) grids coated with anti-human IgG serum. Most HBsAg positive patients appeared to be liver-symptoms-free carriers. In the CSF of a 79 years old senile demented man HBsAg was proved serologically. Several herpesviral particles complexed with globular material and spherical structures for 15 to 25 nm in diameter were visualized in the same CSF on EM grids coated with anti-human IgG serum. The findings support the importance of herpesviruses in mental illness. Penetration of HBsAg through the blood-brian barrier might be involved as an iatrogenic factor in the course of late psychoses.

Appearance and some unusual features of Epstein-Barr virus antibody production in infectious mononucleosis and other health disorders.

The indirect immunofluorescence (IF) test and the complement-fixation reaction (CFR) were used in examination of over 1500 sera obtained from patients with infectious mononucleosis (IM) and other health disorders. The evidence obtained supports a direct aetiological relationship between Epstein-Barr virus (EBV) and IM and points on a relationship of EBV to some other lymphadenopathies and health disorders. The incidence of the IgG type antibody against virus capsid antigen (EB-VCA) and soluble antigen (CF-SA) obtained from EBV genome-positive cells among different age groups of patients is described along with results of long-term examinations of serum samples from IM patients. The appearance and dynamics of production of both types of EBV antibody and their persistence in the organism varied. Long-lasting oscillations, in particular of the EB-VCA antibody levels were found in sera of patients with prolonged health disorders following IM. The diagnosis value of the IF test and the CFR is discussed.

Psychopharmaca and electroconvulsive therapy in relation to viral antibodies and interferon. Experimental and clinical study.

Chlorpromazine (CHP) in a concentration of 8.3 microgram per ml medium inhibited attachment and multiplication of CEC and of various cell lines. The same CHP concentration applied to grown CEC monolayers inhibited the growth of TBEV but not the growth of HHV 1, while interferon was slightly stimulated. The clinical course of fatal subcutaneous TBEV infection in mice was not affected by daily CHP treatment while brain and serum interferon were stimulated. Antibody formation in TBEV infected or HHV 1 immunized mice was not affected significantly by CHP application. The following data were obtained in a longitudinal study on 28 hospitalized schizophrenic patients: 89 percent of them revealed at least some serological findings which indicate an actual HHV 1 infectious process-elevated NAB titers, high ratio between complement requiring and complement not requiring NAB titers. A low interferon activity was observed in serum and cerebrospinal fluid of 93 percent and 14 percent patients, respectively. No regular correlation between HHV 1-NAB-both complement requiring and complement not requiring-and of interferon on one side and between hospitalization. CHP treatment and electroconvulsive therapy on the other side has been observed.

Purification of mouse cell interferon induced by isotopically labelled double-stranded f2 phage RNA.

The dynamics of interferon formation by an established cell line of mouse fibroblast (L cells) and by mouse peritoneal leukocytes induced by double-stranded RNA extracted from E. coli f2 phage is described. The L cells produced interferon at a lower rate, the maximum values were obtained at 12 to 20 hours after induction, and the production was ultimately dependent on the established cell line used and on the presence of DEAE-dextran during induction. The mouse peritoneal leukocytes (MPL), on the other hand, did not require DEAE-dextran and the maximum of interferon production was reached between 6 and 12 hours after induction. Both the L cell- and the MPL-interferons were purified and concentrated so that the final specific biologic activity was 100-to 300-fold higher than that of the initial preparations (1 to 5 X 10(6) interferon units per mg protein). Polyacrylamide gel electrophoresis showed similar migration profiles for the preparations of both interferons. The smaller part of the activity was situated in a broader, slow-moving peak and the greater part formed a sharp, high and fast-moving peak. Using 3H uridine-labelled f2 ds-RNA for induction of interferon it was found that one of the radioactivity zones coincided with the fast-moving activity peak of the purified and concentrated interferon.

Morphological, cytological and biological observations on viruses isolated from patients with subacute thyroiditis de Quervain.

New data on viruses isolated from patients with subacute thyroiditis de Quervain are reported. Characteristic morphological, cytological, some physico-chemical and biological features of the isolated viruses are described. A possible role of these viruses in human and animal health disorders is discussed. The isolated viruses remain unclassified so far.

Isolation and some serological and epidemiological data on the viruses recovered from patients with subacute thyroiditis de Quervain.

Virological and serological methods were used in examination of 28 patients suffering from subacute thyroiditis de Quervain. Attempts to isolate a presumed viral agent from 8 patients were performed by inoculation of serum, urine, and aspiration biopsies of thyroid glands taken at different stages of the illness, into tissue cultures of different types of human and animal cells. Recovery of a cytopathic viral agent on cells of a rabbit lung continuous line was successful in 5 cases. Serological cross reactions exist between the isolated viruses and patient serum but not with serum of healthy people. Cases with the acquired illness and positive antibodies against the isolated viruses who had been in close and prolonged contact with patients suffering from subacute thyroiditis de Quervain were also investigated.
