OBJECTIVE: To study the effect of mitogen activated protein kinase (MAPK) on the proliferation of human embryonic lung fibroblast (HELF) induced by 60Co irradiation and the relationship between MAPK and angiotensin II (A II). METHODS: Cell proliferation was measured with enzyme-labeling method; angiotensin II, MAPK and type I precollagen synthesis as well as the role of sodium nitroprusside on the inhibition of A II were evaluated adopting immunohistochemical technique combined with image analysis after 1-5 Gy 60Co gamma-ray irradiation on the cultured HELF. RESULTS: Irradiation at 1-5 Gy promoted cell proliferation and type I precollagen synthesis; the syntheses of A II and MAPK were also increased in the irradiated cells. Exogenous A II enhanced cell proliferation, but sodium nitroprusside inhibited the action of A II. CONCLUSIONS: HELF proliferation induced by 60Co irradiation is a chain reactive course, in which, A II and MAPK are involved. MAPK may play a role of limiting valve in the signal transduction of cellular proliferation.
OBJECTIVE: To investigate the relationship between tumor angiogenesis and clinical pathology, as wel as the prognosis in osteosarcoma. METHODS: Intratumoral microvessel density (MVD) in 70 cases of osteosarcoma was assessed immunohistochemically by using the specific endothelial cell markers F VIII-RA and CD31. RESULTS: There was no correlation between the MVD and the tumor size, Price's grade, as well as Dahlin's classification; however, significant correlation was found between the MVD and the neoplastic osteoid classification, new WHO classification in 1993, as well as the proliferating cell nuclear antigen (PCNA) labelling index. Microvessel counts were associated with overall survival by Kaplan-Meier analysis. An average vessal count of less than 31 (x 200) suggested a better survival, but a higher vessel count of more than 31 (x 200) showed a trand to worse the overall survival. CONCLUSION: The results suggest a significant relationship between MVD and prognosis; moreover, MVD may be a useful prognostic indicator in osteosarcomas.
For patients with chest cancer who are in advanced stage, aged, or/and with respiratory insufficiency, the key to the success of open-chest surgery is intensive management of the respiratory tract and proper treatment of complications in perioperative period. From May 1984 to November 1995, 1661 patients with esophageal and lung cancer underwent thoracotomy in our hospital. 198 of the 1661 patients fit in with the standards of advanced, aged or respiratory insufficiency. The patients were aged over 70 years, with the length of esophageal cancer over 10 cm and, the MVV less than 50% (least 24.3%). The results the patients were satisfactory because a series of the effective measures of perioperative management were tahen. They incladed thyrocrico-puncture and use of a fine tube through which drugs were injected to irritate cough sputum out; examination and suction with bronchofiberoscope; tracheo-bronchial lavage through bronchofiberoscope with antibiotics solution; and tracheotomy using ventilator appropriately. Perioperative intensive management is very important for extending operation indication and tiding over critical period.
The purpose of this study is to observe the macro, micro and ultrastructural changes of the pituitary, adrenal, pancreas and spleen in the experimental primary brain death. The results showed: 1. acinar zymogen granule of pancreas decreased, karyopyknosis of islet cells formed. 2. dilation and blood stasis of spleen sinus happened 3. adrenocortical karyopyknosis formed, mitochondrial crista vanish. 4. Pituitary karyopyknosis and cytolysis developed. 5. there was correlation between the extent of lesions and time after brain death.
The aim of this study is to characterize the pattern of release of soluble vascular cell adhesion molecule-1 (sVCAM-1) in patients with acute cerebral infarction. The level of serum sVCAM-1 was measured serially with two-layer antibody sandwich enzyme-linked immunosorbent assay (ELISA) in 89 patients with acute cerebral infarction, 43 patients with cerebral haemorrhage and 30 normal controls. It is shown that the level of serum sVCAM-1 in patients with cerebral infarction 24 hours after the onset was higher than that in patients with cerebral haemorrhage and normal controls (P < 0.01). The level of serum sVCAM-1 was significantly higher in the patients with large cerebral infarction than that in patients with medium and small cerebral infarctions (P < 0.01). The level of serum sVCAM-1 in patients with cerebral infarction increased from 24 hours to 7 days and decreased from 7 days to 14 days, but it was still higher than that in patients with cerebral haemorrhage and normal controls on the 14th day. The level of serum sVCAM-1 was higher in patients with infection as compared with those without. These findings suggested that sVCAM-1 is closely related to the development of acute cerebral infarction. It is essential to have further study on the change of serumsVCAM-1 in cerebral infarction.
By measurement and analysis, the postoperative image was simulated in standard facial image of the saddle nose using computer imaging and the software developed by us. When the simulated image is accepted by both the surgeon and the patient, the computer gives data to instruct the operation. The authors described how to obtain the standard facial image and how to reduce error. The computer imaging simulation makes the cosmetic operation accurate, quantitative and imaged, and its applications will be broadened.
The aim of this study was to investigate the regulation and clinical significance of serum acute phase proteins. The patients were divided into three groups: group I consisted of 11 patients with less than 20% of TBSA; group II 10 patients with 20-50% of TBSA and group III 11 patients with more than 50% of TBSA. Blood samples were taken at 8, 24, 48 and 72 h postburn, and in group III additional blood samples were taken on the 7th, 14th, 21st day or in septic episode. The concentration of CRP and C3, Tf, PA were detected by rate nephelometry, agar immunodiffusion and rocket immunoelectrophoresis respectively. Compared with controls, the peak value of CRP was observed during 48-72 h (P < 0.01), C3 decreased at 8 h and lowest at 24 h. Tf was lowered at 48 h, PA and Tf shared the similar changes. However, all four variables had no marked difference during infection and sepsis. The results showed no correlation between C3 and CRP (r = 0.0885, P > 0.05). The results suggested that: 1. CRP mainly reflected the extent of injury but not sepsis; 2. increased CRP was not the causative factor of decreased C3. Tf reduction accelerated bacterial growth.
OBJECTIVE: A model in neonatal rats was established to study pathophysiology of perinatal hypoxic-ischemic brain damage. METHODS: 7-9 day old Wistar rats were subjected to unilateral carotied artery ligation followed by hypoxic state for 3 hours (10% O2 + 90% N2, at 37 degrees C). Magnesium sulfate (0.5 mg/gBW) and ligustrazin (0.1 mg/gBW) was separately given a 30 min before hypoxia. Superoxide dismutase (SOD) and malonic dialdehyde (MDA) in the cerebral cortex and serum MDA were measured immediately after hypoxia. Neuropathologic examination was made in 4 weeks after hypoxia. RESULTS: It was found that SOD and MDA in the cerebral cortex and serum MDA in the hypoxicischemic group were significantly increased in comparision with the normal control group (P < 0.01), and that all these parameters either in the magnesium sulfate group or in the ligustrazin group were lower than in the hypoxic-ischemic group (P < 0.05). The hypoxic-ischemic group showed that there were neuronal degenerations in the gray matter, hippocampus and cerebellum which were reduced in the two treatment groups. CONCLUSION: Our results indicate that oxide free radical formation is one of the pathogenic factors of perinatal hypoxic-ischemic brain damage and magnesium sulfate and ligustrazin reduce hypoxicischemic brain damage due to indirect anti-oxidation.
Among total 67 postoperative complications resulted from osteotomy of vertebral arch and body in 439 cases of kyphosis, massive bleeding accounted for 2 cases (0.46%); spondylolisthesis, 6 (1.37%); paraplegia caused by compressing of the spinal cord, 5(1.14%); root pain, 6(1.37%); loss of correction, 26(29.55%); leaking and subcutaneous accumulation of CST, 5(1.14%); incision necrosis, 16(3.64%); lifting of luque rod with peneatration through the skin, 1(0.23%). Causes, treatment and prevention of these complications have been discussed in this study. We emphasize that the operation should be done after the stable of ankylosing spondylcitis. The degree of correction by osteotomy at one-level should be limited.
To study histologically the biological behaviour of the grafts and the reactions in the receipt area after reconstruction of temporomandibular joint (TMJ), we have reconstructed experimentally removed TMJs on purebred juvenile pigs by transplanting autogenous costo-chondral grafts. The findings of continuously survey of histologic changes in the receipt areas and the grafts showed that autogenous grafts healed well with the surrounding soft tissues, and induced the mesenchymal-like cells from the receipt region, rather than the periosteal cells which might be left, to differentiate and ossify both within and at the surface of them, and they themselves were gradually replaced by the new-formed bone; and that active osteogenesis could also be seen within the thick tissues far from the graft, which could not be the result of graft's induction. It may be concluded that the new bone, both from the induction of the graft which acts as a framework and somehow from the outside tissues, reconstructs the new condyle and determines the postsurgical growth of the mandible.
Trypan blue and Indian ink made in Fuzhou as vital staining dyes were injected into the abdominal cavities of rabbits. Nine days later, the tonsil, thymus, spleen and lymph nodes were examined. Numerous pigment granules were observed in the macrophages of the spleen and lymph node. However, no phagocytosis of pigment granules was found in the palatine tonsil and thymus. It indicated that the function of palatine tonsil as a lymph organ is to contact exogenous antigens and to take part in immune response. It is important for children, especially infants, to establish a complete immune mechanism. The Tonsils reach their maximum development in childhood and thereafter decline. Once the palatine tonsils become chronic foci, surgical removal of the diseased tonsils is necessary.
To investigate the possibility of regeneration and functional restoration of vestibular hair cells after toxic destruction, gentamicin (150 mg/kg per day) was given to guinea pigs subcutaneously for 10 days to induce damage of vestibular hair cells. The control animals received the same volume of saline. At 2, 4, 12 and 24 weeks after the treatment, animals were sacrificed and their vestibular organs prepared for scanning electronic microscopic study. Change of the hair cell density was studied using quantitative analysis techniques. Electronystagmogram, induced with ice water, was taken before and at 2, 4, 12, and 24 weeks after the treatment. Immature stereocillia bundles were noticed in regions with heavy hair cell loss in the utricle and crista ampullaris 2 weeks after treatment and were still visible 24 weeks after the treatment. Hair cell density (number of hair cells/100 microns basilar membrane length) decreased significantly at 12 weeks after treatment as compared with the control (P < 0.01), and showed partial recovery at 24 weeks after treatment as compared with 12 weeks after treatment (P < 0.05) which was still below normall level (P < 0.05). Vmean showed significant reduction at 4 and 12 weeks after treatment (P < 0.01), and recovery at 24 weeks (P < 0.05), which was still below the normal level (P < 0.01). SEM results indicated the potential of repair of vestibular hair cells in guinea pigs after damage which may occur 24 weeks after the damage. Quantitative analysis of hair cell density further suggests that regeneration is the mechanism of repair. The regenerated hair cells seen in this study may play a role in the restoration of vestibular function.
OBJECTIVE: To evaluate enhanced CT examination in the differential diagnosis of local tumor recurrence and postradiation fibrosis when a mass appeared is observed after radiation therapy of carcinoma of nasopharynx. METHODS: 159 cases of nasopharyngeal carcinoma were examined by CT, including 37 cases of local recurrence(RT) after radiation therapy and 54 cases of postradiation fibrosis (RF) 68 cases of nasopharyngeal carcinoma (PT) before radiation therapy sened as comparison. Elscint sprint 2000 CT machine was used. The CT number of plain and enhanced examination of the mass appearing after radiation therapy were measured. Variance analysis was used as statistical management for CT number difference of different kinds of mass lesions before and after contrast enhancement. RESULTS: In the RT cases, the average CT number of the mass lesion for plain scan were 50.40 +/- 8.10 Hu, and for enhanced scan it was 73.74 +/- 7.37 Hu. In the RF cases plain scan 48.62 +/- 10.15 Hu, enhanced scan 56.74 +/- 8.69 Hu, In the control cases (PT), the CT number in the plain scan of lesion was 48.32 +/- 9.20 Hu, and 72.38 +/- 7.75 Hu in the enhanced examination. These measurement showed obvious enhancement of recurrence tumor. The CT number of contrast scan of RT and RF was obviousy different (P < 0.01). CONCLUSION: If a mass lesion appears after radiation therapy in carcinoma of nasopharyx, enhancement of the lesion in CT scan is an important criterion for differential diagnosis.
Two compounds were isolated from the leaves and stems of Trigonella foenum-graecum, and on the basis of spectral analysis, their structures were elucidated as gamma-schizandrin and scopoletin. They were isolated from T. foenum-grecum for the first time.
A malignant transformed mammary epithelial cell line (11A1) was transfected with liposome encapsulated eukaryotic expression plasmid pCMV-neo-RB, yielding 4 constant clones which have obvious phenotypic reversion changes, and named 11A1-R1-R4 respectively. Further experiments showed that the 11A1-R1 behaved like normal epithelial cells in both morphological and biological characteristics, with decreased clonogenicity in solid argar medium as well as decreased tumorigenicity. Northern blot hybridization showed increased expression of RB gene and decreased expression of c-myc gene in 11A1-R1, 11A1-R2 cells compared to 11A1 cells. This was an ideal phenotypic reversion model for epithelial transformed cell line and demonstrated that the RB gene can reexpress and suppress malignant phenotype in RB inactive cells.
