[Late effects of damaging action of doxorubicin on the reproductive system and progeny of rats].

The effect of doxorubicin, an anthracycline antibiotic, on the reproductive system and progeny was studied in rats 1 month after a single administration of the drug in the MID of 6.85 mg/kg. It was shown that doxorubicin induced a decrease in the number of the mature oocytes in the rat ovary and an increase in the pre- and post-implantation death rate. The rats exposed to the antibiotic 1 month before the mating with intact male rats had unchanged fertility and pregnancy. In the fetuses isolated from the female rats treated with the antibiotic, development of some parts of the skeleton appeared to be retarded. One fetus and one young rat had severe developmental anomalies in the skeletons: shorter right anterior and hind extremities and finger aplasia. The survival rate of the progeny from the female rats exposed to the antibiotic was markedly decreased. No retardation in the physical development of the progeny was observed.

[Myelotoxicity of epirubicin].

Epirubicin (pharmorubicin, India), an antitumor antibiotic of the anthracycline group, was studied in regard to its effect on peripheral blood, bone marrow and lymphoid organs (the thymus and spleen) of CBA mice after its intraperitoneal administration in a single dose equal to the MIC (7.8 mg/kg) and in a course dose (1/5 of the MIC 5 times a day). The cytogenetic impairments induced by the cytostatic were estimated on metaphase plates with the bone marrow specimens and by counting the peripheral blood erythrocytes with micronuclei (the micronuclear test). It was shown that epirubicin induced cytogenetic disturbances in the hemopoietic cells within the first 72 hours. The antibiotic had a marked reversible effect on the erythroid population and lymphoid tissues and a moderate toxic action on the granulocyte population. The antibiotic did not affect thrombocytopoiesis. The single administrations had a more pronounced and prolonged myelotoxic and lymphotropic effect.

[Method of spectral analysis of the rounded EMG and its role in the study of physiologic tremor].

A new computerized method for the assessment of electrical activity of muscles is described. The method is based on statistical processing of the spectral parameters of circumscribing EMG. The method is intended for examination of normal and pathological tremor. The authors describe the results of the use of the method for examination of physiological tremor which may serve as standard criteria for the identification of pathological deviations.

[Study of the pathogenesis of tremor in parkinsonism by the technic of spectral analysis of the rounded EMG using a computer].

Tremor was studied in 2 forms of parkinsonism. A new method of computerized spectral EMG analysis was employed in the investigation. Amplitude and frequency characteristics of tremor in parkinsonism have been identified, which make it possible to objectify the diagnosis at the early stages of the disease and to facilitate its differentiation from other types of pathological tremor. The developed method may be used for assessing the efficiency of the chemotherapeutical and surgical treatment of parkinsonism. Some suggestions were made concerning the mechanism of pathological tremor.

[Spectral electromyographic analysis of essential tremor].

The authors have developed a new method of computerized statistical analysis of the spectral parameters of the circumscribing electromyogram in patients with essential tremor (ET), making use of objective frequency and amplitude parameters. The results have shown that ET differed from physiological tremor not only by amplitude but also by frequency characteristics. Four groups of patients have been identified which differed in relation to both parameters. Clear-cut criteria have been defined which may be used in the differential diagnosis between ET and parkinsonism-associated tremor. It has been suggested that the pathological mechanisms of ET and parkinsonism-associated tremor differ in their location.

[Effect of pH on the properties of the chemostatic culture of Canida utilis].

The effect of hydrogen and hydroxyl ions on the physiological features of the yeast C. utilis VKMU-1668 was studied. High acidity inhibited yeast growth and uncoupled pathways of the energy and constructive metabolism: normal respiration was disturbed and the electron transport chain was damaged in the site of cytochromes and not flavins. Hydroxyl ions also inhibited yeast growth and uncoupled pathways of the energy and constructive metabolism: oxygen uptake and the content of flavin adenone dinucleotide increased, dehydrogenase activity upon the use of glycerol decreased significantly, and the absolute amount of all cytochromes declined slightly. The chemical composition of cellular polymers at all pH values tested was stable enough. The amount of major metabolites--volatile oils and ketoacids--was insignificant.

[Effect of the redox potential on the growth of aerobic microorganisms].

The effect of redox potential was studied on the growth of the following aerobic microorganisms: Candida utilis, Bacillus megaterium, Pseudomonas fluorescens. The action of oxidizing agents (K3Fe(CN)6, KIO3, K2Cr207 and KMnO4) and reducing agents (ascorbic acid, sodium thioglycolate, K4Fe (CN)6 and Na2S2O3) on the growth rate was investigated. K3Fe(CN)6, ascorbic acid, sodium thioglycolate and K4Fe(CN)6 were found to be suitable for buffering the Eh of the medium. The potential could be shifted by 50--200 mV by adding various reducing and oxidizing agents. The rate of growth of C. utilis, Bac. megaterium and Ps. fluorescens did not depend on the potential value.

[Principle growth indices of a chemostat Candida utilis culture resistant to acid pH values].

The kinetics of growth of the Candida utilis chemostat culture 1668-3-37 was studied in a synthetic medium with ethanol at different values of pH and temperature. Chemostat curves were obtained for the pH of the medium of 4.5 and 3.0 and the temperature of 32 degrees C. The following growth characteristics were determined: the maximal growth rate (mumax), the economical coefficient (Y), the substrate (saturation) constant (Ks), the rate of ethanol uptake (q), maintenance energy (m). The minimal amount of ethanol inhibiting the yeast growth was assayed in short-term experiments under periodic conditions with shaking. The value of mumax was 0.35 hr-1 when the yeast was cultivated at 32 degrees C and the pH 4.5, and 0.32 hr-1 at the pH 3.0. The value of Ks varied by an order of magnitude at different pH values when the chemostat culture was grown at D=0.2 hr-1: 36.0 mg/litre at the pH of 3.0 and 3.75 mg/litre at the pH of 4.5. The value of m was close to 0 at the pH of 4.5 and equaled 5--7 mg of ethanol per gram of dry biomass per hour at the pH of 3.0; it was still higher when the temperature of cultivation was increased to 38 degrees C. The minimal substrate (ethanol) concentration inhibiting the yeast growth was constant at different cultivation conditions (pH 3.0 or 4.5 and temperature 32 or 38 degrees C), being equal to 0.45% (v/v) of ethanol.

[Cell adhesion in a chemostat culture of Candida utilis under the influence of supraoptimal temperature and elevated acidity].

The effect of the supraoptimal temperature (38, 40 degrees C) on the chemostat culture of Candida utilis was studied. The above factor caused a part of the biomass to float as a thin layer of foam to the surface of the medium. After an hour, the concentration of the cells on the surface could be four times as high as that within the medium. The content of protein was the same in the cells taken from the surface and from the depth. Singular cells or their groups (2 or 4--8 cells) were found deep in the medium whereas cells on the surface were aggregated forming conglomerates of 20--100 and more cells. The simultaneous action of the elevated tmperature and the acid pH value made flotation of cells onto the surface more stable and protracted (it could be maintained in a chemostat for weeks).
