De novo cell-free synthesis of human interferon.

Biologically active human interferon was synthesized de novo in a cell-free mouse extract stimulated with messenger RNA from induced human fibroblasts. The identity of the antiviral activity as human interferon was demonstrated by its antigenic and species specificity. Some characteristics of the cell-free synthesis were described.

[Studies on the influence of glucose infusion on parameters of hemostasis].

In continuation of previous in vitro experiments the influence of glucose infusions on the following haemostatic functions was investigated: bleeding time, platelet count, platelet aggregation, release reaction, fibrinogen concentration, partial thromboplastin time. Five volunteers with normal metabolism a glucose infusion (100 g) was given for two hrs. Before, 1, 2, 3, 4 and 5 hrs after the beginning of the infusion blood sugar, insulin level and haemostatic parameters were determined. Apart from an increase in the glucose and insulin level, a prolonged bleeding time, increased platelet count, inhibition of platelet aggregation and release reaction occurred. Simultaneously, fibrinogen concentration increased and partial thromboplastin time shortened. The possible causes of these changes in haemostasis during glucose supply are discussed.

[Impulse cytophotometry in neoplasm prevention. The behavior of normal vaginal smears in the impulse cytophotometric histogram].

Evaluation of histograms of normal cervical and vaginal smears, produced by ""rapid flow through'' cytophotometry. Normal cytological cervical and vaginal smears were examined for their appearance in histograms in order to find criteria for the classification of ""normal non-pathological'' cytological slides. Simultaneously, the reliability of the standardized method for preparation of vaginal and cervical smears was checked. The diagrams and data produced by the histograph were also coded on line and analyzed by a computer; thus obtaining an objective mathematical analysis and avoiding errors due to subjective visual analysis of the histograms. The objective interpretation of histograms of normal vaginal smears is an important step to the realisation of automatic pre-screening.

Thiostrepton-resistant mutants of Bacillus subtilis: localization of resistance to the 50S subunit.

A number of thiostrepton-resistant mutants of Bacillus subtilis were obtained. The thi mutations map proximally to strA. Effects of thiostrepton on polyphenylalanine synthesis with ribosomes of S-100 fractions from parent and mutant strains indicated that resistance was localized to the ribosomes. Furthermore, effects of thiostrepton on binding of [3H]GTP to ribosomes and 50S subunits from thiostrepton-sensitive and -resistant strains localized the site of resistance to the 50S subunit. In addition, revertants from thiostrepton-resistance to thiostrepton-sensitivity were obtained. Ribosomes and 50S subunits from these thiostrepton-sensitive revertants were sensitive to thiostrepton similar to parental sensitive B. subtilis.

Binding of N-substituted erythromycyclamines to ribosomes.

Several N-substituted erythromycylamines were evaluated for their ability to inhibit the binding of [(14)C]erythromycin to ribosomes. The association and dissociation constants for the binding of each compound to Escherichia coli ribosomes were determined. These studies have resulted in the development of three types of probes for topological studies of the erythromycin-binding site and the ribosome: the chemically reactive bromoacetamido, the photoreactive N-(2)-nitro-4-azidophenyl)glycinamido, and the fluorescent fluorescein isothiocyanate derivatives of 9(S)-erythromycylamine.

Binding of thiostrepton to ribosomes from thiostrepton-sensitive and -resistant Bacillus subtilis strains.

Binding of [(35)S]thiostrepton to ribosomes from thiostrepton-sensitive and -resistant strains of Bacillus subtilis was studied. Ribosomes from thiostrepton-resistant strains bound relatively little thiostrepton compared with ribosomes from thiostrepton-sensitive B. subtilis. In addition, ribosomes from revertant strains that were obtained as thiostrepton-sensitive revertants from thiostrepton-resistant strains bound [(35)S]thiostrepton similarly to ribosomes from the sensitive parental strain.

Prevention of premature delivery in a unicornuate uterus by cervical cerclage.

A 26-year-old woman had two premature deliveries when 6 months pregnant. Neither produced a living child. A hysterosalpingogram was done, confirming a diagnosis of unicornuate uterus. When the patient became pregnant again, a third and fourth time, cervical cerclage was done in both cases. Pregnancies were terminated by cesarean section because of breech presentation and malformation of the uterus and in each case a healthy living child was delivered. In our view, in order to prevent premature deliveries, when a diagnosis of uterus malformations has been confirmed, a cerclage should be performed.

Lead content of human hair (1871-1971).

Samples of antique and contemporary human hair were analyzed for lead by atomic absorption spectroscopy. Antique hair contained a significantly greater amount of lead than did contemporary hair.
