Most antiviral CD8 T cells during chronic viral infection do not express high levels of perforin and are not directly cytotoxic.

Despite the frequency of HIV-specific CD8 T cells, most HIV-infected patients do not control viral replication without antiviral drugs. Although CD8 T cells are important in containing acute HIV and simian immunodeficiency virus (SIV) infection, CD8 T-cell functions are compromised in chronic infection. To investigate whether functional deficits are specific to HIV, the phenotypic and functional properties of HIV, Epstein-Barr virus (EBV), and cytomegalovirus (CMV)-specific CD8 T cells, labeled with HLA A2.1 or B8 tetramers, were compared in 35 HIV-infected and 9 healthy donors. Cytotoxic T lymphocytes express the cytolytic molecules perforin and granzymes, and are thought to be CD45RA(+)CD27(-). Although most HIV- specific cells are antigen experienced and express granzyme A (median, 85%), few express high levels of perforin (median, 10%) or CD45RA (median, 14%) or have down-modulated CD27 (median, 12%). Perforin expression by HIV-specific cells is not significantly different from that of EBV- or CMV-specific cells in the same donors or in healthy donors. EBV- and CMV-specific cells, like HIV-specific cells, are often not cytotoxic when tested directly ex vivo. HIV-specific T-cell expression of other phenotypic markers is similar to that of EBV- and CMV-specific CD8 T cells in healthy donors. However, CMV-specific cells (and, to a lesser extent, EBV-specific cells) in HIV-infected donors are more likely to be CD27(-), CD45RA(+), and GzmA(+). These results suggest that the chance to eradicate an infection by T-cell-mediated lysis may be undermined once an infection becomes chronic. Impaired antiviral cytotoxicity during chronic infection is not specific to HIV but likely represents the immune response to chronic antigenic exposure.

CD8-Positive T-Lymphocytes↗

HMG2 interacts with the nucleosome assembly protein SET and is a target of the cytotoxic T-lymphocyte protease granzyme A.

The cytotoxic T-lymphocyte protease granzyme A induces caspase-independent cell death in which DNA single-stranded nicking is observed instead of oligonucleosomal fragmentation. A 270- to 420-kDa endoplasmic reticulum-associated complex (SET complex) containing the nucleosome assembly protein SET, the tumor suppressor pp32, and the base excision repair enzyme APE can induce single-stranded DNA damage in isolated nuclei in a granzyme A-dependent manner. The normal functions of the SET complex are unknown, but the functions of its components suggest that it is involved in activating transcription and DNA repair. We now find that the SET complex contains DNA binding and bending activities mediated by the chromatin-associated protein HMG2. HMG2 facilitates assembly of nucleoprotein higher-order structures by bending and looping DNA or by stabilizing underwound DNA. HMG2 is in the SET complex and coprecipitates with SET. By confocal microscopy, it is observed that cytoplasmic HMG2 colocalizes with SET in association with the endoplasmic reticulum, but most nuclear HMG2 is unassociated with SET. This physical association suggests that HMG2 may facilitate the nucleosome assembly, transcriptional activation, and DNA repair functions of SET and/or APE. HMG2, like SET and APE, is a physiologically relevant granzyme A substrate in targeted cells. HMG1, however, is not a substrate. Granzyme A cleavage after Lys65 in the midst of HMG box A destroys HMG2-mediated DNA binding and bending functions. Granzyme A cleavage and functional disruption of key nuclear substrates, including HMG2, SET, APE, lamins, and histones, are likely to cripple the cellular repair response to promote cell death in this novel caspase-independent death pathway.

Infrared thermoimages display of body surface temperature reaction in experimental cholecystitis.

AIM: To display the thermoimages of the body surface in experimental cholecystitis, to observe the body surface temperature reaction in visceral disorders, and to study if the theory of body surface-viscera correlation is true and the mechanism of temperature changes along the meridians. METHODS: By injecting bacteria suspension into the stricture bile duct and gallbladder, 21 rabbits were prepared as acute pyogenic cholangiocholecystitis models, with another 8 rabbits prepared by the same process except without injection of bacteria suspension as control. The body surface infrared thermoimages were continuously observed on the hair shaven rabbit skin with AGA-782 thermovision 24h before, 1-11 d after and (2,3 wk) 4 wk after the operation with a total of over 10 records of thermoimages. RESULTS: Twelve cases out of 21 rabbits with cholecystitis revealed bi-lateral longitudinal high temperature lines in its trunk; with negative findings in the control group. The high-temperature line appeared on d1-d2, first in the right trunk, after the preparation of the model, about 7 d after the model preparation, the lines appeared at the left side too, persisting for 4 wk. The hyper-temperature line revealed 1.1-2.7 degrees higher than before the model preparation, 0.7-2.5 degrees higher than the surrounding skin. The length of the high temperature line might reach a half length of the body trunk, or as long as the whole body itself. CONCLUSION: The appearance of the longitudinal high temperature lines at the lateral aspects of the trunk in the experimental group is directly bound up with the experimental animals pyogenic cholecystitis, with its running course quite similar to that of the Gallbladder Channel of Foot Shaoyang, but different to the zones of hyperalgesia and site of referred pain in cholecystitis.

Infrared thermography display of cortical temperature in cats.

OBJECTIVE: To use infrared thermography to take cat cerebral cortical temperature in order to visualize the temperature of its entire cerebral cortex as an image. METHODS: After performing craniotomy for exposure of cerebral hemispheres in 52 cats, their cortical temperatures were displayed and analyzed by computerized infrared thermovision. RESULTS: The temperature distribution of the cerebral cortex was uneven, with a maximum difference of 2.3 degrees C among different cortical areas. The temperature in the cortical anterior-inferior area (including the Ant. Ectosylvian, the lower section of Mid. Ectosylvian and the Ant. Sylvian) was higher compared to the temperature in the posterior-super-parts (Post. Suprasylvian, Mid. Suprasylvian, Post. Lateral and Ant. Lateral). Locations with higher or lower temperatures showed little change within three days after craniotomy, and the cortical temperature held steady. CONCLUSION: The use of cortical infrared thermo-images for display of cat cortical temperature is possible and has many advantages over traditional methods. This new neuroimaging method has a practical value in neurological research.

CT characteristics of cervical Castleman's disease.

Castleman's disease is an uncommon disorder of benign lymph-node hyperplasia. Among all the lesions, less than 10% occurs in the neck. Our case differs from those cases previously reported. Multiple lesions were found on both sides of the neck. Contrast-enhanced CT scan showed dotty and stripy enhancement around these lesions, and rim-like enhancement was also found on delayed scan at 5 min as well. The possibility of Castleman's disease should be considered in multiple cervical masses that show well-defined marked and homogeneous enhancement, especially when stripy enhancement is found around the lesions simultaneously.

A comparison of intrazeolite and solution singlet oxygen Ene reactions of allylic alcohols.

The singlet oxygen ene reactions of four allylic alcohols and for comparison an allylic ether have been examined both in solution and in zeolite Y. Brønsted acid sites in the zeolite were shown to induce decomposition of several of the allylic alcohols. Treatment of the zeolites with pyridine removed these acid sites and allowed intrazeolite reactions of the allylic alcohols without interference from decomposition. Control reactions with an allylic alcohol that is inert to decomposition provided evidence that the presence of pyridine in the zeolite labyrinth does not influence the product composition.

[Degradation of proteinaceous substrates by xylotrophic basidiomycetes].

The ability of various xylotrophs to produce extracellular proteolytic enzymes has been studied, with emphasis on medium-related factors regulating their secretion. Direct measurement of proteolytic activity in the culture liquid and postelectrophoresis determination of protease activity in polyacrylamide gel copolymerized with gelatin demonstrated that the secreted enzymes are quantitatively and qualitatively diverse. Activity levels of extracellular proteolytic enzymes strongly depend on pH and contents of protein and carbohydrate in the medium. All secreted proteases notably differed in molecular weight (of 51 kDa or higher and in excess of 95 kDa) and belonged mostly to two classes of proteolytic enzymes (serine proteases and metalloproteinases).
