[Gene rearrangement studies in Hashimoto's thyroiditis and primary lymphoma of thyroid].

OBJECTIVE: To study the immunoglobulin gene rearrangement patterns in Hashimoto's thyroiditis (HT) and primary thyroid lymphoma (PTL), and to analyze the relationship between the two diseases. METHODS: Formalin-fixed and paraffin-embedded tissues of 11 cases of PTL and 38 cases of HT as well as their clinical data, were retrieved. The latter group was further subcategorized into classic HT and suspicious PTL. Gene rearrangement studies for immunoglobulin heavy chains and light chains were carried out by polymerase chain reaction (PCR) using VH, FR3A and FR3kappa primers. RESULTS: There was an increasing trend in immunoglobulin gene rearrangement rate for classic HT (10.7%), suspicious PTL (40.0%) and PTL (72.7%) groups. In general, a female predilection was observed. This sex predilection however was less obvious in the PTL group. There was no relationship between serum antibody (both thyroglobulin and thyroid peroxidase) titers and gene rearrangement patterns. CONCLUSIONS: HT and PTL show morphologic overlaps and may not be clearly distinguished on the basis of light microscopy alone. PCR-based immunoglobulin gene rearrangement study may be helpful in the detection of cases with early lymphomatous transformation of HT.

[Advances in primary thyroid lymphoma].

Primary thyroid lymphomas (PTLs) are closely correlated with the autoimmune reaction of thyroid. However, the molecular mechanisms of PTLs are still unclear. It is really necessary to improve the ability to differentiate between benign and malignant PTLs along with the introduction of some new molecular biology methods. The diagnosis and prognosis of PTLs depend on their histological features, pathological classification, and clinical stages. Customized therapy of PTLs becomes possible with the further advances in lymphoma's pathological classification, clinical stages, and international prognosis index standard.

[Screening of differentially expressed genes in colorectal cancer using human whole genomic oligonucleotide microarrays].

OBJECTIVE: To screen the differentially expressed genes in human colorectal cancer (CRC) tissue. METHODS: Affymetrix oligonucleotide microarrays HG-U133 representing 32,264 human genes including 19,308 known genes and 12,956 expressed sequence tags (ESTs) were used to detect the gene expressions of CRC tissue paired with normal mucosa tissue. The microarray findings were confirmed by real-time quantitative reverse transcriptase-polymerase chain reaction (FQ-PCR). The gene expression profiles were analyzed by intersection and complement, rank sum test and t test. RESULTS: Totally 3,125 genes and ESTs expressed differentially were detected in normal and cancer tissues, consisting of 974 up-regulated and 2,151 down-regulated genes with 247 ESTs present in CRC tissue and absent in normal mucosa and 162 ESTs absent in CRC tissue but present in normal mucosa. A percent of 80.1% of the differentially expressed genes were not reported in the literatures. CONCLUSION: The strategy of data mining provides a foundation for filtering molecular markers and interpreting molecular carcinogenesis of CRC.

[Differences in morphology and immunohistochemistry between hepatic angiomyolipoma and renal angiomyolipoma].

OBJECTIVE: To investigate the clinicopathological and immunohistochemical characteristics of hepatic angiomyolipoma (AML) and renal AML, and to compare the similarities and differences between the two. METHODS: Hematoxylin-eosin staining and SP immunohistochemical staining were performed on the specimens resected during operation of 12 cases of hepatic AML, 2 males and 10 females, aged 41, and 38 renal AML cases, 14 males and 24 females, aged 43. Wilcoxon rank test was made on the proportions of the 3 kinds of myoid cells and fat tissues. RESULTS: All tumors were composed of three basic components: blood vessels, fatty tissue, and myoid cells epithelioid, spindle, or intermediate in shape. Epithelioid cells accounted for more than 50% of the cells in 6 out of the 12 hepatic AML cases often with apparent cellular and nuclear atypia (12/12), and accounted for less than 25% in all renal AML cases. Spindle-shaped cells were scarcely observed in hepatic AML and were common in renal AML. Nuclear atypia was observed in 20 out of the 38 renal AML cases, 18 cases being mild and 1 being moderate. Fat cells accounted for less than 50% in hepatic AML, and more than 50% in renal AML (P = 0.001). Extramedullary hematopoietic tissue was often seen in hepatic AML but never observed in renal AML. Strong positivity of HMB45 and focal positivity of SMA were common in hepatic AML and strong positivity of SMA and sporadic positivity of HMB45 were observed in the renal AML specimens. CONCLUSION: The histological composition of AML is highly variable in the proportion and distribution of the 3 component cells between hepatic AML and renal AML.
