Organ explant culture of adult Syrian golden hamster pancreas.

An organ explant culture system has been developed for long term maintenance of adult pancreatic tissue from the Syrian golden hamster. Gastric and duodenal lobe explants of up to 0.5 cm2 size were placed in tissue culture dishes (60 mm2) on Gelfoam sponge rafts to which was added 5 ml of CMRL medium 1066 supplemented with heat inactivated newborn bovine serum, L-glutamine, hydrocortisone, insulin, and antibiotics. Dishes were placed in a controlled atmosphere chamber, which was gassed with 45% O2, 50% N2, and 5% CO2 and incubated at 36.5 degrees C. Viability of the tissues was determined by light and electron microscopy as well as by [3]thymidine incorporation. Explants were viable for up to 70 d. Zymogen granule-containing cells characteristic of acinar cells and mucus-containing cells characteristic of ductal cells were present throughout this period. However, endocrine cells were only present for the 1st wk in culture.

Characterization of bovine pancreatic ductal cells isolated by a perfusion-digestion technique.

Bovine pancreatic ductal cells isolated by perfusing an enzyme solution into the lumen of the main duct were obtained as sheets of cells. Morphologic features of these cells were those of pancreatic ductal epithelial cells. These cells also contained alcian blue/periodic acid-Schiff positive material and bound lectins, and they stained for keratin in the same manner as intact ductal epithelium. In culture, the plating efficiency was high (13.6%) as determined by DNA content before and after 24 h plating, perhaps due to the gentle isolation technique and the isolation of sheets of cells rather than a single cell. Cell doubling time was 34.4 h in Eagle's minimal essential medium with 10% heat inactivated fetal bovine serum and antibodies, and over 95% of the cells incorporated [3H]thymidine during a 6 h labeling period after 4 d in primary culture. Isolated cells grew best in medium CMRL 1066 with 10% heat inactivated fetal bovine serum as determined by measuring DNA content.

Methods for transport and long-term maintenance of spiny dogfish sharks.

Spiny dogfish shark (Squalus acanthias) embryos were transported from Maine to the Maryland laboratory where they were maintained for 20 months. During this maintenance period, these sharks progressed from a yolk-sac stage to feeding stage. Animals were maintained at 10 degrees C in artificial seawater containing penicillin and streptomycin and were fed frozen brine shrimp and krill. The ease with which these animals could be maintained in the laboratory for an extended period of time greatly enhanced their usefulness as an experimental animal in a variety of studies.

Risk factors for fractured neck of femur in the elderly.

During a 15-month period, 47 elderly female patients admitted to hospital with a fractured neck of femur were studied immediately prior to discharge, in comparison with 34 elderly female control patients undergoing elective surgery who had been admitted over the same period to the same orthopaedic wards. The principal differences between the two populations were that the fracture patients had a lower forearm trabecular bone density, with lower bodyweight (both lower muscle mass and lower fat content), increased body sway, worse eyesight and reduced mental acuity. The serum biochemistry of the two groups was almost indistinguishable except that the fracture patients tended to have slightly lower concentrations of proteins. There was no evidence to implicate dietary vitamin D deficiency, osteomalacia, oestrogen deficiency or alcoholism in the aetiology of the fracture, nor was there evidence to suggest alterations in endocrine function with respect to cacitonin or parathyroid hormone.

Evaluation of two regimens to immobilise the knee after injections of yttrium-90.

Two regimens to immobilise the knee after injection were compared in patients receiving intra-articular yttrium-90 for persistent synovitis of the knee, the patients being randomly allocated to receive either complete bed rest or full mobilisation with the affected knee in a firm splint. There was no difference in the percentage of the radioisotope retained in the knee or in that taken up by other tissues between the two regimens. It is concluded that after intra-articular injections of yttrium-90 strict bed rest in hospital is unnecessary.

Amniotic fluid cells and the lecithin/sphingomyelin ratio.

Analysis of 86 samples of amniotic fluid included a count of the total number of cells per cubic millimeter, the percent of fatty cells, and the lecithin/sphingomyelin ratio. In the period of gestation studied, 35 to 43 weeks, the average rise in the lecithin/sphingomyelin ratio with each week of gestation was only 0.22 (r = 0.27, P less than 0.02), but the correlations of the lecithin/sphingomyelin ratio with the number of fatty cells per cubic millimeter and with the nonfatty cells per cubic millimeter were highly significant (r = 0.64, P less than 0.001; and r = 0.53, P less than 0.001, respectively). The association of neutral fat-laden cells with the increase in lecithin could be due to a common origin from the skin of the fetus. Alternatively, exfoliation of fatty and nonfatty cells may be stimulated by the same mechanism that causes production of lung surfactant. It is suggested that surfactant lecithin becomes adsorbed onto the surface of the cells in amniotic fluid.

The cytological difference between amniotic fluids of male and female fetuses.

Counts of cyanophilic squamous cells in 48 specimens of amniotic fluid obtained during the last three months of pregnancy gave the correct diagnosis of the sex of the fetus in every case. It is confirmed that the fetal genital tract is the source of a considerable exfoliation of cyanophilic squamous cells. Counts of total cells per mm3 on 38 specimens showed that amniotic fluid from female fetuses at any one week of gestation. The cytological sex difference should be taken into consideration in the interpretation of differential cell count estimations of fetal maturity.

The karyopyknotic index of amniotic fluid cells and its relationship to oestrogen concentration.

Estimations of karyopyknotic index and amniotic fluid oestrogen concentration were made on 107 specimens of amniotic fluid obtained between the 25th and 42nd weeks of pregnancy. The karyopyknotic index was shown to fall with advancing gestation as the amniotic fluid oestrogen concentration increased. The negative correlation between karyopyknotic index and amniotic fluid oestrogen concentration was highly significant when the fetus was female but was not significant when the fetus was male. Intermediate cells from the gential tract of the female fetus can account for the cytological difference between amniotic fluid from male and female fetuses. The fall in amniotic fluid karyaryopyknotic index towards term which suggests that the fetal vagina responds in the same way as the maternal vaginal epithelium to the circulating hormones and not to the oestrogens in the amniotic fluid.

Sputum cytology of metastatic choriocarcinoma: a case report.

A 32-year-old woman who complained of cough and hemoptysis was found to have a metastatic trophoblastic tumor in her lungs and in a supraclavicular lymph node. A regressed primary teratoma of the ovary was considered the most likely origin of the tumor. Sputum cytology showed malignant cells, many of which were in elongated fiberlike forms and some of which had orange-staining cytoplasm. These cytologic appearances of choriocarcinoma are described and compared with the brief descriptions available in the literature. The possibility of misdiagnosis as squamous carcinoma is emphasized. The correct diagnosis is of particular importance because choriocarcinoma of gestational origin, which is similar cytologically to that of teratomatous origin, responds well to chemotherapy.
