How an expert corporate fitness program might be designed.

In this paper, the ideas and concepts of expert systems were combined with the principles of fitness to demonstrate how an expert system could be designed to prescribe exercise routines for corporate employees. Since the examples and the rules of fitness used herein were just a small fraction of what could be used in building such a system, it would be possible to spend years developing a huge system that employs thousands of rules. However, as the supply of skilled labor continues to fall short of corporations' needs, more effort will be brought to bear on maintaining fitness in the work force. Such expert systems may be a reality in the not-too-distant future.

Reprogramming cell fate in the developing Drosophila retina: transformation of R7 cells by ectopic expression of rough.

In the developing eye of Drosophila, the homeo box gene rough is required in the developing photoreceptor cells R2 and R5 for the correct development of the neighboring R3 and R4 cells. We have expressed rough ectopically in a limited subset of developing ommatidial cells using the sevenless enhancer. Expression of rough in the presumptive R7 cell transforms this cell into an R1-6 type photoreceptor. This transformation is cell autonomous in contrast to the apparent nonautonomy of the rough mutant phenotype and depends on the presence of the sevenless gene. We propose that in wild type, rough functions autonomously in the specification of R2/5 photoreceptor cell identity but by itself cannot initiate neural development.

Chaoptin, a cell surface glycoprotein required for Drosophila photoreceptor cell morphogenesis, contains a repeat motif found in yeast and human.

Chaoptin is a photoreceptor cell-specific membrane protein. Analysis of chaoptin mutants demonstrates that this glycoprotein plays a critical role in photoreceptor cell morphogenesis. We have deduced chaoptin's primary structure from the cDNA sequence and examined its membrane topology. Chaoptin is largely composed of 41 potentially amphipathic repeats. Remarkably homologous repeats have been reported in both yeast and human, suggesting their general importance as a structural and/or functional motif. Chaoptin synthesized in vitro is associated with rough endoplasmic reticulum microsomal membrane in an integral fashion, consistent with its extraction characteristics in vivo. The resistance to proteolytic digestion of in vitro synthesized and processed chaoptin suggests that it is primarily localized to the extracellular leaflet of the lipid bilayer. These data are consistent with the proposal that chaoptin is involved in the adhesion between photoreceptor cell membranes.

Transcatheter arterial chemoembolization with anticancer drug in iodized oil for primary hepatic carcinoma.

Transcatheter arterial chemoembolization using a suspension of anticancer drug in iodized oil combined with Gelfoam embolization was performed in 85 patients with advanced primary hepatic carcinoma. Long-term retention of iodized oil in the tumor was observed in 90% of the cases, and changes in tumor size were observable by abdominal X-ray film on follow-up studies. There was a decrease of tumor size in all cases and the overall 1-year survival rate was 43%. In tumors whose diameter was less than 20 cm and in patients without portal vein thrombosis the survival rate was nearly 60%. Combined treatment with anticancer drug in iodized oil and Gelfoam embolization of the proper hepatic artery was superior to use of the suspension alone. There was also better detection of small cancer nodules by the suspension as compared to conventional angiography.
