Regulation of metal transporters by dietary iron, and the relationship between body iron levels and cadmium uptake.

Iron (Fe) plays essential roles in biological processes, whereas cadmium (Cd) is a toxic and non-essential metal. Two metal transporters, divalent metal transporter 1 (DMT1) and metal transporter protein 1 (MTP1), are responsible for Fe transport in mammals. Here, we studied the effect of dietary Fe on the expression of these metal transporters in peripheral tissues, and the uptake by these tissues of Cd. Mice were fed an Fe-sufficient (FeS: 120 mg Fe/kg) or Fe-deficient (FeD: 2-6 mg Fe/kg) diet for 4 weeks. The total Fe levels in the body were evaluated by measuring tissue Fe concentrations. Tissue Cd concentrations were determined 24 h after the mice received a single oral dose of Cd. Animals fed a FeD diet showed depletion of body Fe levels and accumulated 2.8-fold higher levels of Cd than the FeS group. Quantitative real time RT-PCR revealed that whereas DMT1 and MTP1 were both ubiquitously expressed in all FeS peripheral tissues studied, DMT1 was highly expressed in brain, kidney, and testis, whereas MTP1 was highly expressed in liver and spleen. Depletion of the body Fe stores dramatically upregulated DMT1 and MTP1 mRNA expression in the duodenum as well as moderately upregulating their expression in several other peripheral tissues. The iron response element positive isoform of DMT1 was the most prominently upregulated isoform in the duodenum. Thus, DMT1 and MTP1 may play an important role in not only maintaining Fe levels but also facilitating the accumulation of Cd in the body of mammals.

Significance of a solitary perivascular mononuclear infiltrate in lung allograft recipients with mild acute cellular rejection.

BACKGROUND: Although a solitary prominent perivascular mononuclear infiltrate is diagnostic of mild acute rejection (A2) in lung allograft recipients, its significance is still poorly defined. We evaluated the significance of a solitary perivascular mononuclear infiltrate and its correlation with clinical outcome in lung allograft recipients. METHODS: Thirteen patients had mild acute rejection as diagnosed by the presence of a solitary perivascular mononuclear infiltrate. The patients were divided into 2 groups based on subsequent treatment: treated (Group 1) and non-treated (Group 2) patients. We analyzed the difference between the 2 groups according to clinical presentation, histologic parameters and outcome. RESULTS: Nine patients were women (69%), 4 were men (31%); 12 were white and 1 was African American. Ages at the time of biopsy ranged from 20 to 68 years, with a mean of 47.2 years and a median of 52 years. Eight had a history of single-lung transplant and 5 had a history of double-lung transplant. The most common reasons for transplantation were emphysema (n = 6) and cystic fibrosis (n = 3). Nine patients (65.4%) showed decreased rejection grade or no evidence of acute rejection (Group 1) after treatment. Four patients who were untreated had persistent multifocal mild or worsening moderate rejection on subsequent biopsy (Group 2). CONCLUSIONS: Treated and untreated patients with mild rejection based on a solitary perivascular infiltrate have similar clinical presentations and histologic characteristics. Solitary mononuclear infiltrates showed persistence or progression without therapy and therefore need to be treated as, not segregated from, the "usual" forms of mild acute allograft rejection.

Two chemists in two Koreas.

Taikyu Ree (1902-92) and Seung Ki Li (1905-96) were the most famous and influential scientists in twentieth-century Korea. Trained at Kyoto Imperial University during the Japanese occupation period (1910-45), both followed parallel career paths until 1945 but after that year those paths began to diverge. In 1948 Taikyu Ree moved to the University of Utah where he ran an informal graduate programme to train Korea chemists during the 1950s and 1960s. He became a model scholar in pursuit of knowledge for its own sake. In 1950 Seung Ki Li moved to North Korea where he succeeded in industrialising a new synthetic fibre from polyvinyl alcohol. He became a popular hero who demonstrated the utility of science. His success also helped Kim Il Sung, North Korea dictator, to consolidate his political ideology of Self-Reliance. Despite their different career patterns in very different circumstances, they shared something common; apolitical views and commitment to their work. An investigation of the careers of these two chemists illustrates the interwoven scientific, ideological and economic developments of South and North Korea during the second half of the twentieth century.

Direct injection of immature dendritic cells into irradiated tumor induces efficient antitumor immunity.

Although there are several ways to load tumor antigens to DCs, in vitro preparation of tumor antigens and manipulation of DCs are usually required. Therefore, to develop a simple antitumor immunization method, we examined if direct injection of DCs into tumor apoptosed by ionizing IR could induce efficient antitumor immunity. Ionizing IR with 15 Gy induced apoptosis in tumor maximally after 6 hr. Injection of DCs i.t. into IR tumor induced strong cytotoxicity of splenocytes against tumor cells compared to i.t. injection of DCs or ionizing IR of tumor, both of which induced weak cytotoxicity. In an animal study, i.t. injection of DCs into IR tumor induced therapeutic antitumor immunity against a tumor established at a distant site. Moreover, when TNF-alpha or LPS was added as a danger/maturation signal to DC suspension before i.t. injection, antitumor immunity was significantly potentiated compared to a group treated with i.t. injection of DCs into IR tumor. Our results suggest that injection of DCs into tumor apoptosed by ionizing IR might be a simple and efficient method of immunization against tumor.

Crystallization and preliminary X-ray crystallographic analysis of a ytfG gene product from Escherichia coli.

The Escherichia coli ytfG gene product, with NAD(P)H:quinone oxidoreductase activity, was crystallized by the hanging-drop vapour-diffusion method at 296 K. A 1.78 A data set has been collected using synchrotron radiation at Pohang Light Source, South Korea. The crystal belongs to the primitive trigonal system, with unit-cell parameters a = b = 81.7, c = 76.8 A. Analysis of the packing density shows that the asymmetric unit probably contains one monomer, with a solvent content of 48.8%.

Traumatic aneurysm of the supraclinoid internal carotid artery and an associated carotid-cavernous fistula: vascular reconstruction performed using intravascular implantation of stents and coils. Case report.

This report documents the treatment of a traumatic aneurysm of the supraclinoid internal carotid artery (ICA) that was associated with a carotid-cavernous fistula (CCF), which appeared following closed head trauma. This life-threatening lesion, which is very rare, required aggressive management achieved using intravascular stents and coils. A 19-year-old man presented with severe traumatic intracerebral and subarachnoid hematoma after he had suffered a severe closed head injury in a motor vehicle accident. Cerebral angiography performed 11 days after the injury demonstrated a traumatic aneurysm and severe narrowing of the right supraclinoid ICA, which was consistent with a dissection-induced stenosis associated with a direct CCF. Both lesions were successfully obliterated with preservation of the parent artery by using stents in conjunction with coils. Follow-up angiography obtained 7 months postoperatively revealed persistent obliteration of the aneurysm and CCF as well as patency of the parent artery. The patient remained asymptomatic during the clinical follow-up period of 14 months. Endovascular treatment involving the use of a stent combined with coils appears to be a feasible, minimally invasive option for treatment of this hard-to-treat lesion.

Immunohistochemical analysis for histopathological subtypes in pediatric medulloblastomas.

Medulloblastomas occurring in children represent a histological spectrum of varying anaplasia and nodularity. In order to determine whether immunohistochemical markers might be useful parameters in subclassifying these tumors, 17 pediatric medulloblastomas, including nine diffuse/non-anaplastic, four diffuse/anaplastic, three nodular/non-anaplastic and one nodular/anaplastic subtypes, were studied. In the present report, we investigate the expression of neural cell adhesion molecule (NCAM), nerve growth factor receptor (NGFR), neurofilament (NF), synaptophysin (SYN), glial fibrillary acidic protein (GFAP), S100, Bcl-2, and Ki-67 by using the immunohistochemistry against specific antibodies. This study showed that NGFR, NF, GFAP and S100 were not detected in anaplastic subtypes of medulloblastomas (0/5), while non-anaplastic subtypes were mainly expressed within the nodules. All 17 tumors were reactive for NCAM, SYN and Bcl-2. In addition, Ki-67 labeling indices for anaplastic subtypes (39.0 +/- 7.42%) were significantly higher than that of non-anaplastic medulloblastomas (11.4 +/- 8.04%; P < 0.0001). These results suggest that immunohistochemical markers are a useful adjunct in characterizing subtypes of pediatric medulloblastomas.

Merits of intralesional fibrin glue injection in surgery for cavernous sinus cavernous hemangiomas. Technical note.

In planning surgical treatment for extraaxial cavernous hemangiomas, care should be taken to control severe tumor bleeding. The authors present a case of a large cavernous hemangioma of the cavernous sinus, which was completely removed with the aid of multiple intratumoral injections of fibrin glue. This novel method is very effective for preventing excessive blood loss during surgery for this type of lesion.
