[Surface photovoltaic spectroscopic investigation of Bacteriorhodopsin (BR)].

The surface photovoltaic spectra (SPS) of Bacteriorhodopsin-based films were systematically measured. The results show that the BR films are characterized by the specific photovoltaic effect. The SPS response of the BR films depends on the fabrication and humidity of the film. The intermediate M412 and O640 of BR are observed in the SPS of HEPES-containing BR film, indicating that HEPES has very important function to stabilize intermediate M412. The observations are explained in terms of the photocycle of BR and the mechanism of photovoltage generation.

Role of alpha 5 beta 1 integrin in determining malignant properties of colon carcinoma cells.

We characterized the expression of alpha 5 beta 1 integrin in two distinct phenotypes of colon carcinoma cell lines. Highly invasive colon cell lines (designated Group I cell lines) expressed higher levels of integrin alpha 5 beta 1 mRNA and protein than did poorly invasive colon cell lines (designated Group III cell lines). The relatively high expression of integrin alpha 5 beta 1 in Group I cell lines resulted in strong enhancement of cell adhesion to fibronectin (FN) tissue culture plates, whereas Group III cell lines showed little or no enhancement of cell adhesion by coating. There was no significant difference between Group I and Group III cell lines with respect to cell adhesion to laminin and collagen IV. Cell adhesion to FN in Group I cells was mainly mediated by integrin alpha 5 beta 1 because a monoclonal anti-alpha 5 subunit antibody could block cell adhesion to FN, whereas anti-alpha 2 and anti-alpha 3 antibodies had no effect on cell adhesion to FN. The divergence of alpha 5 beta 1 expression in these two distinct colon carcinoma phenotypes suggested that high expression of alpha 5 beta 1 might contribute to malignant progression in this model system. To test this hypothesis, GEO cells, a Group III cell line that did not express alpha 5 integrin, were transfected with the alpha 5 subunit. Stable transfection of alpha 5 sense cDNA into a typical GEO-limiting dilution clone led to the expression of alpha 5 subunit mRNA and cell surface alpha 5 beta 1 protein. The alpha 5 sense transfectants showed enhanced attachment to FN-coated plates and were more tumorigenic when the cells were injected into athymic nude mice. These results indicate that inappropriately high alpha 5 beta 1 integrin expression contributes to malignant progression in colon carcinoma.

HLA class II antigens and T lymphocytes in human nasal epithelial cells. Modulation of the HLA class II gene transcripts by gamma interferon.

BACKGROUND: Nasal polyps are characterized by a proliferation of the epithelial layer of the mucosa, cellular infiltrates and other pathological changes; however the mechanisms involved in polyp pathogenesis remain largely unclear. OBJECTIVES: We have taken two different approaches to study the cellular events involved in nasal polyposis. METHODS: First, through use of immunohistochemical methods, we have studied the expression of HLA class II antigens in epithelial cells of nasal polyps and the distribution of lymphocytes in the epithelium and in the subepithelial layer in patients with clinical conditions, such as asthma, atopy, aspirin intolerance or cystic fibrosis, and in subjects with an absence of concomitant diseases. Second, in order to investigate whether HLA class II expression is controlled at the pre- or post-transcriptional level, we studied the effect of interferon gamma (INF gamma) on epithelial cells in primary culture, which were derived from HLA class II negative and HLA class II positive nasal polyps. Total RNA was extracted from the cells and reverse-transcribed, and the c-DNA corresponding to DR, DP, DQ loci was amplified by PCR. RESULTS: Expression of HLA class II antigens by the epithelia of nasal polyps was more common in the presence rather than in the absence of concomitant asthma, atopy or cystic fibrosis (59% versus 40%). HLA-DR was the only HLA class II antigen expressed in the seven polyps taken from cystic fibrosis patients. The number of CD8+ cells was significantly higher in polyps associated with known clinical conditions and HLA class II antigen expression than it was in 'isolated' polyps and in HLA class II negative polyps. RNA transcripts for at least one or all three HLA-DR, DP and DQ antigens were detected in 10 cultures of the II HLA class II positive polyps. Conversely, 8 of 10 cultures derived from HLA class II negative polyps did not express HLA class II transcripts in the absence of INF gamma. Adding INF gamma (100 U/ml) to the latter cell cultures caused expression of transcripts of one or more HLA class II genes. CONCLUSIONS: We have shown that HLA class II antigens were more frequently detected in polyps of patients with an identified clinical syndrome than in those of asymptomatic subjects. Our results also suggest that IFN gamma regulates expression of HLA class II antigens in airway epithelial cells of the nasal polyps at the transcriptional level, and that cultured cells from nasal polyps represent a suitable model to investigate immune mechanisms involved in diseases such as atopy, asthma and cystic fibrosis.

Cellular ras gene activity is required for full neoplastic transformation by the large tumor antigen of SV40.

To investigate the role of the cellular ras gene product in neoplastic transformation by the SV40 large tumor antigen (SVLT), murine C3H10T1/2 cells were rendered deficient in Ras activity by transfection with inducible or constitutive antisense ras gene constructs or through the introduction of the dominant-negative mutant, ras(asn17). Consistent with previous results, SVLT-induced morphological transformation was unaffected by the down-regulation of c-ras gene product activity. On the other hand, colony formation in soft agar and tumorigenicity in nude mice were drastically reduced in c-Ras-deficient cells. In addition, SVLT expression in C3H10T1/2 cells led to increased c-Ras activity, as determined by an increase in the Ras-bound GTP/GTP + GDP ratio. These results suggest that c-Ras is required for full neoplastic transformation by SVLT.

Dibutyryl cAMP prevents increased vascular permeability caused by air embolism in isolated rat lungs.

BACKGROUND AND HYPOTHESIS: Venous gas bubbles are routinely detected in astronauts undergoing extravehicular activities at lower suit pressure. Venous air embolism increases the pulmonary arterial pressure and the vascular permeability leading to acute lung injury. In the present study we evaluated the protective effect of dibutyryl cAMP, aminophylline, and pentoxifylline on the pulmonary vasculatures after air embolism. METHOD: In isolated and perfused rat lungs, we induced air embolism by introducing air bubbles into the pulmonary artery. We measured the pulmonary arterial pressures and capillary pressure. Vascular permeability was determined by measuring the filtration coefficient (Kf) and the protein concentration in the lung lavage fluid. RESULTS: Air infusion caused pulmonary hypertension and increased vascular permeability, resulting in pulmonary edema. The Kf (in g.min-1.cm H2O-1.100 g-1) increased from 0.44 +/- 0.05 at baseline to 2.98 +/- 0.47 after air infusion. Pretreatment with DBcAMP prevented the increase in Kf (0.63 +/- 0.09) caused by air embolism without altering the hemodynamics. Aminophylline and pentoxifylline did not prevent lung injury induced by air embolism. Although aminophylline did not alter the response of pulmonary arterial pressure to air infusion, it elevated the capillary pressure to 5.1 +/- 0.4 mmHg, which was significantly greater than that in the lung receiving air infusion alone. CONCLUSION: Our results suggest that DBcAMP prevents the increase in vascular permeability caused air embolism. The ineffectiveness of aminophylline and pentoxifylline on the prevention of air embolism-induced lung injury remains for further investigation.

[Clinical study on reduction of postpartum bleeding by methyl carprost suppository].

OBJECTIVE: To observe the effects of methyl carprost suppository on postpartum bleeding in normal parturients. METHODS: Three hundred and sixty normal parturients were randomized into study group (n = 260) and the control group (n = 100). Parturients in study group were administered methyl carprost suppository tents either vaginally (n = 130) or via the rectum (n = 130). The controls were given 10IU oxytocin immediately after the delivery of anterior shoulder of the fetus either intramuscularly (n = 50) or intravenously (n = 50). The amount of bleeding with in 2 hours postpartum was measured. RESULTS: The mean amount of bleeding for parturients who received vaginal or rectal methyl carprost was 146.4 ml and 134.2 ml, respectively. In contrast, the mean amount of bleeding for parturients who received oxytocin intramuscularly or intravenously was 267.9 ml and 210.6 ml, respectively. The differences in the amount of bleeding between the two groups were statistically significant (P < 0.01), whereas differences between the routes of drug administration within each group were not significant (P > 0.05). CONCLUSION: Methyl carprost suppository is more effective than oxytocin in controlling postpartum bleeding. Drug administration through the rectum is easy to perform and, therefore, suitable for use in rural areas.

[Distribution and significance of endothelin-1 and endothelin receptor a mRNA in liver after endotoxemia].

OBJECTIVE: To study the distribution and significance of endothelin-1 (ET-1) and endothelin receptor A (ETAR) mRNA in liver after endotoxemia using situ hybridization. METHODS: 30 rats were randomized into control and endotoxin groups. The endotoxin group was injected with endotoxin at a dose of 10 mg/kg body wt. Hybridization of ET-1 and ETAR mRNA of the hepatic tissue was proceeded at 3, 6, 12 and 24 hours after endotoxin administration. RESULTS: By in situ hybridization, the cell expression of ET-1 mRNA was hepatic sinusoids, the endothelial cells of portal vein, and Kupffer cell and the cell expression of ETAR mRNA was the small artery smooth muscle cells of hepatic lobule and the hepatic sinusoidal lining cells. The accumulation of grain was observed in many cells of the liver. The number of accumulating grain increased in unit scope. CONCLUSION: ET-1 is synthesized by the hepatic sinusoids, hepatic vascular endothelial cells, and Kupffer cells, ETAR may be located on the hepatic vascular smooth muscle cells and Ito cells (fat-storing cells) around sinusoid. Endotoxin may, on transcription and translation level, lead to the increase of ET-1 and ETAR in synthesis and release.

[Clinical observation on Doula delivery].

OBJECTIVE: To improve the obstetricare quality during labour and delivery, our hospital carried out Doula delivery which is an appropriate technology in promoting natural delivery reported by Dr M Klaus, USA. METHODS: Mothers were accompanied by senior experienced midwife who afforded continuous physical, psychological and emotional support during the whole course of labour. From April to August 1996, 46 cases of primigravida were selected as study group. Another 336 primigravida given birth in the same period were taken as controls, who were accompanied by their family member during their active phase of labour. RESULTS: The results showed that the cesarean section rate was 6.5% in Doula group while 20.8% in the control group. The difference was significant (P < 0.05). The duration of labour and volume of postpartum bleeding both declined obviously in the study group (P < 0. 05). CONCLUSION: Doula delivery offered better quality of health care during labour and decreased cesarean section rate duration of labour and postpartum bleeding.

Expression and cellular location of endothelin-1 mRNA in rat liver following endotoxemia.

OBJECTIVE: To study the effect of endotoxin on the alteration of the transcription, expression and cellular location of endothelin-1 (ET-1) mRNA in the hepatic tissue. METHODS: Wistar rats were divided into control and endotoxic group. The rats in the control group were injected with saline, and those in the endotoxic group with endotoxin at a dose of 10 mg.kg-1 body wt. ET-1 hepatic homogenate was assayed by radioimmunoassay at 3, 6, 9, 12 and 24 h after endotoxin administration. Dot blot was used to identify and quantify ET-1 mRNA of the hepatic tissue. Hybridization of ET-1 of the hepatic tissue was proceeded at 3, 6, 12 and 24 h after endotoxin administration. RESULTS: ET-1 concentrations and the level of ET-1 mRNA increased rapidly and reached the peak at 6 h, and remained high at 24 h after endotoxin administration. By in situ hybridization, ET-1 mRNA was found in hepatic sinusoids, endothelial cells of portal vein and kupffer cells. CONCLUSIONS: Endotoxin may be the principal stimulating factor for expression of ET-1 mRNA in hepatic tissue. Endotoxin may affect transcription and translation level of ET-1, leading to increase of the synthesis and release of ET-1. The hepatic vascular endothelial cells, hepatic sinusoids and Kupffer cells all synthesize and release ET-1.

The embryonic development of the human lamina cribrosa.

OBJECTIVE: To investigate the embryonic development of lamina cribrosa of human eyes. METHODS: The lamina cribrosa of 291 eyes were studied including 260 eyes taken from embryos and fetus of 7 weeks to full term and 31 eyes from infants under 1 year of age by using light microscopy (LMS), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). RESULTS: From observations, it was found that the development of the lamina cribrosa was lagged behind the development of the optic nerve. CONCLUSIONS: In eyes taken from embryos or fetus less than 4 months of pregnancy, only ectodermal primordium of the lamina cribrosa could be seen. The initial scleral part of the lamina cribrosa began its formation in the 5th month of fetus. The fibrous tissue originating from the choroid and optic nerve sheath contributed to the formation of the lamina cribrosa in the 8th month fetus. At this time, the morphology of the lamina cribrosa is similar to that of an adult, however the thickness of the structure is still getting increased up to one year after birth.

[Mulivariate analysis of prognostic factors in non-small cell lung cancer].

OBJECTIVES: To find independent prognostic factor of non-small cell lung cancer (NSCLC), identify the population with "high-risk" of recurrence and death in resected NSCLC, and direct adjuvant therapy for clinical doctors. METHODS: 85 resected patients from NSCLC were followed up. All patients were subjected to the study by light microscope, immunochemistry, PCR-SSCP method, and DNA image analysis. Thirteen clinical, pathological, and biological factors were analysed by Cox regression model. RESULTS: The median follow-up period was 47 months. The overall 3-year survival rate was 66% and the overall 5-year survival rate 61%. Cox regression showed significant univariate predictors (P < 0.05) of early recurrence and death for NSCLC were lymphnode status, pTNM, tumor emboli, microvessel count, Ki-67 antigen labeling, DNA relative content and p53 gene mutation. Microvessel count (P < 0.001) and pTNM (P = 0.006) were independent predictors of early recurrence and cancer-death. According to the multivariate model for predicting early recurrence and cancer death, prognostic index (PI) was calculated. CONCLUSION: The PI may be a useful clinical tool for evaluating prognosis of patients and identifying the population with "high-risk" of recurrence and death in NSCLC.

[SCA1, SCA2, MJD/SCA3 (CAG)n mutation detection and analysis in patients with hereditary spinocerebellar ataxia from Chinese families].

OBJECTIVE: To assess the frequency of the SCA1, SCA2, MJD/SCA3 CAG trinucleotide repeat expansions ((CAG)n) among individuals diagnosed with hereditary spinocerebellar ataxia (SCA) from Chinese families. METHOD: The SCA1, SCA2, MJD/SCA3 (CAG)n mutation were detected with the polymerose chain reaction (PCR), denaturing polyacrylamide gel and silver staining technique in 79 patients with autosomal dominant SCA from 50 Chinese families. RESULTS: Among 50 kindreds, 2% (1/50) had the SCA1, (CAG)n, 6% (3/50) had the SCA2, (CAG)n, whereas 48% (24/50) were positive for the MJD/SCA3 (CAG)n. Thus, together SCA1, SCA2, and MJD/SCA3 represent 56% (28/50) of the autosomal dominant ataxias in our group. In two SCA1 patients the CAG repeat was expanded to 53-62 repeats, whereas in normal ivdividuals was 12-36 repeats. In seven SCA2 patients the CAG repeat was expanded to 43-47 repeats, whereas in normal ivdividuals was 22-30 repeats. In forty-two MJD/SCA3 patients the CAG repeat was expanded to 63-78 repeats, whereas in normal ivdividuals was 15-38 repeats. The SCA1, SCA2, MJD/SCA3 (CAG)n mutation were excluded in the other 28 SCA patients from 22 families. CONCLUSION: The frequency of MJD/SCA3 is substantially higher than that of SCA1 and SCA2 in the autosomal dominant SCA from Chinese families. Chinese patients with MJD/SCA3 are non-Portuguese patients with MJD/SCA3. Clinical expressions of the various SCAs overlap one another, making a diagnostic classification based on phenotype inaccurate in many instances. It is important for SCA clinical study to make a SCA gene diagnosis and genomic classification.

[Polymorphism and pologenetic affinities of genotype in pathogenic yeast].

The classification of pathogenic yeast, which is one of the most common reasons of opportunistic infection in human body, will be useful for the epidemiological survey and clinical works. The genotypic polymorphism of intergenus, interspecies and intraspecies in 48 isolates, including Candida, Cryptococcus, Torulopsis, Trichosporon, Saccaromyces had been administered carefully with patterns amplified by RAPD (Randomly amplified polymorphic DNA) in 53 primers. The pologenetic affinities had been evaluated by similarity coefficients obtained from these profiles. The results indicated that there are significant difference among intergenus, interspecies and intraspecies in Candida and related yeast. The similarity coefficients among Candida and Crytococcus, Trichosporon were maintained about 80%. The similarity coefficients among interspecies of Candida ranged from 82%-87%, and ones of intraspecies of different Candida species were more than 90%. The genotypic typing of species except C. guilliermondii seems to be related to morphological classification.

[Screening of Candida albicans fluconazole--resistant mutation strains].

We adopted the method of miminum inhibitary concentration (MIC) assay according to the method recommened by National Committee for Clinical Laboratory Standard (NCCLS), and selected 2 fluconazole--susceptible strains from over 50 strains of Candida albicans which were preserved in our center (these strains were all isolated from clinical patients). The number of the strains are: BMU8945 (MIC value is 0.125 microgram/ml) and BMU8977 (MIC value is 0.25 microgram/ml). In addition, we also picked one strain of ATCC14053 (MIC value is 0.5 microgram/ml) as our research starting strains. After the induction of UV and DES, we obtained nearly 1000 colonies of fluconazole--resistant strains (MIC > 30 micrograms/ml). After subculture, we found that the characterization of drug--resistance of these strains could be inheredited stably.
