Comparative genomics of the lactic acid bacteria.

Lactic acid-producing bacteria are associated with various plant and animal niches and play a key role in the production of fermented foods and beverages. We report nine genome sequences representing the phylogenetic and functional diversity of these bacteria. The small genomes of lactic acid bacteria encode a broad repertoire of transporters for efficient carbon and nitrogen acquisition from the nutritionally rich environments they inhabit and reflect a limited range of biosynthetic capabilities that indicate both prototrophic and auxotrophic strains. Phylogenetic analyses, comparison of gene content across the group, and reconstruction of ancestral gene sets indicate a combination of extensive gene loss and key gene acquisitions via horizontal gene transfer during the coevolution of lactic acid bacteria with their habitats.

Impairments in immune cell function in B cell chronic lymphocytic leukemia.

B cell chronic lymphocytic leukemia (B-CLL) is a common clonal B cell leukemia that is often accompanied by a multitude of immune abnormalities. Each immune defect may be linked to several of the common complications affecting B-CLL patients. Furthermore, the combined abnormalities constitute a significant immunodeficiency for each patient. Importantly, some of the immune dysfunctions are potentially very relevant to the in vivo survival status of the leukemic B cell. The elucidation of these abnormalities in the circulating non-malignant immune cells of B-CLL patients has generated important insights into the biology of the disease. This discussion reviews the immune abnormalities of the clonal malignant B cells, the polyclonal B cells, and the immunoregulatory T cells and natural killer cells in B-CLL. In addition, we indicate the potential for immunotherapeutic protocols as innovations in treating this disease.

Integrated maps of the chromosomes in Dictyostelium discoideum.

Detailed maps of the six chromosomes that carry the genes of Dictyostelium discoideum were constructed by correlating physically mapped regions with parasexually determined linkage groups. Chromosomally assigned regions were ordered and positioned by the pattern of altered fragment sizes seen in a set of restriction enzyme mediated integration-restriction fragment length polymorphism (REMI-RFLP) strains each harboring an inserted plasmid that carries sites recognized by NotI, SstI, SmaI, BglI and ApaI. These restriction enzymes were used to digest high molecular weight DNA prepared from more than 100 REMI-RFLP strains and the resulting fragments were separated and sized by pulsed-field gels. More than 150 gene probes were hybridized to blots of these gels and used to map the insertion sites relative to flanking restriction sites. In this way, we have been able to restriction map the 35 mb genome as well as determine the map position of more than 150 genes to with approximately 40 kb resolution. These maps provide a framework for subsequent refinement.

Long-term central venous catheters: issues for care.

Long-term central venous catheters allow the safe administration of chemotherapy, blood and blood products, total parenteral nutrition, fluids, and other medications. Despite their benefits, the risk of certain complications (e.g., fibrin sleeve and mural thrombus formation, infection, catheter occlusion, extravasation, and catheter malposition) exist for every person who has a catheter. Thus, preventative measures, recognition of early signs and symptoms of complications, and adequate care of utmost importance.

[Experimental studies of electroplating-resin bonding systems].

Optimized conditioning of galvanically produced gold frameworks (Auro-Galva-Crown-System) by means of Silicoater, Silicoater MD and Rocatec techniques gives rise to a reliable bond to veneering resins. After experimental stress (thermocycling) the strength of the compound could be found between 17 and 26 MPa and SEM investigations of the contact zone showed no crevices.

[Microbiological studies of self-disinfecting alginate impression materials].

Using microbiologically experimental methods and observations corresponding to praxis the efficiency of the addition of antiseptics to alginate is evaluated. The addition of chlorhexidine to the alginate leads to a considerable reduction of the amounts of germs, but an one hundred percent disinfection of the alginates is not always performed. An influence of the alginate materials and the water quality on the antimicrobial efficacy of chlorhexidine has been proved. The always occurring contamination of the impression tray rules out a complete stopping of infection between the patient and the laboratory staff.

[Preclinical biological testing of veneer composite materials].

The toxicity of commercial (Dentacolor, Visio-Gem) and other light reactive veneer composite materials under development (DG) was investigated by the hemolysis test. In contrast to a standard composite filling material (Evicrol) the veneer composite materials caused only small hemolysis rates.

[Experimental microbiological research on instrument and denture disinfection with a disinfectant spray for dental practice and the use of patients. 2. Virological studies; the evaluation summary].

Carrier experiments using Echo- and Influenza-viruses on denture basis resins and fragments of orthodontic appliances resulted in good and even very good virucidal effects of disinfectant sprays Desident and Fesia-sept. The application of the disinfectant as spray seem to be advantageous, but is not recommendable. The diverse causes of this conclusion are discussed and an alternative way is proposed.

[Experimental microbiological research on instrument and denture disinfection with a disinfectant spray for dental practice and the use of patients. 1. The formulation of the problem and bacteriological studies].

Three disinfectant sprays (Arugeen, Desident and Fesia-sept) proved to be very efficient against pure cultures of aero bacteria species, Cand. albicans, saliva flora and entero- and influenza viruses. The methods included carrier experiments using denture basis resins, fragments of orthodontic appliances and screws (instead of instruments).

[In vitro microbiological research on oral ointments].

Two oral ointments were investigated of their antimicrobial activities using agar diffusion test and suspension method with aerobic germs and Candida albicans. The oral ointment Parodontal-Mundsalbe-neu was better then Parodontal-Mundsalbe-alt with regard to intensity, begin and end of action. In our experiments the field of activity of both ointments against the species tested was identically.

[Haemotoxicity of dental luting cements].

A glass ionomer luting cement (AquaCem) shows a relatively low haemolytic activity in comparison with two zinc phosphate cements. Especially the initial irritation by this cement is smaller. Although it is possible that AquaCem particularly, in unfavourable cases, may damage the pulpa dentin system; this is due to the slowly decrease of the haemolytic activity with increasing of the probes. We found that Adhesor showed in dependence of the batches a varying quality.
