Evidence from knockout mice against physiologically significant aquaporin 8-facilitated ammonia transport.

Aquaporin (AQP)8-facilitated transport of NH(3) has been suggested recently by increased NH(3) permeability in Xenopus oocytes and yeast expressing human or rat AQP8. We tested the proposed roles of AQP8-facilitated NH(3) transport in mammalian physiology by comparative phenotype studies in wild-type vs. AQP8-null mice. AQP8-facilitated NH(3) transport was confirmed in mammalian cell cultures expressing rat or mouse AQP8, in which the fluorescence of a pH-sensing yellow fluorescent protein was measured in response to ammonia (NH(3)/NH(4)(+)) gradients. Relative AQP8 single-channel NH(3)-to-water permeability was approximately 0.03. AQP8-facilitated NH(3) and water permeability in a native tissue was confirmed in membrane vesicles isolated from testes of wild-type vs. AQP8-null mice, in which BCECF was used as an intravesicular pH indicator. A series of in vivo studies were done in mice, including 1) serum ammonia measurements before and after ammonia infusion, 2) renal ammonia clearance, 3) colonic ammonia absorption, and 4) liver ammonia accumulation and renal ammonia excretion after acute and chronic ammonia loading. Except for a small reduction in hepatic ammonia accumulation and increase in ammonia excretion in AQP8-null mice loaded with large amounts of ammonia, there were no significant differences in wild-type vs. AQP8-null mice. Our results support the conclusion that AQP8 can facilitate NH(3) transport but provide evidence against physiologically significant AQP8-facilitated NH(3) transport in mice.

[Effects of chemotherapy in recurrent endometrial carcinoma].

OBJECTIVE: To explore the effects of chemotherapy in recurrent endometrial carcinoma. METHODS: A retrospective study was carried out on the clinical data of 20 cases of recurrent endometrial carcinoma hospitalized from May. 1992 to Jun. 2002. RESULTS: The recurrence rate of these 20 cases was 11.0%. The recurrence occurred (17 +/- 10) months after the first therapy. Eleven of the 20 patients (55.0%) had accepted chemotherapy with (6 +/- 4) courses and were followed up for (17 +/- 16) months after the second therapy. Fifteen cases (75.0%) died of cancer, 4 cases survived with tumor, and only 1 case (5.0%) was tumor-free. Univariate model revealed that chemotherapy and number of chemotherapy course were related with the prognosis (P = 0.0146, P = 0.0353). Radiotherapy affected the prognosis too (P = 0.0414). But pathologic subtype, stage, tumor cell differentiation, age, recurrent time, surgery, chemotherapy scheme, drug infusion way , and endocrine therapy had no relation with the prognosis (P = 0.5517, 0.5763, 0.1879, 0.880, 0.453, 0.0672, 0.0795, 0.5956, and 0.2035). Multivariate model revealed that chemotherapy had influence on the prognosis (beta = -1.349, OR = 0.259, P = 0.026). CONCLUSION: Combined treatment raises the survival of the recurrent endometrial carcinoma patients, in which chemotherapy plays a leading role and influences the prognosis remarkably.

17 beta-estradiol activates PI3K/Akt signaling pathway by estrogen receptor (ER)-dependent and ER-independent mechanisms in endometrial cancer cells.

Cellular response to estrogen is mediated both by estrogen receptor (ER) binding to estrogen response element (ERE) and by non-nuclear actions like activation of signal transducing pathways. The main aims are to study if PI3K/Akt signaling pathway can be activated by 17beta-estradiol (E2) via non-nuclear action and to investigate the relationship of the action of E2 and ER in endometrial cancer cells expressing with different status of ER. The levels of phosphorylated Akt (Ser473) (P-Akt) and total Akt were examined by western blot and Akt kinase activity was measured in cells after stimulation with 1 microM E2 at different time points. Inhibitory role of LY294002 on activation of Akt induced by E2 and its estrogen antagonist, ICI182780 were also tested. P-Akt/Akt was used as a measure of activation of Akt. We found that maximum P-Akt/Akt and Akt kinase activity took place at 30 min in Ishikawa cells and 15 min in HEC-1A cells and the activation persisted for at least 2 h after stimulation with 1 microM E2. The activation of Akt elicited gradually with increasing doses of E2. PI3K inhibitor, LY294002, stopped the activating Akt in a dose-dependent manner and 50 microM LY294002 completely blocked the activation of Akt induced by E2. ICI182780 could block the activation of PI3K/Akt in ER-positive Ishikawa cells but not in HEC-1A cells with poor-expressed ER. This study demonstrated that E2 is able to promptly activate PI3K/Akt signal pathway in Ishikawa cells in an ER-dependent manner and ER-independent in HEC-1A cells. Blockage of PI3K/Akt cascade may become a potential and effective way to control endometrial carcinoma, especially in ER-negative cancers, which show no response to endocrinal therapy.

Urea and urine concentrating ability in mice lacking AQP1 and AQP3.

Aquaporin-1 (AQP1) and aquaporin-3 (AQP3) water channels expressed in the kidney play a critical role in the urine concentrating mechanism. Mice with AQP1 or AQP3 deletion have a urinary concentrating defect. To better characterize this defect, we studied the influence of an acute urea load (300 mumol ip) in conscious AQP1-null, AQP3-null, and wild-type mice. Urine was collected and assayed every 2 h, from 2 h before (baseline) to 8 h after the urea load. Mice of all genotypes excreted the urea load in approximately 4 h with the same time course. Interestingly, despite their low baseline, the AQP3-null mice raised their urine osmolality and urea concentration progressively after the urea load to values almost equal to those in wild-type mice at 8 h. In contrast, urine non-urea solute concentration did not change. Urine volume fell in the last 4 h to about one-fourth of basal values. AQP1-null mice increased their urine flow rate much more than AQP3-null mice and showed no change in urine osmolality and urea concentration. The urea load strongly upregulated urea transporter UT-A3 expression in all three genotypes. These observations show that the lack of AQP3 does not interfere with the ability of the kidney to concentrate urea but impairs its ability to concentrate other solutes. This solute-selective response could result from the capacity of AQP3 to transport not only water but also urea. The results suggest a novel role for AQP3 in non-urea solute concentration in the urine.

Growth and activation of PI-3K/PKB and Akt by stromal cell-derived factor 1alpha in endometrial carcinoma cells with expression of suppressor endoprotein PTEN.

BACKGROUND: Mutation or deletion in the phosphatase and tensin homologue deleted on chromosome ten (PTEN) gene has been identified as an important cause of endometrial carcinoma; stromal cell derived factor-1alpha (SDF-1alpha) exerts growth-promoting effects on endometrial cancer cells through activation of the PI-3 kinase/Akt pathway and downstream effectors such as extracellular-responsive kinase (ERK). In this study, a plasmid containing the PTEN gene was transfected into Ishikawa cells to investigate the difference in growth and signal transduction between Ishikawa-PTEN and Ishikawa cells after SDF-1alpha stimulation, and to study mechanisms of the involvement of PTEN protein in endometrial carcinoma development. METHODS: Ishikawa cells were transfected with a plasmid (pLXSN-PTEN) containing the PTEN gene and a plasmid (pLXSN-EGFP) with enhanced green fluorescent protein (EGFP). Cells were then screened to obtain Ishikawa-PTEN cells and Ishikawa-neo cells that can both stably express PTEN protein and EGFP. Expression of PTEN protein, phosphorylation levels of AKT and ERK (pAKT and pERK) and growth differences in Ishikawa-PTEN, Ishikawa-neo and Ishikawa cells before and after SDF-1alpha stimulation were then determined by Western blots and MTT assays. RESULTS: Western blot analysis showed that Ishikawa cells produced PTEN after transfection with the PTEN gene. At 15 minutes after SDF-1alpha stimulation, the pAKT level of Ishikawa-PTEN cells was lower than that of Ishikawa-neo cells and Ishikawa cells. There was no significant difference in pERK levels among the three cell lines. The positive effect of SDF-1alpha on Ishikawa-PTEN cells growth was markedly less than the effect on Ishikawa-neo and Ishikawa cells. However, in the absence of SDF-1alpha stimulation (baseline), the pAKT level in Ishikawa-PTEN cells was less than that in Ishikawa cells. There was a significant difference in growth between the Ishikawa-PTEN cells and the Ishikawa-neo cells. CONCLUSIONS: PTEN gene transfection can regulate the level of pAKT but not pERK in Ishikawa-PTEN cells. PTEN protein may suppress the growth-promoting effect of SDF-1alpha on endometrial carcinoma by inhibiting the PI-3K/AKT signal transduction pathway.

Mouse model of inducible nephrogenic diabetes insipidus produced by floxed aquaporin-2 gene deletion.

Transgenic mouse models of defective urinary concentrating ability produced by deletion of various membrane transport or receptor proteins, including aquaporin-2 (AQP2), are associated with neonatal mortality from polyuria. Here, we report an inducible mouse model of AQP2 gene deletion with severe polyuria in adult mice. LoxP sequences were inserted into introns 1 and 2 in the mouse AQP2 gene by homologous recombination in embryonic stem cells. Mating of germ-line AQP2-loxP mice with tamoxifen-inducible Cre-expressing mice produced offspring with inducible homozygous Cre-AQP2-loxP, which had a normal phenotype. Tamoxifen injections over 10 days resulted in AQP2 gene excision, with undetectable full-length AQP2 transcript in kidney and a >95% reduction in immunoreactive AQP2 protein. Urine osmolality decreased from approximately 2,000 to <500 mosmol/kgH(2)O after 4-5 days, with urine output increasing from 2 to 25 ml/day. Urine osmolality did not increase after water deprivation. Interestingly, AQP3 protein expression in the collecting duct was increased by about fivefold after AQP2 gene excision. Mild renal damage was seen after 6 wk of polyuria, with collecting duct dilatation, yet normal creatinine clearance and serum chemistries. These results establish the first adult model of nephrogenic diabetes insipidus (NDI) caused by AQP2 deficiency, with daily urine output comparable to body weight, although remarkable preservation of renal function compared with non-inducible NDI models.

Application of surface-enhanced laser desorption/ionization time-of-flight-based serum proteomic array technique for the early diagnosis of prostate cancer.

AIM: To identify the serum biomarkers of prostate cancer (PCa) by protein chip and bioinformatics. METHODS: Serum samples from 83 PCa patients and 95 healthy men were taken from a mass screening in Changchun, China. Protein profiling was carried out using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). The data of spectra were analyzed using two bioinformatics tools. RESULTS: Eighteen serum differential proteins were identified in the PCa group compared with the control group (P < 0.01). There were four proteins at the higher serum level and 14 proteins at the lower serum level in the PCa group. A decision tree classification algorithm that used an eight-protein mass pattern was developed to correctly classify the samples. A sensitivity of 92.0% and a specificity of 96.7% for the study group were obtained by comparing the PCa and control groups. CONCLUSION: We identified new serum biomarkers of PCa. SELDI-TOF MS coupled with a decision tree classification algorithm will provide a highly accurate and innovative approach for the early diagnosis of PCa.

Decreased saliva secretion and down-regulation of AQP5 in submandibular gland in irradiated rats.

The molecular mechanisms of radiation-induced xerostomia remain unclear. The purpose of this study was to investigate the alterations of aquaporins (AQPs) and Na(+)/K(+)-ATPase in irradiated rat submandibular glands and to test the hypothesis that down-regulation of AQP5 expression in irradiated salivary glands is one of the mechanisms of radiation-induced xerostomia. Saliva from control and irradiated rat submandibular glands was analyzed. The mRNA level of AQP5 in the submandibular glands was assessed by semi-quantitative RT-PCR and in situ hybridization. The protein expression of AQP5, AQP1 and Na(+)/K(+)-ATPase was determined by Western blotting and immunohistochemistry. The body weight, submandibular gland weight, and saliva secretion of irradiated rats significantly decreased by 12, 24 and 32% on day 3 and 24, 16 and 38% on day 30 postirradiation, respectively. There was a significant increase in the protein concentration and osmolality of saliva in irradiated rats on days 3 and 30 postirradiation. However, there was no significant difference between irradiated and control rats in total saliva protein secretion. RT-PCR analysis showed that mRNA expression of AQP5 was significantly down-regulated by 37 and 51% in irradiated rats on days 3 and 30 postirradiation, respectively. Immunoblotting showed that the AQP5 protein level was decreased by 40 and 60% in irradiated glands, in contrast to the slight reductions of AQP1 and Na(+)/K(+)-ATPase proteins. Immunohistochemical analysis demonstrated that loss of AQP5 protein occurred throughout the irradiated glands, while no significant reduction was detected in AQP1 and Na(+)/ K(+)-ATPase labeling density. These results suggest that the preferential down-regulation of AQP5 with minor effects on AQP1 and Na(+)/K(+)-ATPase may contribute to radiation-induced salivary dysfunction.

[Protective effects of glycyrrhizin on adriamycin nephropathy in rats].

OBJECTIVE: To observe the effects of glycyrrhizin on laminin (LN) expression in kidney tissue and excretory quantity of urine protein of rats with adriamycin nephropathy, and to explore the protective effects of glycyrrhizin on glomerulosclerosis. METHODS: Eighteen SD rats were randomly divided into 3 groups: normal control group (n=6), untreated group (n=6) and glycyrrhizin-treated group (n=6). Adriamycin nephropathy was induced in rats in the last two groups by intravenous injection of adriamycin. The rats in the glycyrrhizin-treated group were fed glycyrrhizin for eight weeks, whereas the rats in the normal control group and untreated group were fed normal saline solution for eight weeks too. The levels of 24 h urine protein (Upr), serum creatinine (sCr), blood urea nitrogen (BUN) and serum cholesterol (Ch) of rats in each group were examined before treatment and after the treatment for four and eight weeks. The renal morphological changes were observed under a microscope. The expression level of LN in renal tissue was detected by streptavidin-biotin peroxidase complex (SABC) method. RESULTS: The levels of 24 h Upr after the treatment for four and eight weeks in the glycyrrhizin-treated group were both significantly decreased as compared with those in the untreated group. The pathological morphological changes of renal tissue in the glycyrrhizin-treated group were remarkably alleviated, and the expression level of LN in renal tissue was also decreased in the glycyrrhizin-treated group as compared with those in the untreated group. CONCLUSION: The glycyrrhizin exerts certain protective effects on adriamycin nephropathy in rats by reducing excretory quantity of urine protein, decreasing expression level of LN in renal tissue, improving renal function and lessening the severity degree of glomerulosclerosis so as to retard the development of glomerulosclerosis.

[Regulatory effect of 17beta-estradiol on expression of orphan nuclear receptor ERRalpha in endometrial carcinoma cell lines].

BACKGROUND & OBJECTIVE: Estrogen receptor-related receptor alpha (ERRalpha), a member of the subfamily of orphan nuclear receptors, could compete with estrogen receptor alpha (ERalpha) to bind the same target genes and interfere in ER signal pathway. Therefore, it might be associated to the tumorigenesis of endometrial carcinoma. This study was to explore the regulatory effect of 17beta-estradiol (17beta-E(2)) on ERRalpha expression, and to elucidate the relationship between ERRalpha and ER signal pathway in endometrial carcinoma cell lines. METHODS: ERalpha-positive cell line Ishikawa and ERalpha-negative cell line HEC-IA were treated with different concentrations of 17beta-E(2) (1x10(-10) mol/L, 1x10(-8) mol/L, and 1x10(-6) mol/L) for 24 h and 48 h, respectively. The levels of ERRalpha mRNA and protein were examined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. 17beta-E(2) (1x10(-8) mol/L) and complete ER inhibitor ICI182,780 (1x10(-6) mol/L) were given concomitantly to observe the change of ERRalpha expression. RESULTS: The levels of ERRalpha mRNA and protein in Ishikawa cells were down-regulated after stimulated for 24 h and 48 h by different concentrations of 17beta-E(2). The maximal effect was observed at the concentration of 1x10(-8) mol/L. When 17beta-E(2) and ICI182,780 were given simultaneously to Ishikawa cells, this down-regulation was blocked. However, the level of ERRalpha mRNA, not protein, in HEC-IA cells was up-regulated after stimulated by different concentrations of 17beta-E(2) for 24 h. After stimulated by 17beta-E(2) for 48 h, the level of ERRalpha protein was up-regulated, which could not be blocked by ICI182,780. CONCLUSIONS: 17beta-E(2) can down-regulate the expression of ERRalpha in Ishikawa cells, which is mediated by ERalpha. 17beta-E(2) can up-regulate the expression of ERRalpha in HEC-IA cells, but this regulation cannot be blocked by ICI182,780.

[Genetic polymorphisms in the promoter region of cyclooxygenase-2 and their association with risk of gastric cancer].

OBJECTIVE: To examine the genetic polymorphisms in the promoter region of cyclooxygenase-2 ( COX-2) and evaluate their association with the risk of gastric cancer. METHODS: Single-strand conformational polymorphism and DNA sequencing were used to screen the genetic variants of the COX-2 promoter region. Total 323 patients with gastric cancer and 646 frequency-matched controls were genotyped by polymerase chain reaction-restriction fragment length polymorphism method. Odds ratios (OR) and 95% confidence intervals (CI) were estimated by logistic regression. Haplotype frequency was estimated using Haplo. stat software. RESULTS: Three single nucleotide polymorphisms, including - 1290A > G, -1195G > A, and -765G > C were identified in the promoter of COX- 2. Case-control analysis showed an increased risk of developing gastric cancer for the -1290AG (OR 1.64; 95% CI 1.03-2.61), -1195AA (OR 2.68; 95% CI 1.65-4.33), and -765CG (OR 2.66; 95% CI 1.53-4.64) genotype carriers, respectively, compared with noncarriers. A greater risk of developing gastric cancer was observed for the A(-1290) -A(-1195) -C(-765) haplotype compared with the A(-1290) -G(-1195) -G(-765) haplotypes (OR 11.80; 95% CI 2.43-57.25). CONCLUSION: Genetic polymorphisms in COX-2 promoter region may play a role in gastric carcinogenesis.

[Effect of conjugated linoleic acid on the gene expression of the fatty acid binding proteins in resistant rat].

OBJECTIVE: To study the effect of conjugated linoleic acid on gene expression of fatty acid binding proteins (ap2) in white obesity tissue of rats with insulin resistance, and explore the mechanism of resisting diabetes by CLA. METHODS: Male Wistar rats were randomly separated to control group, high-fat group and high fat + CLA group (0.75g, 1.50g, 3.00g by diet weight), Using reverse transcription polymerse chain reaction (RT-PCR) technique to measure the expression level of ap2 and Peroxisome proliferators-activated receptor--gamma (PPARgamma) mRNA expression. RESULTS: The FFA, serum insulin and glucose levels of rats with insulin resistance were higher than that of rats fed with basic diet, supplement of CLA can decrease FFA, serum insulin and glucose levels of rats with insulin resistance, and CLA can increase the expression of ap2 and PPARgamma mRNA in adipose tissue of rat with insulin resistance. CONCLUSION: CLA can improve insulin resistance of obese rat and increase the expression of ap2 mRNA,possibly acting through activing PPARgamma.

[Preliminary study of membrane estrogen receptor expression in endometrial carcinoma cells].

OBJECTIVE: To analyze if there are membrane estrogen receptors (ER) in endometrial carcinoma and if there is some relationship between membrane ER and nuclear ER. METHODS: The cell membrane and total cell ERalpha and ERbeta expressions of high and moderate differentiation endometrial carcinoma cells (Ishikawa and HEC-1A cells) were analyzed. Intermittent immunofluorescence dyeing and fluorescent microscopy were carried out with the cells treated with polyformaldehyde and Triton X-100. Intermittent immunofluorescence dyeing and flow cytometry were carried out with the live cells and the cells treated with Triton X-100 respectively. RESULTS: There were fluorescences on the membrane of the Ishikawa and HEC-1A cells which were treated with polyformaldehyde. When the cells were treated with Triton X-100, the fluorescences were also seen inside the cells. The fluorescence intensity of ERalpha and ERbeta in Ishikawa cell membrane (1.09 +/- 0.21, 1.27 +/- 0.33) was stronger than the control, but there were no significant differences (P > 0.05). When treated with Triton X-100, the total cell fluorescence intensity of ERalpha and ERbeta in Ishikawa cell (4.21 +/- 0.34, 4.69 +/- 1.96) was stronger than the membrane (P < 0.05). The ERalpha and ERbeta fluorescence intensity of HEC-1A cell membrane (1.58 +/- 0.13, 1.49 +/- 0.04) were stronger than the control (P < 0.05). The fluorescence intensity of ERalpha and ERbeta of the HEC-1A cell (2.34 +/- 0.33, 2.52 +/- 0.15) was stronger than the membrane also (P < 0.05). The membrane ERalpha fluorescence intensity of Ishikawa was lower than HEC-1A (P = 0.028). But the total cell ERalpha fluorescence intensity of Ishikawa was higher than HEC-1A (P = 0.002). CONCLUSIONS: There are membrane ER on endometrial carcinoma cells Ishikawa and HEC-1A. The membrane ER must have some similarity to the nuclear receptor. There is no direct correlation between the quantity of the membrane ER and nuclear ER.

[Association between genetic polymorphism in xeroderma pigmentosum G gene and risks of laryngeal and hypopharyngeal carcinomas].

OBJECTIVE: To study the association between polymorphism of DNA repair gene xeroderma pigmentosum G (XPG) Asp1104His and the risks of developing laryngeal and hypopharyngeal carcinomas. METHODS: Totally 175 patients with laryngeal or hypopharyngeal carcinoma and 525 cancer-free controls were genotyped for the polymorphism by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The odds ratio (OR) and 95% confidence interval (CI) were calculated using unconditional logistic regression model. RESULTS: Compared with those having the Asp/Asp genotype, patients having the XPG 1104Asp/His genotype had a higher risk for laryngeal carcinoma (OR = 2.46, 95% CI = 1.15-5.24, P < 0.05), but not for hypopharyngeal carcinoma (OR = 1.36, 95% CI = 0.87-2.12, P > 0.05). In addition, the XPG 1104Asp/His genotype appeared to be associated with well differentiated squamous cell carcinoma in both larynx and hypopharynx (OR = 1.88, 95% CI = 1.05-3.40, P < 0.05 ). CONCLUSION: The XPG Asp1104His polymorphism may play a role in the development of laryngeal and hypopharyngeal carcinomas.

FASL -844C polymorphism is associated with increased activation-induced T cell death and risk of cervical cancer.

The FAS receptor-ligand system plays a key role in regulating apoptotic cell death, and corruption of this signaling pathway has been shown to participate in tumor-immune escape and carcinogenesis. We have recently demonstrated (Sun, T., X. Miao, X. Zhang, W. Tan, P. Xiong, and D. Lin. 2004. J. Natl. Cancer Inst. 96:1030-1036; Zhang, X., X. Miao, T. Sun, W. Tan, S. Qu, P. Xiong, Y. Zhou, and D. Lin. 2005. J. Med. Genet. 42:479-484) that functional polymorphisms in FAS and FAS ligand (FASL) are associated with susceptibility to lung cancer and esophageal cancer; however, the mechanisms underlying this association have not been elucidated. We show that the FAS -1377G, FAS -670A, and FASL -844T variants are expressed more highly on ex vivo-stimulated T cells than the FAS -1377A, FAS -670G, and FASL -844C variants. Moreover, activation-induced cell death (AICD) of T cells carrying the FASL -844C allele was increased. We also found a threefold increased risk of cervical cancer among subjects with the FASL -844CC genotype compared with those with the -844TT genotype in a case-control study in Chinese women. Together, these observations suggest that genetic polymorphisms in the FAS-FASL pathway confer host susceptibility to cervical cancers, which might be caused by immune escape of tumor cells because of enhanced AICD of tumor-specific T cells.

Down-regulation of signal transducer and activator of transcription 3 expression using vector-based small interfering RNAs suppresses growth of human prostate tumor in vivo.

PURPOSE: Signal transducer and activator of transcription 3 (Stat3) is constitutively activated in a variety of cancers and it is a common feature of prostate cancer. Thus, Stat3 represents a promising molecular target for tumor therapy. We applied a DNA vector-based Stat3-specific RNA interference approach to block Stat3 signaling and to evaluate the biological consequences of Stat3 down-modulation on tumor growth using a mouse model. EXPERIMENTAL DESIGN: To investigate the therapeutic potential of blocking Stat3 in cancer cells, three small interfering RNAs (siRNA; Stat3-1, Stat3-2, and Stat3-3) specific for different target sites on Stat3 mRNA were designed and used with a DNA vector-based RNA interference approach expressing short hairpin RNAs to knockdown Stat3 expression in human prostate cancer cells in vitro as well as in vivo. RESULTS: Of the three equivalently expressed siRNAs, only Stat3-3 and Stat3-2, which target the region coding for the SH2 domain and the coiled-coil domain, respectively, strongly suppressed the expression of Stat3 in PC3 and LNCaP cells. The Stat3-1 siRNA, which targeted the DNA-binding domain, exerted no effect on Stat3 expression, indicating that the gene silencing efficiency of siRNA may be dependent on the local structure of Stat3 mRNA. The Stat3 siRNAs down-regulated the expression of Bcl-2 (an anti-apoptotic protein), and cyclin D1 and c-Myc (cell growth activators) in prostate cancer cells. Inhibition of Stat3 and its related genes was accompanied by growth suppression and induction of apoptosis in cancer cells in vitro and in tumors implanted in nude mice. CONCLUSIONS: These data indicate that Stat3 signaling is a promising molecular target for prostate cancer therapy and that vector-based Stat3 siRNA may be useful as a therapeutic agent for treatment of prostate cancer.

[Regulation of orphan receptor ERR alpha by estrogen and progesterone in endometrial carcinoma cell line].

OBJECTIVE: To explore the relationship between expression of ERR alpha mRNA and estrogen and progesterone and to elucidate the function of ERR alpha in endometrial carcinoma. METHODS: The expression of ERR alpha mRNA was examined by reverse transcription polymerase chain reaction. Endometrial carcinoma cell line Ishikawa was dealt with different concentrations of 17beta-estradiol (10(-10) mol/L, 10(-8) mol/L and 10(-6) mol/L) for 15 min, 30 min and 24 h and 17beta-estradiol and ER inhibitor-ICI182780 were given concomitantly to observe the change of ERR alpha mRNA. Different concentrations of progesterone (10(-8) mol/L, 10(-7) mol/L, 10(-6) mol/L and 10(-5) mol/L) were also given to Ishikawa cell line for 24 h to observe the change of ERR alpha mRNA. RESULTS: The expression level of ERR alpha mRNA was slightly higher than that of the control group after being stimulated for 15 min, 30 min and 24 h by 10(-10) mol/L 17beta-estradiol. However the expression level of ERR alpha mRNA was significantly lower than that of the control group after being stimulated for 15 min, 30 min and 24 h by 10(-8) mol/L and 10(-6) mol/L of 17beta-estradiol. When 10(-6) mol/L of ICI182780 and 10(-8) mol/L of 17beta-estradiol were given simultaneously to Ishikawa cell line, this down-regulation was blocked. After being stimulated for 24 h by 10(-7) mol/L, 10(-6) mol/L and 10(-5) mol/L of progesterone, the expression levels of ERR alpha mRNA were significantly higher than that of the control group, but no change was found in 10(-8) mol/L of progesterone. CONCLUSION: 17Beta-estradiol can down-regulate the expression of ERR alpha mRNA and this regulation is mediated by estrogen receptor. Progesterone can up-regulate the expression of ERR alpha mRNA.
