Quantity of nucleolar silver-stained proteins is related to proliferating activity in cancer cells.

The relationship between the silver-stained proteins of interphase nucleolar organizer regions (NORs) and proliferative activity was studied in 12 established cell lines derived from different tumor types. Quantitative evaluation of NOR proteins was carried out on silver-stained cells, by means of an automated image analyzer and the cell proliferation rate was measured with [3H] thymidine labeling. A linear relationship was found between cell duplication activity and the amount of silver-stained NOR proteins independently of the type of cancer cells (r = 0.94, p less than 0.001). Measurement of silver-stained NOR protein could thus be used as a simple estimate of cancer cell proliferation rate.

The silver-stained proteins of interphasic nucleolar organizer regions as a parameter of cell duplication rate.

The relationship between interphasic silver-stained proteins of the nucleolar organizer regions (Ag-NOR proteins) and cell replication rate has been studied in 13 established neuroblastoma cell lines. The quantity of Ag-NOR proteins was measured in silver-stained cells by means of an automated image analyzer. The results indicated that the amount of Ag-NOR proteins is strictly proportional to the proliferative activity of the cells. Cell lines with a difference of only 4 h in doubling time were characterized by a statistically significant difference in Ag-NOR protein amount. The Ag-NOR protein quantity can therefore be used as a parameter for evaluating the cell proliferation rate.

Relationship between interphasic nucleolar organizer regions and growth rate in two neuroblastoma cell lines.

The relationship between the quantity of silver-stained interphasic nucleolar organizer regions (NORs) and nuclear synthetic activity, caryotype, and growth rate was studied in two established neuroblastoma cell lines (CHP 212 and HTB 10). Statistical analysis of silver-stained NORs revealed four times as many in CHP 212 cells compared with HTB 10 cells. No difference was observed in the ribosomal RNA synthesis between the two cell lines. The caryotype index was 1.2 for CHP 212 and 1.0 for HTB 10 cells. The number of chromosomes carrying NORs and the quantity of ribosomal genes was found to be the same for the two cell lines. Doubling time of CHP 212 cells was 20 hours compared with 54 hours for HTB 10 cells. In CHP 212 cells bindering of cell duplication by serum deprivation induced a progressive lowering (calculated at 48, 72, and 96 hours) of the quantity of silver-stained interphasic NORs. Recovery of duplication by new serum addition induced, after 24 hours, an increase of the quantity of silver-stained interphasic NORs up to control levels. In the light of available data, these results indicate that the quantity of interphasic NORs is strictly correlated only to the growth rate of the cell.

Monomeric and polymeric forms of alpha-1 antichymotrypsin in sera from patients with probable late onset Alzheimer's disease.

Patients with probable late-onset Alzheimer's disease (l-AD) had higher levels of serum alpha 1-antichymotrypsin (alpha 1-ACT) than those found in patients with vascular dementia (VD) and healthy elderly controls, when assessed by a competitive enzyme-linked immune assay. Serum alpha 1-ACT was also characterized by SDS PAGE electrophoresis. Western blot and computer-assisted optical density reading (OD). Using a polyclonal affinity-purified antibody specific for human alpha 1-ACT, one band with the apparent MW of 60 and another with 180 kD in sera from all subjects were clearly detectable. OD of both alpha 1-ACT bands from patients with l-AD was higher than that from VD patients, the 180-kD form being 2.65 times higher than that observed from patients with VD. Serum levels of other acute phase proteins from l-AD were comparable to those observed in VD patients. A slight but nonstatistical increment of serum IL-6 was noted in patients with l-AD. Serum alpha 1-ACT was purified from 3 of these l-AD patients by a two-step affinity chromatography technique. After Western blot, purified alpha 1-ACT showed two or three different bands which immune-reacted with an antibody specific for alpha 1-ACT. The apparent MWs were 60, 120 and 180 kD. In human sera the serpin was present mainly in a monomeric form, but it could also form SDS stable dimers and trimers. Both monomeric and SDS stable polymeric forms of alpha 1-ACT appeared to be increased in sera from patients with l-AD.

Diagnostic value of silver-stained interphasic nucleolar organizer regions in breast tumors.

Seventy-six specimens of normal breast tissue and benign and malignant breast lesions were studied to assess the mean area occupied by silver-stained proteins of the nucleolar organizer regions (MNORA) of the nucleolus. The assessment was performed with a computer-assisted image analyzer. The results indicate that only 30% of malignant lesions have a MNORA value greater than that of normal breast tissue or benign lesions. On the other hand, MNORA values of ductal carcinoma in situ were significantly greater than those of epitheliosis (papillomatosis). MNORA values were also significantly different in grade I and grade III invasive ductal carcinomas, the latter exhibiting the highest MNORA values of all the cases observed. Evaluation of MNORA values may therefore help in differentiating benign epithelial proliferations from ductal carcinomas in situ. Furthermore, because there is evidence that MNORA values are indicative of the cell duplication rate, MNORA values may ultimately be considered an objective prognostic parameter in addition to grading for invasive ductal carcinomas.
