The interaction between orthographic and phonological information in children: an fMRI study.

We examined the neural representations of orthographic and phonological processing in children, while manipulating the consistency between orthographic and phonological information. Participants, aged 9-15, were scanned while performing rhyming and spelling judgments on pairs of visually presented words. The orthographic and phonological similarity between words in the pair was independently manipulated, resulting in four conditions. In the nonconflicting conditions, both orthography and phonology of the words were either (1) similar (lime-dime) or (2) different (staff-gain); in conflicting conditions, words had (3) similar phonology and different orthography (jazz-has) or (4) different phonology and similar orthography (pint-mint). The comparison between tasks resulted in greater activation for the rhyming task in bilateral inferior frontal gyri (BA 45/47), and greater activation for the spelling task in bilateral inferior/superior parietal lobules (BA 40/7), suggesting greater involvement of phonological and semantic processing in the rhyming task, and nonlinguistic spatial processing in the spelling task. Conflicting conditions were more difficult in both tasks and resulted in greater activation in the above regions. The results suggest that when children encounter inconsistency between orthographic and phonological information they show greater engagement of both orthographic and phonological processing.

The role of the basal ganglia and cerebellum in language processing.

The roles of the cerebellum and basal ganglia have typically been confined in the literature to motor planning and control. However, mounting evidence suggests that these structures are involved in more cognitive domains such as language processing. In the current study, we looked at effective connectivity (the influence that one brain region has on another) of the cerebellum and basal ganglia with regions thought to be involved in phonological processing, i.e. left inferior frontal gyrus and left lateral temporal cortex. We analyzed functional magnetic resonance imaging data (fMRI) obtained during a rhyming judgment task in adults using dynamic causal modeling (DCM). The results showed that the cerebellum has reciprocal connections with both left inferior frontal gyrus and left lateral temporal cortex, whereas the putamen has unidirectional connections into these two brain regions. Furthermore, the connections between cerebellum and these phonological processing areas were stronger than the connections between putamen and these areas. This pattern of results suggests that the putamen and cerebellum may have distinct roles in language processing. Based on research in the motor planning and control literature, we argue that the putamen engages in cortical initiation while the cerebellum amplifies and refines this signal to facilitate correct decision making.

Weaker top-down modulation from the left inferior frontal gyrus in children.

Previous studies have shown that developmental changes in the structure and function of prefrontal regions can continue throughout childhood and adolescence. Our recent results suggested a role for the left inferior frontal cortex in modulating task-dependent shifts in effective connectivity when adults focus on orthographic versus phonological aspects of presented words. Specifically, the top-down influence of the inferior frontal cortex determined whether incoming word-form information from the fusiform gyrus would have a greater impact on the parietal areas involved in orthographic processing or temporal areas involved in phonological processing (Bitan, T., Booth, J.R., Choy, J., Burman, D.D., Gitelman, D.R. and Mesulam, M.-M., 2005. Shifts of Effective Connectivity within a Language Network during Rhyming and Spelling. J. Neurosci. 25, 5397-5403.). In the current study, we find that children displayed an identical pattern of task-dependent functional activations within this network. In comparison to adults, however, children had significantly weaker top-down modulatory influences emanating from the inferior frontal area. Adult language processing may thus involve greater top-down cognitive control compared to children, resulting in less interference from task-irrelevant information.

Specialization of phonological and semantic processing in Chinese word reading.

The purpose of this study was to examine the neurocognitive network for processing visual word forms in native Chinese speakers using functional magnetic resonance imaging (fMRI). In order to compare the processing of phonological and semantic representations, we developed parallel rhyming and meaning association judgment tasks that required explicit access and manipulation of these representations. Subjects showed activation in left inferior/middle frontal gyri, bilateral medial frontal gyri, bilateral middle occipital/fusiform gyri, and bilateral cerebella for both the rhyming and meaning tasks. A direct comparison of the tasks revealed that the rhyming task showed more activation in the posterior dorsal region of the inferior/middle frontal gyrus (BA 9/44) and in the inferior parietal lobule (BA 40). The meaning task showed more activation in the anterior ventral region of the inferior/middle frontal gyrus (BA 47) and in the superior/middle temporal gyrus (BA 22,21). These findings are consistent with previous studies in English that suggest specialization of inferior frontal regions for the access and manipulation of phonological vs. semantic representations, but also suggest that this specialization extends to the middle frontal gyrus for Chinese. These findings are also consistent with the suggestion that the left middle temporal gyrus is involved in representing semantic information and the left inferior parietal lobule is involved in mapping between orthographic and phonological representations.

Developmental and skill effects on the neural correlates of semantic processing to visually presented words.

Functional magnetic resonance imaging (fMRI) was used to explore the neural correlates of semantic judgments to visual words in a group of 9- to 15-year-old children. Subjects were asked to indicate if word pairs were related in meaning. Consistent with previous findings in adults, children showed activation in bilateral inferior frontal gyri (Brodmann area [BA] 47, 45) and left middle temporal gyrus (BA 21). Words with strong semantic association elicited significantly greater activation in bilateral inferior parietal lobules (BA 40), suggesting stronger integration of highly related semantic features. By contrast, words with weak semantic association elicited greater activation in left inferior frontal gyrus (BA 45) and middle temporal gyrus (BA 21), suggesting more difficult feature search and more extensive access to semantic representations. We also examined whether age and skill explained unique variance in the patterns of activation. Increasing age was correlated with greater activation in left middle temporal gyrus (BA 21) and inferior parietal lobule (BA 40), suggesting that older children have more elaborated semantic representations and more complete semantic integration processes, respectively. Decreasing age was correlated with activation in right superior temporal gyrus (BA 22) and decreasing accuracy was correlated with activation in right middle temporal gyrus (BA 21), suggesting the engagement of ancillary systems in the right hemisphere for younger and lower-skill children.

Integrin binding characteristics of the disintegrin-like domain of ADAM-15.

Although discovered as potent inhibitors of a (IIb) ss (3) -mediated platelet aggregation, snake venom disintegrins are now known to bind to other integrins according to different degrees of potency and specificity. More recently, homologues of the disintegrinlike loop have been found as a discrete domain in the ADAM family, yet the potency and specificity of each of these domains in terms of integrin binding is relatively unknown. In this present study, we have selected the disintegrin-like domain (dd) of ADAM-15 (designated as ddADAM-15), the only RGD containing domain in the ADAM family, for a structure/function study. Experimentally, the ddADAM-15 and a number of mutants in which the RGD-containing loop was substituted by cognate regions from ADAM-2, -12 and -19 were tested in terms of integrin-binding activity. For comparison with ADAM-15, an additional mutant (dd (den) ADAM-15) was designed based upon the RGD-containing loop of snake venom dendroaspin, a disintegrin-like integrin antagonist. The results showed that ddADAM-15 is an inhibitor of platelet aggregation, though with less potency than dd (den) ADAM-15. None of the other mutants exhibited significant inhibition of platelet aggregation. ddADAM-15 was found to have higher binding ability for a (2) ss (1) and a (9) ss (1) than the ADAM-2 derived mutant which appeared to be more selective for a (V) ss (3) and a (4) ss (1) than either ddADAM-15 or its ADAM-19 based mutant. The integrin-binding properties of ddADAM-15 were completely abolished by point mutation within the RGD motif (R (64) GD ? A (64) GD). These results suggest a more subtle contribution of this loop sequence to defining the functionality of the ADAMs compared to dendroaspin.

Developmental changes in the neural correlates of semantic processing.

Functional magnetic resonance imaging (fMRI) was used to explore the neural correlates of semantic judgments in the auditory modality in a group of 9- to 15-year-old children. Subjects were required to indicate if word pairs were related in meaning. Consistent with previous findings in adults, children showed activation in bilateral superior temporal gyri (BA 22) for recognizing spoken words as well as activations in bilateral inferior frontal gyri (BAs 47, 45) and left middle temporal gyrus (BA 21) for semantic processing. The neural substrates of semantic association and age differences were also investigated. Words with strong semantic association elicited significantly greater activation in the left inferior parietal lobule (BA 40), whereas words with weak semantic association elicited activation in left inferior frontal gyrus (BAs 47/45). Correlations with age were observed in the left middle temporal gyrus (BA 21) and the right inferior frontal gyrus (BA 47). The pattern of results for semantic association implies that the left inferior parietal lobule effectively integrates highly related semantic features and the left inferior frontal gyrus becomes more active for words that require a greater search for semantic associations. The developmental results suggest that older children recruit the right inferior frontal gyrus as they conduct a broader semantic search and the left middle temporal gyrus to provide more efficient access to semantic representations.

Electro-optic modulation in hybrid solgel doped with Disperse Red chromophore.

An electro-optically active hybrid solgel doped with Disperse Red 13 has been developed by use of a simple solvent-assisted method. It permits a high loading concentration and has low optical loss at 1550 nm. A channel waveguide amplitude modulator has been fabricated by use of active and passive hybrid solgel materials. The device shows an electro-optic coefficient of 14 pm/V at 1550 nm and stable operation.

Analysis of SAW properties in ZnO/AlxGa1-xN/c-Al2O3 structures.

Piezoelectric thin films on high acoustic velocity nonpiezoelectric substrates, such as ZnO, AlN, or GaN deposited on diamond or sapphire substrates, are attractive for high frequency and low-loss surface acoustic wave devices. In this work, ZnO films are deposited on AlxGa1-xN/c-Al2O3 (0 < or = chi < or = 1) substrates using the radio frequency (RF) sputtering technique. In comparison with a single AlxGa1-xN layer deposited on c-Al2O3 with the same total film thickness, a ZnO/AlxGa1-xN/c-Al2O3 multilayer structure provides several advantages, including higher order wave modes with higher velocity and larger electromechanical coupling coefficient (K2). The surface acoustic wave (SAW) velocities and coupling coefficients of the ZnO/AlxGa1-xN/c-Al2O3 structure are tailored as a function of the Al mole percentage in AlxGa1-xN films, and as a function of the ZnO (h1) to AlxGa1-xN (h2) thickness ratio. It is found that a wide thickness-frequency product (hf) region in which coupling is close to its maximum value, K(2)max, can be obtained. The K(2)max of the second order wave mode (h1 = h2) is estimated to be 4.3% for ZnO/GaN/c-Al2O3, and 3.8% for ZnO/AlN/c-Al2O3. The bandwidth of second and third order wave modes, in which the coupling coefficient is within +/- 0.3% of K(2)max, is calculated to be 820 hf for ZnO/GaN/c-Al2O3, and 3620 hf for ZnO/AlN/c-Al2O3. Thus, the hf region in which the coupling coefficient is close to the maximum value broadens with increasing Al content, while K(2)max decreases slightly. When the thickness ratio of AlN to ZnO increases, the K(2)max and hf bandwidth of the second and third higher wave modes increases. The SAW test devices are fabricated and tested. The theoretical and experimental results of velocity dispersion in the ZnO/AlxGa1-xN/c-Al2O3 structures are found to be well matched.

The effect of the single substitution of arginine within the RGD tripeptide motif of a modified neurotoxin dendroaspin on its activity of platelet aggregation and cell adhesion.

The Arg-Gly-Asp (RGD) tripeptide unit is a cell-cell and cell-extracellular matrix recognition sequence of some integrins that is found within several extracellular matrix glycoproteins and dendroaspin, a disintegrin-like venom protein isolated from the snake venom of the Dendroaspis jamsonii. In the present study, the RGD motif in dendroaspin was substituted by Lys-Gly-Asp (KGD), His-Gly-Asp (HGD), Gln-Gly-Asp (QGD) and Ala-Gly-Asp (AGD) denoted as KGD-den, HGD-den, QGD-den and AGD-den, respectively. Each of the mutants exhibited activity as inhibitor of ADP-induced platelet aggregation with IC50 values of 0.26, 2.5, 6, and 17 microM for KGD-den, HGD-den, QGD-den, and AGD-den, respectively, as compared with RGD-den (IC50 = 0.18 microM). Interestingly, HGD-den was approx. two-fold more potent and a more selective inhibitor than either the KGD-den or QGD-den counterpart at blocking A375-SM human melanoma cell adhesion to fibrinogen (beta3-mediated). KGD-den, HGD-den, and QGD-den were preferentially antagonists of A375-SM human melanoma cell adhesion to fibrinogen rather than to fibronectin (alpha5beta1-, beta3-mediated). Both HGD-den and KGD-den were equipotent as inhibitors of human erythroleukaemia (HEL) cell adhesion to fibrinogen (IC50 = 0.15 microM) and also preferential inhibitors of HEL cell adhesion to fibrinogen (beta3 and beta1-mediated) rather than to fibronectin. These findings show that the presence of the arginine within the RGD motif of dendroaspin is not obligatory and substitution of this residue can modulate inhibitory potency and integrin binding selectivity.
