[Effect of prenatal stress upon parameters of the behavioral response induced by a tonic pain focus in male and female rats during postnatal ontogenesis].

The long-term effects of prenatal stress (immobilization of females during the last week of pregnancy) were studied on pain sensitivity to a prolonged irritant in Wistar rats during prepubertal, pubertal periods of development and in adults. Pain sensitivity was evaluated from indices of the biphasic behavioral response in the classical formalin test--the intensity of patterns of flexes, shakes and time spent licking, and from duration of the first, acute and the second, tonic phases and from the duration of interphase. Consequences of the prenatal stress manifested themselves differently in the patterns of the response organized at the spinal and supraspinal levels, during tonic phase mainly, differently in females and males; there were peculiarities of effects of prenatal stress in each age period. The data obtained suggest that the consequences of the prenatal stress at the spinal level manifest themselves in activation of modulating descending facilitating and in suppression of inhibitory monoaminergic systems in prepubertal and pubertal rats, and on the contrary, in activation of descending inhibitory and suppression of facilitating systems in adults. Furthermore, considerable evidence is obtained corroborating the idea of inhibitory nature of the interphase and the mechanisms of modulation of acute and tonic phases in the formalin test during different age periods of the individual development.

Prenatal stress alters time characteristics and intensity of formalin-induced pain responses in juvenile rats.

Previous studies found that stressful events during pregnancy can alter the offspring's pain sensitivity to the phasic nociceptive stimuli. The present data constitute the first demonstration of the consequences of prenatal stress to formalin-induced pain in juvenile rats. Injection of formalin into a hind paw of a 25-day-old rat that had not been stressed prenatally produced the typical biphasic specific nociceptive behavioral response consisting of an early short phase lasting 1-4 min followed by a second prolonged phase (12-24 min). Between them there was an interphase that lasted 6-9 min during which the specific behaviors were not shown. This period is generally considered to be a period of inactivity. Prenatally stressed rat pups showed significant increase in flexing+shaking behaviors and in the duration of the second phase of formalin-induced pain in flexing+shaking and licking behaviors and decrease of the duration of the interphase. Disinhibition of the pain behaviors during the interphase was greatly more pronounced in female than in male rats. Sex differences indicate increased vulnerability of inhibitory processes to prenatal stress in females compared with males. These data also underline the importance of understanding the nature of the interphase and provide data on the mechanisms that underlie that component of the formalin test.

Effect of prenatal stress on tonic pain in rats.

The effect of prenatal stress on specific biphasic behavioral response in 25-day rat pups was studied using the experimental model of formalin-induced tonic pain. Prenatally stressed rats showed hypersensitivity to tonic nociceptive stimulation manifested in increased amplitude and duration of the response. Behavioral responses observed during the interphase period attest to impairment of inhibitory processes (more pronounced in females). These findings suggest that the interphase interval is the period of active inhibitory process rather than rest period.

[Effect of initial (spontaneous) arterial pressure on systemic hemodynamic orthostatic responses].

In anesthetized rats, a reverse linear correlation of arterial pressure (AP) shifts was found within the range of initial AP over 95 mm Hg under the head-up tilts at 30 and 45 degrees. Within the range beneath 95 mm Hg this correlation was transformed into the direct one. Under 60 degrees tilt such correlation only occurred within the range beneath 95 mm Hg. The direction and degree of the arterial system responsiveness seems to depend on the tilt value as well as on the range of the initial AP.

The dynamics of two different tests of laterality in rats.

Female rats were tested for behavioural laterality in a T-maze (TM) and a "handling by the tail" (TH) test, and vaginal smears were taken every day. The two tests did not correlate. There was a population bias of left choices in TH, but in TM most of the animals were ambilateral. Statistical analysis showed that animal learning in successive trials influenced their lateral choice, but it was similar in both tests. The difference in lateral bias in rats in the two tests depended mostly on differences in internal laterality. Rats in proestrus had a left-side bias in TM. There was no dependency between direction of lateral choice in TH and estrus cycle.

[Sensitivity of the cat lateral geniculate body neurons to orientation of the brightness gradient vector].

A new property of visual neurons: their sensitivity to orientation and the vector brightness gradient, was revealed and described. Receptive fields of the lateral geniculate body neurons in the cat have preferred orientation maximum reaction (average mean of orientation sensitivity coefficient--0.55 +/- 0.20). The preferred orientation mainly has a radial or tangential trend in the visual field. Temporal characteristics of the neuronal responses were analysed. A role of inhibition processes in the orientation sensitivity is discussed.

[Nociceptive system response to repetitive painful stimuli during chronic psychoemotional stress in humans].

Repeated nocigenic stimuli applied to the skin evoked a hyperalgesia in healthy subjects while in patients with neurasthenia which is a natural model of a chronic psychoemotional stress, they evoked a hyperalgesia accompanied by sensitization. The differences in the nociceptive system responses show particularities in reactivity (plasticity) of a wide dynamic range of the spinal neurons under chronic psychoemotional stress that is due to disorders in supraspinal descending modulation system of plasticity in the spinal nociceptive neurons.

[Pain sensitivity during chronic psychoemotional stress in humans].

In a natural "model" of a chronic psychoemotional stress in humans, (neurasthenia) a decrease in the nociceptive threshold was found. The decrease resulting from weakening of supraspinal tonic descending inhibition which, in its turn, evidently was due to a diminished activity (or dysfunction) of the brain opioid system occurring in a long-lasting psychoemotional stress.

[The effect of an increased erythrocyte count on rapid blood flow fluctuations in the microvessels of the rat brain].

Polycythemia was achieved by means of substitution of the rat own blood (2 ml) with the same volume of washed out erythrocytes, the hematocrit increasing from 44 to 54%. Excessive erythrocytes altered the structure of the erythrocyte flow, its diameter being 3-13 mu. The rate of occurrence of the plasma lumens decreased. The hematocrit increase, however, did not affect the amplitude and period of the oscillations. The character of distribution of periodic components of the fluctuations of the blood flow linear velocity did not change either in the vessels under study. The data obtained suggests that the erythrocytes do not affect the occurrence of the blood flow oscillations in the microcirculation system. The main part in their formation seems to be played by the leucocytes differing by their rheological features from erythrocytes.

Blood flow fluctuations in cerebral cortex microvessels.

Studies on the dynamics of the blood flow in cerebral microvessels with the diameter of erythrocyte flow (5-13 micron) carried out by the cinema-TV method showed that the linear velocity of the blood flow in microvessels experienced considerable fluctuations the amplitude of which varied within 10 to 1500 micron/s and the period within 0.2 to 7 s. It was established that variations in linear velocity of the blood flow were most frequently observed during 0.5-1 s. The autocorrelation analysis showed there was periodicity in the blood flow fluctuations in most microvessels which varied within a wide range of 0.3 to 3 s. A linear dependence was found between the amplitude and the period of fluctuations. The observed variations in the blood flow are likely to be caused by the changes in local resistance when leukocytes and erythrocyte aggregates passed through microvessels.

[Blood flow fluctuations in the microvessels of the rat temporal muscle].

Dynamic characteristics of the blood flow linear velocity in microvessels on the surface of rat's temporal muscle were determined by means of accelerated cinemicrography. The blood flow velocity varied from 200 to 5000 microseconds in microvessels with diameter 4-13 mu. The blood flow fluctuated with period of 0.3-7.0 sec and amplitude 0.5-100% of the average. Auto- and crosscorrelation analysis showed a positive interrelationship between velocities shifted in time by 0.5 sec in 37% of the cases. The interrelationships did not exist between rapid changes of the blood flow in either adjacent or distant microvessels. The sharp blood flow changes seem to be due to periodical passage of leucocytes and aggregated erythrocytes which increases the resistance of blood current in the microvessels.

[Blood flow in the microvessels of the cerebral cortex].

To measure dynamic characteristics of linear velocity of the blood flow in microvessels on the surface of rat's brain, the method of accelerated cinemicrography was used. The blood flow velocity varied from 500 to 5000 mu/sec in microvessels with diameter of 7-15 mu. The blood flow fluctuated with the rate of 1-3 oscillations per second and the amplitude 20-50% of the average value. Auto- and crosscorrelation analysis of the processes revealed in 35% of the cases a positive correlation between shifts of velocities and the frequency of their occurrence. No correlation existed between rapid changes of the blood flow in adjacent or distant microvessels. The sharp changes of blood flow seem to be due to periodical passage of leucocytes and accumulated erythrocytes which increase resistance against the blood flow in microvessels.
